Type II protein secretion systems (T2SS) are molecular machines that promote specific transport of folded periplasmic proteins in Gram-negative bacteria, across a dedicated channel in the outer membrane. Secreted substrates, released to the milieu or displayed on the cell surface, contribute to bacterial adaptation to a range of habitats, from deep-sea waters to animal and plant tissues. The past decade has seen remarkable progress in structural, biochemical and functional analysis of T2SS and related systems, bringing new mechanistic insights into these dynamic complexes. This review focuses on recent advances in the field, and discusses open questions regarding the secretion mechanism. To study effects of traffic during daytime and other human activities, we conducted surveys on the migration of Tibetan antelope Pantholops hodgsoni along the Qinghai-Tibet highway between Kunlun Mountain Pass to Wudaoliang,using line transects (LT) from June to September in 2001 and 2002, Data were collected for 64 days with 17 days in 2001, and 47days in 2002. The Stationary Observation Method (SOM) was also adopted for the investigation of status of the disturbance from traffic when antelopes passing the highway and the traffic situation from August 6 to 28 (6 : 30-19: 00), 4 to 20 (6: 00- 18: 00) and from December 21 to 29 (9:00- 19: 00) in 2002. The results showed that,the period of migration in 2002 was more prolonged than in 2001, and it took at least 45 min for each group to cross the highway, with success ratio of only 30.2 %, and the traffic flow of summer daytime was 76/h, while the winter‘s traffic flow was 29/h. Highway and rail traffic should be managed during daylight hours from June to August, to allow for the migration of the antelopes. Also conservation education and a further survey should be emphasized for the protection on the species [Acta Zoologica Sinica 50 (4) : 669-674, 2004].  相似文献

国家公园内人与野生动物冲突严重,建立野生动物肇事保险机制是减缓冲突的重要举措。以大熊猫国家公园周边社区为例,分析建立国家公园以及农户风险偏好对野生动物肇事保险购买意愿的影响,并探讨冲突程度和制度信任的中介效应。研究结果表明建立国家公园显著增加了农户野生动物肇事保险购买意愿,而农户风险偏好对保险购买意愿会产生负向显著影响。冲突程度和制度信任是重要的中介变量,冲突程度和制度信任均对农户保险购买意愿产生正向显著影响。建立国家公园显著增加了人与野生动物冲突严重程度,进而对保险购买意愿产生正向影响。此外,建立国家公园显著降低了农户制度信任,进而对保险购买意愿产生负向影响。农户风险偏好对野生动物肇事损失程度产生正向显著影响,进而对保险购买意愿产生正向影响。农户风险偏好对制度信任影响并不显著。基于此,研究提出吸纳社会资本参与,拓宽野生动物肇事保险资金来源,开展社区共管、地役权补偿等措施提升社区信任以及建立基于农户自主防护行为的野生动物肇事保险机制等政策建议。  相似文献

Abstract The cydD gene of Escherichia coli encodes a protein which, together with the CydC protein, probably constitutes a heterodimeric, ABC-family membrane transporter, necessary for biosynthesis of the cytochrome bd quinol oxidase. Here, we demonstrate that a cydD mutant also fails to synthesise periplasmic c -type cytochrome(s), suggesting that the transporter exports haem or some other component involved in assembly of cytochromes that are found in, or exposed to, the periplasm. The CydDC system appears to be the first example of a transporter required for periplasmic cytochrome assembly processes requiring more than one type of haem. A mutant defective in trxB (adjacent to the cydDC operon, and encoding thioredoxin reductase) was unaffected in cytochrome c or bd assembly.  相似文献

A model for a parallel evolution of the genetic code and protein synthesis is presented. The main tenet of this model is that the genetic code, that is, a correspondence between nucleotide and aminoacid coding units, originated from sequence-specific interaction between abiotically synthesized polynucleotides and polypeptides. A sequence-specific binding between oligonucleotides and oligopeptides is supported by experimental findings. Moreover, it is parsimonious enough to be consistent with the relatively simple chemistry of a primordial environment. Proximity between peptides and RNA increased the rate of formation of ester bonds between them. This lead to the accumulation of sequence-specific polypeptide-polynucleotide pairs, that is, of primordial-loaded tRNA. Condensation of short polypeptides into longer products could be catalyzed by a sequence-specific juxtaposition of loaded tRNA over complementary RNA, originating the core of protein synthesis. The accumulation of useful encoded products, for example, catalysts for tRNA loading (primordial aminoacyl-tRNA synthetases) or stabilizers of tRNA-mRNA interactions (primordial ribosomes), permitted the subsequent evolution of protein synthesis and of the genetic code to their mature form. This occurred via a parallel reduction in length of the interacting polynucleotides and polypeptides. Thus, it maintained the correct reading frame of mRNA from the preceding stages of evolution. Members of the ABC transporter superfamily contain two nucleotide binding domains. To date, the three dimensional structure of no member of this super-family has been elucidated. To gain structural insight, the known structures of several other nucleotides binding proteins can be used as a framework for modeling these domains. We have modeled both nucleotide binding domains of the protein CFTR (Cystic Fibrosis Transmembrane Conductance Regulator) using the two similar domains of mitochondrial F1-ATPase. The models obtained, provide useful insights into the putative functions of these domains and their possible interaction as well as a rationale for the basis of Cystic Fibrosis causing mutations. First, the two nucleotide binding domains (folds) of CFTR are each predicted to span a 240–250 amino acid sequence rather than the 150–160 amino acid sequence originally proposed. Second, the first nucleotide binding fold, is predicted to catalyze significant rates of ATP hydrolysis as a catalytic base (E504) resides near the phosphate of ATP. This prediction has been verified experimentally [Ko, Y.H., and Pedersen, P.L. (1995) J. Biol. Chem. 268, 24330-24338], providing support for the model. In contrast, the second nucleotide binding fold is predicted at best to be a weak ATPase as the glutamic acid residue is replaced with a glutamine. Third, F508, which when deleted causes 70% of all cases of cystic fibrosis, is predicted to lie in a cleft near the nucleotide binding pocket. All other disease causing mutations within the two nucleotide binding domains of CFTR either reside near the Walker A and Walker B consensus motifs in the heart of the nucleotide binding pocket, or in the C motif which lies outside but near the nucleotide binding pocket. Finally, the two nucleotide binding domains of CFTR are predicted to interact, and in one of the two predicted orientations, F508 resides near the interface.This is the first report where both nucleotide binding domains of an ABC transporter and their putative domain-domain interactions have been modeled in three dimensions. The methods and the template used in this work can be used to analyze the structures and function of the nucleotide binding domains of all other members of the ABC transporter super-family.  相似文献

The ecosystems of Chernobyl nuclear accident zone, having been deeply transformed by normal long-term agricultural and forestry activity, have been damaged by irradiation after the nuclear accident of 1986 and then influenced by post-accident countermeasures. Ecosystem remediation is being undertaken in this area, including maintenance of the forests, artificial afforestation, and processes of spontaneous self-restoration of the natural coenoses. Both natural and artificial remediation of the landscapes and ecosystems at the abandoned territory improves their capacity to retain radionuclides and their ability to stabilise the processes of redistribution of radionuclides and their spread in the environment. The Chernobyl zone is a territory of great importance for studying ecosystem remediation processes and elaborating on ecological engineering technologies to correct serious environmental problems arising from radionuclides release.  相似文献

澳门路凼连贯公路西侧的湿地保护区是黑脸琵鹭(Platalea minor)重要的越冬地之一,每年的越冬数量稳定增长。2007~2009年度2个冬季的调查结果显示,黑脸琵鹭每年11月初至次年4月底在澳门越冬,12月至翌年2月份种群数量达到最大,超过50只。其中人工湿地是黑脸琵鹭主要的休息场所,沿岸滩涂是其重要觅食地。黑脸琵鹭昼间栖息行为以休息为主,其次是护理行为。栖息地周边主要人为干扰为噪声污染,但噪声水平较低,经噪声预测模型计算出传播至黑脸琵鹭停歇处的噪声水平为45.4dB(A)及46.5dB(A),低于鸟类耐受阈值,警觉受惊行为多数由大型牵引车和直升飞机引起。随着保护区周边地区的开发,车流量将进一步增加,建议加强保护区的科学管理,控制周边交通干线的车流量,并调整直升飞机飞行路线,尽量绕行保护区上空,以减少对黑脸琵鹭栖息的干扰。  相似文献

The authors indicate the origin of the data used by the French Public Health Authority in 1986 to estimate the risk of the radioactive fall out following the Chernobyl accident. The technical means developed in order to establish this data, and the precedent experience gained, are described. The principal results are given. The terms of the 28 May 1986 note to the Academy of Sciences by R. Latarjet, which concluded that the low level of this fallout did not justify any countermeasure, except the control of the imported food, are confirmed. Rational dispositions are required in order to improve the information of the population, referring to the Swedish model, which involves the intervention of the medical staff specialized in radiotoxicology, radiopathology, nuclear medicine, and especially trained.  相似文献

White rats were irradiated by both 131iodine, which was incorporated into the thyroid gland, and 137cesium; these were dosed at about 10 and 6 Gy, respectively. A portion of the experimental animals was fed a diet enriched with powdered Laminaria japonica of known composition. In animals provided with this powder, the frequency of neoplasms was lessened, and the latent period for the major types of tumours was increased, compared to rats not given the powder.
