Summary The effect of Alternaria solani culture filtrate on adventitious shoot regeneration from tuber discs was evaluated using five potato cultivars, which were selected based on their field reaction to Alternaria solani and which represented a range of disease reactions. The antibiotic streptozotocin under a variety of growth conditions rapidly and irreversibly inactivates the capacity to divide or to form colonies of a series of sensitive bacteria, containing the phosphoenolpyruvate-dependent sugar-phosphotransferase system. Cells can be sensitized towards the drug by pregrowth in N-acetyl-glucosamine and can be protected by adding this amino-glucoside to the medium. Starvation for energy, especially for phosphoenolpyruvate, or prevention of the induction of a transport system involved in streptozotocin uptake will protect the cells, while a block in protein synthesis does not. The killed cells neither lyse, nor are they transformed into spheroplasts. At first, the capacity of such dead cells to respire, to swim actively or to keep the cytoplasmic membrane impermeable for small molecules remains intact. Their capacity for over-all RNA and protein synthesis, and for carbohydrate and amino acid uptake by facilitated diffusion or active transport is not affected. However, they loose rapidly their ability to take up carbohydrates by the phospheonolpyruvate dependent process of group translocation or to synthesize inducible enzymes, e.g. the enzyme -galactosidase. These inhibitory effects apparently are caused by the accumulation of phosphorylated, toxic derivates of the antibiotic and eventually lead to a pronounced bacteriostasis. To allow rapid identification of toxic shock syndrome toxin-1 (TSST-1)-producing Staphylococcus aureus strains, a real-time PCR assay for the detection of the tst gene, which encodes TSST-1, was developed. The assay was applied to S. aureus isolates from patients with Wegener's Granulomatosis (WG), as well as isolates that were classified as either community- (CA) or hospital-acquired (HA). No significant difference in the percentage of tst-positive strains was observed between isolates from WG patients and CA isolates (24% and 25%, respectively). In contrast, only 14% of the HA isolates were tst-positive (p<0.05). Investigation of the clonal relationship between tst-positive CA and HA strains could indicated the recent emergence of a virulent S. aureus clone in the community.  相似文献

Pr-IIGL(a), a derivative of the tetrapeptide beta-amyloid 31-34 (Abeta(31-34)), exerts controversial effects: it is toxic in a neuroblastoma culture, but it protects glial cells from the cytotoxic action of Abeta(1-42). For an understanding of this phenomenon, a new pentapeptide, RIIGL(a) was synthetized, and both compounds were studied by different physicochemical and biological methods. Transmission electron microscopic (TEM) studies revealed that Pr-IIGL(a) forms fibrillar aggregates, whereas RIIGL(a) does not form fibrils. Congo red binding studies furnished the same results. Aggregated Pr-IIGL(a) acts as a cytotoxic agent in neuroblastoma cultures, but RIIGL(a) does not display inherent toxicity. RIIGL(a) co-incubated with Abeta(1-42) inhibits the formation of mature amyloid fibres (TEM studies) and reduces the cytotoxic effect of fibrillar Abeta(1-42). These results indicate that RIIGL(a) is an effective inhibitor of both the aggregation and the toxic effects of Abeta(1-42) and can serve as a lead compound for the design of novel neuroprotective peptidomimetics.  相似文献

目的:评价盐酸多柔比星脂质体单药(PLD)与盐酸多柔比星脂质体联合洛铂治疗复发性卵巢癌的安全性和临床疗效。方法:收集2012年4月至2015年10月我科收治的31例复发晚期上皮性卵巢癌患者,根据患者是否存在铂类耐药分为多柔比星组(单药组)15例及多柔比星+洛铂组(对照组)16例。单药组给盐酸多柔比星脂质体50 mg/m~2,静滴;对照组给盐酸多柔比星脂质体20-30 mg/m~2,洛铂30-50 mg/m~2,静脉滴注,两组每21-28天重复一次,观察和比较两组的临床疗效和毒性反应的发生情况。结果:所有患者完成3-8周期,客观有效率(ORR)为38.7%。单药组为33.3%,对照组为占43.8%,两组ORR比较差异无统计学意义(P=0.411)。单药组骨髓抑制的毒副作用较对照组发生率显著升高高(P=0.019),两组其他毒副反应的发生情况比较差异无统计学意义(P0.05)。单药组和对照组中位生存时间(MST)分别为10个月(95%CI:1.242-18.758)、18个月(95%CI:8.261-27.739),中位无进展生存期(PFS)分别为7个月(95%CI:2.210-13.797)、13个月(95%CI:4.368-21.632),两组MST、PFS比较差异均无统计学意义(P0.277)。结论:聚乙二醇脂质体阿霉素单体或聚乙二醇脂质体阿霉素联合洛铂治疗复发性卵巢癌的疗效相当,而聚乙二醇脂质体阿霉素单体的安全性更高。  相似文献

Research on drunken horse grass (Achnatherum inebrians)-endophytic fungi (Epichloë gansuensis, E. inebrians) represented an important area of endophyte research in China. Compared with tall fescue (Festuca arundinacea)-endophyte (E. coenophiala) in the United States and perennial ryegrass (Lolium perenne)-endophyte (E. festucae var. lolii) in New Zealand, A. inebrians-endophyte research in China is becoming a leading branch of endophyte research. The studies of A. inebrians-endophyte symbionts over the past three decades were reviewed, mainly including endophyte distribution, detection methods, infection rate, fungal diversity, stress tolerances of drought, cold, salt, heavy metal, pest, disease and their mechanisms, secondary metabolite of alkaloids, feeding experiments and its ecological functions etc. Research reveals that drunken horse grass itself has no toxicity, the grazed animals will be intoxicated if the grass bears infection by Epichloë endophytes and produces ergot alkaloids. Future research will focus on E. gansuensis and E. inebrians genomics and endophyte functions. In other research we will create new grass cultivars that are 1) sanitized of the endophyte and non-toxic, 2) utilizing novel endophytes that deter birds feeding and 3) utilizing endophytes that are modified to produce novel fungicides, pesticides or medicines. © 2018 Editorial Office of Chinese Journal of Plant Ecology. Colletotrichum lindemuthianum, C. truncatum andC. graminicola, produced metabolites in culture which induced necrotic lesions on susceptible hosts. The highest production was obtained from Richard's medium under shake incubation. The toxic metabolites of these pathogens fluoresce at 254 nm and 366 nm under ultraviolet light. The crude metabolites of theseColletotrichum species inhibited seed germination at the concentration of 100 µg/ml while the potency of the metabolites decreases with increase in dilution to 0.1 µg/ml. Similarly, the metabolites also inhibited growth of seedlings of hosts to the pathogen at 100 µg/ml and the potency again reduced with increasing dilution to 0.10 µg/ml.  相似文献

A study was carried out on 20 water bodies of the same origin in southern Poland. The study objectives included the assessment of toxic metal contamination in the bottom sediments of the water bodies in comparison with the geochemical background and sediments found in the substrate (i.e., vicinity) of the water bodies (i.e., the formations present in the surroundings of the water body itself), thus demonstrating the scale of anthropogenic enrichment of bottom sediments with toxic metals and assessing the cumulative impact on water bodies. The following amounts of toxic metals were found in the bottom sediments of the water bodies examined: 181.7–35200.0 ppm for zinc, 33.3–1648.8 ppm for lead, 1.8–359 ppm for cadmium, 14.0–271.5 ppm for copper, 45.3–167.5 ppm for chromium, and 12–128.5 ppm for nickel. Ratios of the values measured to the geochemical background were as follows: 0.7–135.9 (Zn), 0.6–53.0 (Pb), 0.7–143.6 (Cd), 0.9–18.1 (Cu), 5.0–18.6 (Cr), 1.1–11.7 (Ni).  相似文献

Commercially available food supplements based on microalgae such as Spirulina (Cyanobacteria) or Chlorella (Chlorophyta) are becoming increasingly popular. Both are considered as non-toxic per se but the quality and safety of the final product depends on culturing and manufacturing conditions. This study presents two cases of human poisoning following the simultaneous use of Spirulina and Chlorella food products and a multidisciplinary approach to their evaluation: cytotoxic tests using human whole-blood in vitro and a suite of analytical screenings of over 30 elements, arsenic species and cyanotoxins: cylindrospermopsin (CYN), anatoxin-a (ANA) and three microcystin (MC) analogues. To compare metal content the Food Supplement Metal Index and Toxic Elements Contamination Index were also introduced. In all performed analyses two other commercial products were also investigated. Reported clinical symptoms of poisoning included the spreading of atopic dermatitis, nausea, dizziness, headache and fatigue. Extracts of supplements obtained from affected subjects were found to act pro-necrotically in human neutrophils, while tablets contained higher levels of several metals including Cd, Pb and Hg. All analyzed food supplements contained a significant content of Al. Neither CYN, ANA nor MC were present in any examined product. The quality of both Spirulina-based and Chlorella-based food supplements was very doubtful. The contamination problem of some commercially available microalgae-based supplements appears to be pleiotropic. The present study clearly indicates that such products should be subject to strict and routine monitoring before being registered and distributed as some of them may pose a distinct threat to human health.  相似文献
