Abstract: New details of F-actin organisation in leaf epidermal and stomatal cells were revealed by rhodamine — and fluorescein — phalloidin staining of fixed epidermal peels of Tradescantia virginiana and visualisation by confocal microscopy. Non-specialised epidermal cells contain highly organised arrays of fine cortical actin filaments aligned in transverse or oblique orientations. In interphase guard mother cells (GMCs), the arrangement of cortical F-actin changes on the periclinal and anticlinal cell walls at different times during differentiation. Initially, cortical F-actin on the periclinal surfaces is oriented transversely and F-actin is evenly distributed around the anticlinal walls. Following polarisation of the adjacent subsidiary mother cells (SMCs), actin in GMCs concentrates on the lateral anticlinal walls, but not on the transverse walls. Subsequently, F-actin on the periclinal walls reorients to radial and then longitudinal. Organisation of F-actin in SMCs appears to be influenced by the adjacent GMCs and co-ordination in F-actin arrangements in cells of the stomatal complex continues through to the formation of the guard cell pair. Our studies indicate that actin bands marking the division site in prophase cells, and detected in microinjected living material, are a particularly labile subset of F-actin. Actin bands were difficult to preserve, even when aldehyde fixation was avoided, in contrast to all interphase and mitotic F-actin.  相似文献

Summary Self-incompatibility (SI) is reported for an accession ofTradescantia ohiensis. Pollen tube inhibition is stigmatic. The stigma is wet and papillate, the papillr surface bearing conspicuous blebs particularly in the mid- and basal regions of the cell. A proteinaceous pellicle is present on the surface of the papillar cuticle. The penetration of the stains calcofluor white and alcian blue into the cell wall of fresh stigma papillae strongly indicates that the permeability of the papillae is greatest at the mid-region of the cell and not at the tip. When single pollen grains are attached to the tip of a papilla there is either no response at all or the pollen adheres to the papilla. When attached in the mid-region pollen adheres, and often germinates. It is concluded that the sites of pollen receptivity are the mid- and basal regions of the papular cell.  相似文献

The numbers of major chromosome arms of these cytotypes give a nombre fondamentaP series of 14, 28, 42 and 56 which, in combination with meiotic analyses, indicates plants which, in genetical terms at least, are diploid, tetraploid, hexaploid and octoploid. This series has evolved from a 2 n = 14 acrocentric or telocentric karyotype by a combination of Robertsonian fusion and polyploidy. Pseudo-iso-chromosomes are sometimes formed in this evolutionary development and can persist as stable members of normal complements.  相似文献

采用体外渗透和显微注射的方法。将植物微管特效解聚剂甲基氨草磷(APM)引入紫露草雄蕊毛细胞后,发现原来沿着胞质束运动的胞质颗粒运动速度渐慢,进而胞质束消失,颗粒运动停止。显微注射后,还发现APM可通过胞间通道由被注射的细胞向两侧细胞扩散,从而也导致两侧细胞胞质束消失,颗粒运动停止。APM对胞质环流的抑制作用是可逆的。结果表明微管可能是胞质束的重要组份之一,植物胞质环流与微管的聚合与解聚状态有密切关系。  相似文献

Summary The two human homologues of the fission yeast cell cycle protein p13 suc1 displayed structural characteristics consistent with their existing in solution as differently folded monomers despite 81% identity with respect to their primary structures and both being capable of fulfilling the functions of their homologues in fission and budding yeasts. Carboxyfluorescein-labelled p9 CksHs1 and p9 CksHs2 retained their native structures. When microinjected into live stamen hair cells ofTradescantia virginiana, the labelled proteins accumulated in the nuclei of the cells. Summary. Calcium oxalate crystals are by far the most prevalent and widely distributed mineral deposits in higher plants. In Tradescantia pallida, an evergreen perennial plant widely used as an ornamental plant, calcium oxalate crystals occur in the parenchymal tissues of stem, leaf, and root, as well as in flower organs, in the form of either raphides or tetragonal prismatic crystals or both. Energy-dispersive X-ray analysis revealed that C, O, and Ca were the main elements; and K, Cl, and Si, the minor elements. Infrared and X-ray analyses of crystals collected from these tissues detected the coexistence of two calcium oxalate chemical forms, i.e., whewellite and weddellite, as well as calcite, opal, and sylvite. Here, we show for the first time the occurrence of epitaxy in mineral crystals of plants. Epitaxy, which involves the oriented overgrowth of one crystal onto a second crystalline substrate, might explain how potassium chloride (sylvite) – one of the most water-soluble salts – stays insoluble in crystal form when coated with a calcium oxalate epilayer. The results indicate the potential role of crystals in regulating the ionic equilibrium of both calcium and potassium ions. 肌动蛋白间接免疫荧光标记实验观察到紫鸭跖草花粉母细胞减数分裂前期Ⅰ细胞核,中期┘染色体,二分体和四分体均发现较明亮的黄绿色荧光,说明其中含有肌动蛋白。经TRIT-C鬼等环肽荧光标记后,减数分裂前Ⅰ细胞核,二分枝及中期Ⅰ染色体均发现较明亮的红色荧光;CD处理后细胞核和染色体的荧光明显减弱或没有荧光,说明其中存在F-肌动蛋白。  相似文献

A study of stomatal mechanics using the cell pressure probe   总被引:9,自引:2,他引:7  
The relationship between stomatal aperture ( a ) and guard cell pressure ( P g) was measured directly in four different species ( Vicia faba, Tradescantia virginiana, Ginkgo biloba and Nephrolepis exaltata ) using a special cell pressure probe technique. The effect of epidermal turgor ( P ep) on this relationship was also measured in T. virginiana . The relationship was sigmoidal for V. faba and T. virginiana , but entirely convex for G. biloba and N. exaltata. Epidermal turgor was found to have a pronounced closing effect on stomata of T. virginiana . Maximum aperture with full epidermal turgor (0·92 MPa) was about half that with zero epidermal turgor. Also, with full epidermal turgor stomata of T. virginiana did not begin to open until P g was more than 1·25 MPa. These characteristics were used to develop an expression for a as a function of P g and P ep. Results for the different species are compared and discussed in terms of possible advantages and limitations of water economy.  相似文献
