In water-stressed leaves, accumulation of neutral osmotica enhances the heat tolerance of photosynthetic electron transport. There are large diurnal and day-to-day changes in leaf sugar content because of variations in net photosynthetic production, respiration and retranslocation. To test the hypothesis that diurnal and day-to-day variations in leaf sugar content and osmotic potential significantly modify the responses to temperature of photosynthetic electron transport rate, we studied chlorophyll fluorescence rise temperatures (i.e. critical temperatures at break-points in fluorescence versus temperature response curves, corresponding to enhanced damage of PSII centers and detachment of pigment-binding complexes) in the dark at a background of weak far-red light (T(FR)) and under actinic light (T(L)), and responses of foliar photosynthetic electron transport rate to temperature using gas-exchange and chlorophyll fluorescence techniques in the temperate tree Populus tremula L. Sucrose and sorbitol feeding experiments demonstrated strong increases of fluorescence rise temperatures T(FR) and T(L) with decreasing leaf osmotic potential and increasing internal sugar concentration. Similar T(FR) and T(L) changes were observed in response to natural variation in leaf sugar concentration throughout the day. Increases in leaf sugar concentration led to an overall down-regulation of the rate of photosynthetic electron transport (J), but increases in the optimum temperature (Topt) of J. For the entire dataset, Topt varied from 33.8 degrees C to 43 degrees C due to natural variation in sugars and from 33.8 degrees C to 52.6 degrees C in the sugar feeding experiments, underscoring the importance of sugars in modifying the response of J to temperature. However, the correlations between the sugar concentration and fluorescence rise temperature varied between the days. This variation in fluorescence rise temperature was best explained by the average temperature of the preceding 5 or 6 days. In addition, there was a significant year-to-year variation in heat sensitivity of photosynthetic electron transport that was associated with year-to-year differences in endogenous sugar content. Our data demonstrate a diurnal variation in leaf heat tolerance due to changes in sugar concentration, but they also show that this short-term modification in heat tolerance is super-imposed by long-term changes in heat resistance driven by average temperature of preceding days.  相似文献

A GC/EIMS/SIM methodology has been developed to re-examine the path of carbon in photosynthesis. Exposing isolated spinach chloroplasts to 13CO2 on a solid support for a defined period followed by quenching and work-up provided a mixture of labelled sugar phosphates. After enzymatic dephosphorylation and derivatization, the Mox-TMS sugars were analysed using the above method. The purpose of the study was to try to calculate the atom% enrichment of 13C in as many of the individual carbons in each of the derivatized sugars as was practical using diagnostic fragment ions. In the event, only one 45 s experiment provided sufficient data to enable a range of enrichment values to be calculated. This confirmed that D-glycero-D-altro-octulose phosphate was present in the chloroplasts and was heavily labelled in the C4, C5 and C6 positions, in keeping with the hypothesis that it had an inclusive role and a labelling pattern consistent with a new modified pathway of carbon in photosynthesis.  相似文献

花蜜的研究是花生物学中的一个重要内容,探寻实用的方法将方便野外操作。我们分别还用毛细管、注射器、滤纸条和离心法采集了5种花的花蜜,以比较各种方法的优劣,并用3种旋光测糖仪测量了慈姑Sagittaria trifolia L,的雌雄花的花蜜糖含量。目的是为寻找一种适合小型花的花蜜采集测量方法。结果表明,几种方法的适用性受花的大小、形状、蜜的分泌量及蜜腺位置的影响非常大,不同的花要采用不同的方法。对于一般的野外工作建议用毛细管采集后使用便携式旋光测糖仪测其糖含量。特别小的花和蜜量微小的花可以采用离心法收集。  相似文献

Optical activity is directly related to molecular conformation through the anisotropic polarizabilities of molecules and the refractive index of materials. L-amino acids and D-sugars are characteristic essential bioactive molecules. Since molecular recognition and enzyme activity are related to the conformation of substrates, the relevance of optical activity to biological processes is evident. Specific ion, or Hofmeister, effects that occur with electrolytes at moderately high concentrations modify the behavior of interfaces, molecular forces between membranes, of bulk solutions, of enzymes, and even of DNA. Such effects are universal. Here we report a study on the change in optical rotation induced by some sodium salts for the enantiomers of serine and glucose in water solution. The optical rotation is shown to depend on the kind of anion and on the salt concentration. To obtain further insights into the mechanism behind the phenomenon, Fourier transform infrared (FTIR) spectral studies of serine and glucose solutions in electrolytes were also carried out. The results suggest that it is the differences in interactions of anions at specific chemical sites of the solutes that are responsible for the effects. These forces depend strongly on anion polarizability in water. Such specific ion preferential interactions can affect conformation and internal field, and result in significant changes in optical rotation.  相似文献

  • 1 Accurate and sensitive survey and monitoring methods are needed for shrews. We present a new design of hair tube and a new, simple method of species identification from multivariate analysis of four parameters measured from shrew guard hairs using a binocular microscope with incident light.
  • 2 Multivariate analysis of these parameters measured from hairs of known identity showed that they can be used to identify hair to the species level with 85% accuracy.
  • 3 We compared our indices of abundance from hair tubes (the hair tube index) with those from live trapping in 40 field margins. Capture‐mark‐recapture methods showed that capture rate did not vary systematically across sites, so that number of individuals captured was used as an index of abundance.
  • 4 The hair tube index showed a significant association with the number of individuals captured for Sorex araneus and Neomys fodiens. The lack of a significant association for Sorex minutus may be because hair tubes are more sensitive in detecting this species than live trapping.
  • 5 Hair tubes have additional advantages over live trapping, since they do not require frequent checking, are much lighter and cheaper than live traps, and no licence is required for their use in the UK. We therefore recommend consideration of their use in future surveys and monitoring studies of shrews. We provide an equation so that other researchers can use our multivariate method.

BACKGROUND AND AIMS: Floral nectar concentration and chemical composition of 26 plant species native to the temperate forest of southern South America are reported and the relationships with the flower type are evaluated. METHODS: Nectar concentration was measured with a hand refractometer and sugar composition was analysed by gas-liquid chromatography. Plant species were classified into flower type categories based not only on floral features but also on data from the literature and field observations on their pollinators. KEY RESULTS: Most data on nectar are new reports at the generic and/or specific level. Plant species in which more than one population was studied showed significant among-population variation in nectar sugar concentration and composition. Results showed a weak relationship between nectar traits and flower type. Many species had nectar containing 50 % or more sucrose (17 of 26 species), independent of the main pollinator. CONCLUSIONS: Considering that (a) nectar characteristics did not show a clear association with different flower types or with plant taxonomic membership, and (b) different populations of the same species showed large variability in sugar composition, the results suggest that other factors (e.g. historical and environmental) could be involved in determining the sugar composition of the highly endemic plant species from this region.  相似文献

Plant meristems are utilization sinks, in which cell division activity governs sink strength. However, the molecular mechanisms by which cell division activity and sink strength are adjusted to a plant's developmental program in its environmental setting are not well understood. Mitogenic hormonal as well as metabolic signals drive and modulate the cell cycle, but a coherent idea of how this is accomplished, is still missing. Auxin and cytokinins are known as endogenous mitogens whose concentrations and timing, however, can be externally affected. Although the sites and mechanisms of signal interaction in cell cycle control have not yet been unravelled, crosstalk of sugar and phytohormone signals could be localized to several biochemical levels. At the expression level of cell cycle control genes, like cyclins, Cdks, and others, synergistic but also antagonistic interactions could be demonstrated. Another level of crosstalk is that of signal generation or modulation. Cytokinins affect the activity of extracellular invertases and hexose-uptake carriers and thus impinge on an intracellular sugar signal. With tobacco BY-2 cells, a coordinated control of cell cycle activity at both regulatory levels could be shown. Comparison of the results obtained with the root cell-representing BY-2 cells with literature data from shoot tissues or green cell cultures of Arabidopsis and Chenopodium suggests opposed and tissue-specific regulatory patterns of mitogenic signals and signal crosstalk in root and shoot meristems.  相似文献

Transgenic potato ( Solanum tuberosum cv. Prairie) lines were produced over-expressing a sucrose non-fermenting-1-related protein kinase-1 gene ( SnRK1 ) under the control of a patatin (tuber-specific) promoter. SnRK1 activity in the tubers of three independent transgenic lines was increased by 55%−167% compared with that in the wild-type. Glucose levels were decreased, at 17%−56% of the levels of the wild-type, and the starch content showed an increase of 23%−30%. Sucrose and fructose levels in the tubers of the transgenic plants did not show a significant change. Northern analyses of genes encoding sucrose synthase and ADP-glucose pyrophosphorylase, two key enzymes involved in the biosynthetic pathway from sucrose to starch, showed that the expression of both was increased in tubers of the transgenic lines compared with the wild-type. In contrast, the expression of genes encoding two other enzymes of carbohydrate metabolism, α-amylase and sucrose phosphate synthase, showed no change. The activity of sucrose synthase and ADP-glucose pyrophosphorylase was also increased, by approximately 20%–60% and three- to five-fold, respectively, whereas the activity of hexokinase was unchanged. The results are consistent with a role for SnRK1 in regulating carbon flux through the storage pathway to starch biosynthesis. They emphasize the importance of SnRK1 in the regulation of carbohydrate metabolism and resource partitioning, and indicate a specific role for SnRK1 in the control of starch accumulation in potato tubers.  相似文献

采用生理生化分析方法研究了辣椒感染疫病后叶片中几个生化指标的变化。结果表明,染病前后感病品种叶片中可溶性总糖含量持续高于抗病品种;抗病类型品种和感病类型品种的可溶性蛋白含量变化规律均表现为先升高后下降,但接种前其叶片中可溶性蛋白含量两者间无明显差异;抗病类型和感病类型辣椒接种后保护酶活性均升高,而且感病类型的POD和ASP酶活性在接种后120h显著高于抗病类型;高抗类型叶片中PPO活性增加幅度显著大于感病类型,但抗病类型品种PPO活性上升趋势比较平缓。接种后,感病品种PAL活性上升幅度小于高抗品种,接种后96h PAL活性开始逐渐下降。可溶性总糖含量和苯丙氨酸解氨酶可以作为辣椒苗期抗疫病鉴定的生化指标。  相似文献

Endophyte-infected (EI) seeds of Lolium perenne L. were used to attain endophyte-free (EF) population by heating the seeds at 43°C for 15 min and then 57°C for 25 min. Relative water content (RWC), chlorophyll, soluble sugar and starch content of EI and EF populations under normal and drought stress conditions were compared to investigate the effect of endophyte infection on the host plant. Under severe stress, RWC of EI leaf was significantly higher than that of EF leaf, i.e. EI plants took more advantages over EF plants in water-holding ability. Under mild stress, endophyte could enhance soluble sugars in host plants to improve their osmotic ability. With stress intensification, the improvement of endophyte no longer existed, and more photosynthetic products (such as starch) accumulated in EI plants to survive through the undesirable conditions. In the next spring, EI populations will recover more rapidly than EF populations. The biomass of a population is closely related to its photosynthesis. Under severe stress, EI population significantly accumulated more biomass than EF population. As far as photosynthetic pigments were concerned, contents of Chla, Chlb and Car of EI plants were close to those of EF plants, which suggested that endophyte infection didn’t alleviate photosynthetic pigments from being destroyed by drought stress, and endophyte might improve photosynthesis ability of its host plant in other ways. __________ Translated from Acta Ecologica Sinica, 2005, 25 (2) [译自: 生态学报, 2005, 25(2)]  相似文献
