 The root-knot nematode resistance gene Mi-1 in tomato has long been thought to be located in the pericentromeric heterochromatin region of the long arm of chromosome 6 because of its very tight genetic linkage (approx. 1 cM) to the markers Aps-1 (Acid phosphatase 1) and yv (yellow virescent). Using Mi-BAC clones and an Aps-1 YAC clone in fluorescence in situ hybridisation (FISH) to pachytene chromosomes we now provide direct physical evidence showing that Mi-1 is located at the border of the euchromatin and heterochromatin regions in the short arm (6S) and Aps-1 in the pericentromeric heterochromatin of the long arm (6L) close to the euchromatin. Taking into account both the estimated DNA content of hetero- and euchromatin regions and the compactness of the tomato chromosomes at pachytene (2 Mb/μm), our data suggest that Mi-1 and Aps-1 are at least 40 Mb apart, a base pair-to-centiMorgan relationship that is more than 50-fold higher than the average value of 750 kb/cM of the tomato genome. An integrated cytogenetic-molecular map of chromosome 6 is presented that provides a framework for physical mapping. Received: 24 July 1998 / Accepted: 14 August 1998  相似文献

Fruit sink strength or its ability to attract assimilates depends bothon sink activity and size. This study investigated one main component of sinksize, that is the number of fruit cells during tomato plant development. Plantswere grown in a controlled climate chamber under a limiting (LS, six fruits pertruss) and non-limiting (NLS, two fruits per truss and CO2enrichment) supply of carbon assimilates. Under NLS conditions, fruit cellnumber was homogeneous among successive trusses, and fruits contained onaverage1.2×106 more cells than under LS conditions,though differences were not significant on the first truss which underwent thelowest competition. Under LS conditions, an ontogenetic increase in cell numberwas observed in proximal fruits of the upper trusses attributed to theenlargement of the apical meristem during plant development. The decrease ofcell number from proximal to distal fruits within a truss, that was expectedfrom the literature, was generally observed in the LS experiment, with anaverage significant difference of about1.6×106cells between the first and fifth fruits. Nevertheless, whereas the gradient incell number from proximal to distal fruits was steep in the upper trusses, itwas not significant on the lower trusses indicating that this gradient largelydepended on the level of competition during floral development. Thus, under lowassimilate supply, cell division is a main limiting factor for fruit growth,although cell enlargement during further fruit development is also affected,butwas not measured in this work.  相似文献

 Arbuscular mycorrhizal fungi (AMF) and Erysiphe graminis are obligate biotrophic fungi with different outcomes in their interaction with plants, different targeted host tissues, but similar patterns of development and infection processes. These similarities raise the question of whether the two types of biotrophic fungal infections have common features in their regulation. To investigate this question, we compared a number of Ror and Rar barley mutants susceptible to E.graminis f. sp. hordei, as well as their resistant progenitors, for susceptibility to infection by the AMF Glomus mosseae. The two powdery mildew-resistant lines BC Ingrid and Sultan presented a similar reduction in G. mosseae development within roots when compared to the wildtype cultivar Ingrid, indicating a systemic effect of the altered genes in the plant. Ror and Rar mutants, in which susceptibility to powdery mildew is restored, showed increased resistance to AM fungal development in their roots when compared to their progenitors, which suggests that corresponding mutations must have affected genes which differentially modulate symbiotic and pathogenic biotrophic plant-fungus interactions. Aims Many resistance genes against fungal pathogens show costs of resistance. Genetically modified (GM) plants that differ in only one or a few resistance genes from control plants present ideal systems for measuring these costs in the absence of pathogens.Methods To assess the ecological relevance of costs of pathogen resistance, we grew individual plants of four transgenic spring wheat lines in a field trial with three pathogen levels and varied the genetic diversity of the crop.Important findings We found that two lines with a Pm3b transgene were more resistant to powdery mildew than their sister lines of the variety Bobwhite, whereas lines with chitinase (A9) or chitinase and glucanase (A13) transgenes were not more resistant than their mother variety Frisal. Nevertheless, in the absence of the pathogen, both the GM lines of Bobwhite as well as those of Frisal performed significantly worse than their controls, i.e. Pm3b #1 and Pm3b #2 had 39% or 53% and A9 and A13 had 14% or 23% lower yields. In the presence of the pathogen, all GM lines except Pm3b #2 could increase their yields and other fitness-related traits, reaching the performance levels of the control lines. Line Pm3b #2 seemed to have lost its phenotypic plasticity and had low performance in all environments. This may have been caused by very high transgene expression. No synergistic effects of mixing different GM lines with each other were detected. This might have been due to high transgene expression or the similarity between the lines regarding their resistance genes. We conclude that costs of resistance can be high for transgenic plants with constitutive transgene expression and that this can occur even in cases where the non-transgenic control lines are already relatively resistant, such as in our variety Frisal. Transgenic plants could only compete with conventional varieties in environments with high pathogen pressure. Furthermore, the large variability among the GM lines, which may be due to unpredictable transgene expression, suggests that case-by-case assessments are necessary to evaluate costs of resistance.  相似文献

The aim of this study was to investigate the role of concanavalin A (Con A)-cellulose-bound tomato peroxidase for the decolorization of direct dyes. Cellulose was used as an inexpensive material for the preparation of bioaffinity support. Con A-cellulose-bound tomato peroxidase exhibited higher efficiency in terms of dye decolorization as compared to soluble enzyme under various experimental conditions. Both Direct Red 23 and Direct Blue 80 dyes were recalcitrant to the action of enzyme without a redox mediator. Six compounds were investigated for redox-mediating property. Immobilized peroxidase decolorized both dyes to different extent in the presence of all the used redox mediators. However, 1-hydroxybenzotriazole emerged as a potential redox mediator for tomato peroxidase catalyzed decolorization of direct dyes. These dyes were maximally decolorized at pH 6.0 and 40 degrees C by soluble and immobilized peroxidase. The absorption spectra of the untreated and treated dyes exhibited a marked difference in the absorption at various wavelengths. Immobilized tomato peroxidase showed a lower Michaelis constant than the free enzyme for both dyes. Soluble and immobilized tomato peroxidase exhibited significantly higher affinity for Direct Red 23 compared to Direct Blue 80.  相似文献

以小型番茄 Micro-Tom 为材料,利用农杆菌介导法导入花青素调节基因VlmybA2。对抗性筛选出的再生植株进行 GUS 组织染色和 PCR 检测,证明外源基因已经整合到 Micro-Tom 中,转基因番茄根、茎、叶脉、果皮均呈紫色,花色为黄紫嵌合。而野生型的根为白色,茎、叶脉呈绿色,果皮为红色,花为黄色。对转基因番茄的花青素含量、叶片叶绿素含量和光合速率等生理指标进行测定,花青素含量有显著增加,叶绿素含量降低,VlmybA2基因过量表达会降低植株的光合效率,但对植株正常生长影响并不显著。VlmybA2 基因既可增加抗衰老物质花青素含量,又可作为转基因植株的报告基因。  相似文献

Antipathogenic potential of 16 plants was evaluated in the form of aqueous extracts against Peronosclerospora sorghi, causing downy mildew of sorghum. A novel method was developed in which conidial suspension and plant extracts were mixed individually and allowed to stand for 5?min and then used to inoculate the host by sprout-dip method. The sprouts thus inoculated were grown in pots, and disease incidence was observed. Three extracts (Parthenium hysterophorus, Duranta repens and Oxalis latifolia) suppressed the disease at par with chemical fungicide (Mancozeb 75%). In form of spray, D. repens extract with tween-20 adjuvant (1.9% disease) performed closest behind mancozeb (0% disease), while negative control showed 21% disease incidence. Organic management of airborne inoculum of downy mildew of sorghum is feasible and preferable as compared to the use of chemical fungicides, considering human and environmental health concerns. Use of water extract keeps the technology simple so that it can be directly prepared and used by farmers. Short listing of three most effective water extracts would help in self-reliance of farmers, reducing their dependence on commercial products.  相似文献

A frosty mildew was observed on leaves of Salix koreensis in two localities of Korea during 2011 and 2012. The main signs and symptoms were expressed as conical white to cream coloured tufts of the causal fungus on the brown lesions, followed by premature defoliation. Based on morphological observations, cultural characteristics and phylogenetic analyses of rDNA‐ITS, the fungus was identified as Mycopappus alni, which has been known to be associated with frosty mildews on Alnus spp., Betula spp., Crataegus chlorosarca and Pyrus pyrifolia. Pathogenicity test was conducted twice with the same results, fulfilling Koch's postulates. This is the first case of SalixMycopappus association as well as the first report of frosty mildew on S. koreensis.  相似文献
