Fluoride compounds are known as hazardous environmental pollutants that can enter the body with drinking water. Chronic exposure to fluoride leads to development of oxidative stress and can lead to activation of nuclear factor κB (NF-κB). The aim of this work is to clarify the role of NF-kB activation in production of reactive nitrogen and oxygen species, activity of antioxidant enzymes and intensity of lipid peroxidation (LPO) in gastric mucosa of rats during chronic fluoride intoxication.Materials and methodsWe carried out the study on 18 mature male rats of the Wistar line. The animals were divided into 3 groups: control animals (6), group of chronic fluoride intoxication (6), and animals (6), which received the NF-κB inhibitor, namely ammonium pyrrolidine dithiocarbamate (PDTC) in a dose of 76 mg / kg (iNF-κB group) during modeling of chronic fluoride intoxication. To assess the development of oxidative stress we studied superoxide production (O2-), activity of superoxide dismutase (SOD), catalase (CAT) and concentration of free malondialdehyde (MDA). We also assessed NO production and concentration of its metabolites (peroxynitrite, nitrosilated thiol groups, nitrites).ResultsChronic fluoride intoxication leads to NO hyperproduction with subsequent increase in concentration of its later metabolites (peroxynitrite, nitrosilated thiol groups, nitrites). Production of O2- increases, SOD activity decreases, CAT activity increases and MDA concentration also increases. Inhibition of NF-kB activation by PDTC normalizes the parameters studied.ConclusionsActivation of NF-κB during chronic fluoride intoxication leads to the development of hyperproduction of NO and development of oxidative-nitrosative stress.  相似文献

The mucosae of the nasal passages contain a large amount of glands which express secretory proteins as well as phase I and phase II biotransformation enzymes. In this review the metabolic activation, covalent binding and toxicity of chemicals in the Bowman's glands in the olfactory mucosa, in the sero-mucous glands in the nasal septum and in the lateral nasal glands and maxillary glands around the maxillary sinuses are discussed. Light microscopic autoradiographic studies have demonstrated a selective covalent binding of nasal toxicants and carcinogens such as halogenated hydrocarbons and N-nitrosamines, especially in the Bowman's glands following a single systemic exposure, suggesting a high rate of metabolic activation of chemicals in these glands. Special attention is put on the herbicide dichlobenil which induces necrosis in the olfactory mucosa following a cytochrome-P450-mediated metabolic activation and covalent binding in the Bowman's glands.  相似文献

Abstract. Dynamics in hydrology and water chemistry in the Biebrza mires (Poland) were examined by means of a sampling survey that was repeated four times between 1987 and 1992. The dynamics in the vertical stratigraphy of water types in the peat profile are considerable from close to the mire surface to a depth of 50 cm. Water composition in the root zone correlated best with vegetation types during extremely dry or wet conditions. In the root zone of groundwater-fed rich fens with Caricetum limoso-diandrae and Calamagrostietum strictae vegetation, specific groundwater types evolve from the interaction of discharging groundwater from below the root zone and the temporal influence of precipitation and evapotranspiration. The Caricetum limoso-diandrae is fed by the continuous discharge of nutrient-poor, relatively mineral-rich water. The site conditions in the Calamagrostietum strictae are determined by occasional flooding and the presence of discharging mineral-poor groundwater in the lower part of the root zone. In the Caricetum limoso-diandrae and the Calamagrostietum strictae the maximum variations in water level were 56 and 86 cm, respectively. The composition of shallow groundwater of the Betuletum humilis/Caricetum rostratodiandrae fen is diluted most compared to other vegetation types by rainwater in wet periods. In periods of prolonged drought it has a water type that is affected by evapotranspiration and peat mineralisation. The water level varies by only 33 cm. In the Magnocaricion and Glycerietum maximae in the floodplain the water composition is determined by spring flooding of the river and the natural draw-down that occurs in the following summer. Here, maximum variations in water level were 108 and 117 cm, respectively.  相似文献

The paper couples the results obtained by applying the expert and the rapid Macrophyte Quality Indices set up to assess the ecological status of the Italian transitional environments according to the requirements by the Water Framework Directive (2000/60/CE). The indices were validated by comparing the composition of the macrophyte assemblages and the values of some bio-physico-chemical parameters of the water column of 20 stations of the Venice lagoon sampled monthly for one year between 2003 and 2005. In 5 stations out of the 20, the ones which fall within the 5 classes of ecological status suggested by the Water Framework Directive, sedimentation rates, sediment grain-size, and nutrient and pollutant (metals, Polychloro-Dibenzo-Dioxins/Furans, Polycyclic Aromatic Hydrocarbons, Pesticides and Polychlorinated Biphenils) concentrations in surface sediments were also determined. Results showed strong relationships between the trends of these environmental parameters and the composition and structure of macrophyte associations, as well as with the Macrophyte Quality Index assessment. Chlorophyceae showed a trend opposite to Rhodophyceae whose presence was concentrated in oxygenated and transparent environments. Chlorophyceae and the species characterised by low scores prevailed in turbid areas where nutrient and pollutant concentrations were high. Results allowed the identification of the conditions of the “reference sites” (confinement areas and sites with high water renewal) and the integration of the dichotomic key used for the application of the R-MaQI. Infections with the parasitic helminth, Nippostrongylus brasiliensis, cause changes in rat small intestinal goblet cell mucin, particularly in the peripheral sugar residues of oligosaccharide. These changes may correlate with expulsion. In this study, we examined changes in mucin oligosaccharides caused by primary infection and reinfection with N. brasiliensis, using two monoclonal antibodies, HCM31 and PGM34, that react with sialomucin and sulfomucin, respectively. Enzyme-linked immunosorbent assay of jejunal mucins showed that the relative reactivity of mucins with HCM31, but not PGM34, increased up to 16 days after primary infection and 6 days after reinfection, the times when the worms were expelled from the rats. Immunohistochemical studies confirmed that goblet cells stained with HCM31 greatly increased at the time of worm expulsion. These results indicate that the marked increase observed in HCM31-reactive sialomucins may be related to expulsion of the worms.  相似文献

目的:观察电针胃经穴对大鼠胃黏膜损伤修复的血清蛋白质差异表达,为进一步阐明针刺效应的体液机理提供理论依据。方法:用表面增强激光解吸离子化飞行时间质谱(SELDI—TOF—MS)技术和WCX2(弱阳离子交换芯片)对正常大鼠血清和电针大鼠血清进行蛋白质指纹图谱检测分析,通过Biomarker Wizard和Biomarker Patterns System软件判别分析处理数据并结合生物信息学方法筛选差异表达蛋白质。结果:与正常大鼠血清比较,电针大鼠血清蛋白质在质荷比为2000-50000有25个蛋白质峰差异有显著意义,其中有19个标志蛋白在电针胃经大鼠血清中呈现高表达,6个标志蛋白在电针胃经大鼠血清中呈现低表达。结论:电针可促进胃黏膜损伤大鼠血清蛋白质差异表达,这种差异蛋白质可能与电针促进胃黏膜损伤修复效应密切相关。  相似文献

观察木瓜三萜对吲哚美辛致胃黏膜损伤小鼠胃酸分泌及胃黏膜屏障的影响,在此基础上探讨其可能的机制。实验时,将小鼠随机分为正常组、模型组、木瓜三萜(50、100mg/kg)和奥美拉唑(20mg/kg)组。将给药组灌胃给予相应的药物,正常组和模型组灌胃给予0.5%羧甲基纤维素钠溶液,给药6小时后,除正常组外,灌胃给予20mg/kg的吲哚美辛,每天一次,连续7天。末次给药次日,小鼠用水合氯醛麻醉后,固定,剪开腹腔,进行胃黏膜血流量的测定,然后取胃检测胃液量、胃液酸度和胃结合黏液量;检测胃黏膜中表皮生长因子基因(EGF)和三叶因子1基因(TFF1)的mRNA和蛋白表达。研究发现:吲哚美辛致胃黏膜损伤模型组小鼠胃液分泌量,胃液酸度、胃黏膜血流量、胃结合黏液量及胃黏膜组织中EGF和TFF1的mRNA和蛋白表达明显降低,与正常组比较均具有统计学差异(P<0.01);用木瓜三萜预处理后,上述异常的变化均得到了有效逆转,与模型组比较具有显著性差异(P<0.05,P<0.01)。实验结果表明木瓜三萜(50、100mg/kg)对吲哚美辛致小鼠胃黏膜损伤具有较好的保护作用,通过上调EGF和TFF1的表达水平,增加胃液分泌量、胃液酸度、胃黏膜血流量、胃结合黏液量,恢复胃黏膜防御屏障的功能可能是其治疗吲哚美辛致胃黏膜损伤的机制之一。  相似文献
