我们利用水稻苯丙氨酸解氨酶启动子(AQ630)和吲哚乙酰胺赖氨酸酯合成酶基因(iaaL)构建了维管束特异表达的植物表达载体pBAL1并导入烟草基因组中。比较了对照植株、转基因植株的幼茎和根外植体在组织培养中分化能力的变化,结果表明转基因植株茎外植体的不定芽形成明显受到促进,而转化植株根外植体在不定根发生方面对外源IAA的敏感性下降。  相似文献

Germination, growth, and physiological responses of hybridizing Carpobrotus from coastal California to soil salinity were studied. Hybrids are presumably the result of hybridization and introgression between the exotic Carpobrotus edulis, a succulent perennial invading coastal habitats, and the native or long-naturalized C. chilensis. Germination responses were investigated at 0, 10, 20, and 50% seawater. Seedling growth and physiology were compared by irrigating seedlings with solutions of the same seawater concentrations and in low and high nutrients. Germination was inhibited in the presence of salt, but recovered after transferring the seeds to fresh water. Seeds exposed to salt had higher final germination rates than control. Growth of Carpobrotus was slightly enhanced by low seawater concentrations but reduced at high salinity at both nutrient regimes. Leaf cell sap osmolarity increased with increasing soil salinity, and taxa did not differ significantly in this physiological adjustment. Leaf carbon isotope ratios (∂13C) ranged from −28 to −22‰ and became less negative at higher salinities, indicating an improved water use efficiency in the seedlings at high salt concentrations. In addition, ∂13C values were generally less negative at high than at low nutrients. Differences among taxa were generally small. The results show that salinity affects both establishment and growth of hybridizing Carpobrotus. The overall weak species differences in salt tolerance indicate that the exotic C. edulis can occupy the same sites as C. chilensis in terms of salinity. The similarity of hybrids in their response to salinity suggests that they may contribute to the invasion by Carpobrotus.  相似文献

By incorporating Ball-Berry model of stomatal conductance into the models of photosynthesis and transpiration, a model of leaf water use efficiency (WUE) as affected by several environmental variables [irradiance (Ⅰ), vapor pressure deficit (VPD) and atmospheric CO2 concentration (Ca) ] was constructed. Because the environmental variables influenced the photosynthetic rate and transpiration rate in different ways, the changes of leaf WUE with these factors were quite complicated. The rates of photosynthesis and transpiration of wheat leaves were also measured in the phytotron where the environmental variables were kept within certain ranges, and leaf WUE was calculated therefrom. The results of simulation fit quite well with the measurements except at high Ca.  相似文献

Photosynthetic rate and quatum efficiency of grapevine (Vitis vinifera L. cv. Sauvignon blanc) leaves were measured under the field with ample soil water supply, and in phytotron with ample supply of water and mineral nutrients, constant air humidity and CO2 concentration, and optimum air temperature, respectively. Under field conditions CO2 assimilation quantum efficiency of leaves reached its maximum in the morning, which was followed by continuous decrease and midday depression. The leaves intercepting more light energy in the morning showed a higher quantum efficiency. Those leaves subjected continuously to strong irradiance exhibited a more obvious and longer midday depression. Reduction of leaf light interception around midday could reduce midday depression. Shaded leaves had a higher quantum efficiency than leaves under direct sunlight. The diurnal changes in photosynthetic rate and quantum efficiency of leaves were shown to be closely related to the variations in mesophyll resistance to CO2. In phytotron experiments the photosynthetic quantum efficiency of leaves was reduced after a certain period of illumination not only at 1200 μmol · m-2 · s-1 PFD, higher than the saturating light of vine leaves (≈1000 μmol · m-2 · s-1), which was caused by "photoinhibition”, but also at 800 and 200μmol · m-2 · s-1, which was similar to "photoinhibition”. But photosynthetic quantum efficiency of leaves exposed continuously to a very weak PFD (100 μmol · m -2 · s-1) remained contant. The diurnal changes in mesophyll resistance to CO2 of vine leaves could be partly related to photoinhibition. It is considered that, under field conditions without soil water limitation, midday depression of vine leaf photosynthesis could be a result of an increase of the mesophyll resistance induced by multiple effects of strong light, high temperature and low humidity. A higher light interception by canopy plane in the morning may be advantageous to exploit higher photosynthetic potentiality of leaves, but a lower light interception in the middle of day may reduce midday depression. The north-south orientation plane can provide optimum light regime and improve photosynthetic environment in vineyards.  相似文献

The indoleacetic acid-lysine synthetase (iaaL) gene of Pseudornonas syringae subsp. savastanoi was fused to a rice Rchl0 promoter and introduced into tobacco plants. The expression pattern of this chimeric Rchl0-iaaL gene in the transgenic plants was studied, and the endogenous levels of indoleacetic acid (IAA) were assayed with the high-performance liquid chromatography (HPLC) method. The results showed that the Rchl0 promoter could direct the high level expression of the iaaL gene in young inflorescences, fully bloomed flowers, and immature fruits, resulted in a decrease of the IAA levels by 32%–82%. The transgenic plants displayed decreased apical dominance, enlongated stamens, abnormal inflorescences and fruits. The transgenie plant pYL614-T14, in which the endogenous IAA levels in inflorescences were reduced by 82 %, displayed extremely abnormal inflorescence with only two flowers and a much higher potential of vegetative growth than that of the controls after flowering. These Rchl 0-iaaL gene transgenic tobacco plants would pro- vide valuable materials for further studies of physiological functions of IAA in reproductive development.  相似文献

为了探讨癌细胞mtDNA控制区序列的变化特征, 采用PCR产物限制性片段长度多态性(PCR-RFLP)分析与直接测序相结合的方法,对比分析6株人癌细胞系、 6例癌患者及4例健康成人白细胞mtDNA控制区序列。发现第16519位T→C、16 534位A→G、46位T→G和49位A→C突变, 在癌细胞系和癌患者白细胞mtDNA中分别占50%(3/6)和33.3%(2/6), 健康成人白细胞mtDNA中未见此类型突变;第16 278位C→T突变,在癌细胞系mtDNA中占50%(3/6),显著高于正常人群mtDNA中此位点的多态性变异。表明癌细胞和癌患者白细胞mtDNA重链复制起点及其 相邻D环区的特征性突变可能与细胞癌变/或癌的易感性有关。 Abstract: To explore the sequence feature of mitochondrial DNA(mtDNA) control region in human carcinoma cells, polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) and direct sequence techniques were used to analyze the sequence of mtDNA control region of 6 human carcinoma cell lines versus white blood cells which from 6 tumor patients and 4 normal adults. The T to C mutation at np 16 519, A to G mutation at np 16 534, T to G mutation at np 46, and A to C mutation at np 49 was found in 50% (3/6 cases) of carcinoma cell lines and in 33.3%(2/6 cases) of tumor patients, but it was not found in normal adults. The C to T mutation at np 16 278 was found in 50%(3/6 cases) of carcinoma cell lines, it was significantly higher than that of the polymorphism of normal population. These findings suggest that the typical mutation in the starting area of heavy-strand replication and the first half of D-loop region might probably be associated with carcinogenesis or susceptibility of carcinoma.  相似文献

1. The photosynthetic response to elevated CO2 and nutrient stress was investigated in Agrostis capillaris, Lolium perenne and Trifolium repens grown in an open-top chamber facility for 2 years under two nutrient regimes. Acclimation was evaluated by measuring the response of light-saturated photosynthesis to changes in the substomatal CO2 concentration.
2. Growth at elevated CO2 resulted in reductions in apparent Rubisco activity in vivo in all three species, which were associated with reductions of total leaf nitrogen content on a unit area basis for A. capillaris and L. perenne . Despite this acclimation, photosynthesis was significantly higher at elevated CO2 for T. repens and A. capillaris , the latter exhibiting the greatest increase of carbon uptake at the lowest nutrient supply.
3. The photosynthetic nitrogen-use efficiency (the rate of carbon assimilation per unit leaf nitrogen) increased at elevated CO2, not purely owing to higher values of photosynthesis at elevated CO2, but also as a result of lower leaf nitrogen contents.
4. Contrary to most previous studies, this investigation indicates that elevated CO2 can stimulate photosynthesis under a severely limited nutrient supply. Changes in photosynthetic nitrogen-use efficiency may be a critical determinant of competition within low nutrient ecosystems and low input agricultural systems.  相似文献
