Antibiotic resistance genes can act as either cell autonomous or non-cell autonomous genetic markers with which to monitor the excision of plant transposons. To convert spectinomycin resistance from a noncell autonomous resistance to cell autonomous resistance, a transit peptide for chloroplast localization from a petunia ribulose bisphosphate carboxylase (rbcS) gene was fused in-frame to the aadA gene, which confers spectinomycin and streptomycin resistance. Constructs were generated in which the expression of this chimeric gene was prevented by the presence, in the 5 untranslated leader, of the maize transposons Activator (Ac) or Dissociation (Ds). When progeny of tobacco or tomato plants transformed with these constructs were germinated on spectinomycin-containing medium, germinally revertant and somatically variegated individuals could be distinguished.  相似文献

Genetic chimeras of the VFNT cherry tomato line (Lycopersicon esculentum) were produced by mutagenizing seeds with ethyl methane sulfonate (EMS). The chimeras thereby produced were evaluated by progeny-testing the fruits of the genetically mosaic tissue. A total of 2011 M1 plants was grown from treated seeds and evaluated by screening their 95175 (M2) progeny for mutations affecting seedling phenotype. Three vigorous and fertile M1 plants bearing mutant progeny with definitive phenotypes were selected for systematic harvesting and analysis. The specific location of each fruit was noted at harvest time, enabling the mutated sporogenous tissue of the mosaic M1 plants to be traced. Sectoring appeared in both branch and floral tissues. In several cases, mutant progenies were restricted to individual branches or parts thereof. True-breeding recessive mutants whose monogenic mode of inheritance was later established occasionally segregated within M1 fruit progenies at frequencies that indicate a non-homogeneous floral meristem origin. The data emphasize the necessity of making a well-distributed harvest of mosaic plants in order to detect as many variants as possible, as mosaic sectors may or may not recur late in ontogeny and may not contribute to sporogenous tissue early in development.  相似文献

A new allele, SC148, of thesulfurea locus inLycopersicon esculentum was detected in a line derived after repeated selfing of plants that had been regenerated from tissue culture. Like the originalsulf mutant, SC148 displayed two mutant phenotypes: green-yellow speckled plants in which thesulf vag allele is present and pure yellow plants homozygous for thesulf tpura allele. Although the mutant alleles are recessive to wild-type, an unpredictable number of variegated and pura plants appeared in F1 progenies that had been derived from crosses between SC148 and wild-type tomato plants. The presence of the wild-typesulf + allele in these variegated heterozygotes was demonstrated using a cytological marker that is linked tosulf. It is concluded that the mutantsulf allele of SC148, imposes its variegated expression state on the wild-typesulf + allele present insulf +/sulfvag heterozygotes. This behaviour, known as paramutation, has also been described for the originalsulf allele. The SC148 allele, however, seems to induce changes at an earlier stage in development. The analogy of this paramutagenic system to dominant position effect variegation inDrosophila is discussed.  相似文献

Seven fungi associated with fruit rot of tomato were isolated includingFusarium equiseti, F. chlamydosporum, Alternaria solani, Geotrichum candidum, Acremonium recifei, Aspergillus flavus andA. niger. They were all pathogenic on tomato fruits, most pathogenic beingGeotrichum candidum followed byA. niger. Least rot was caused byAlternaria solani. The optimum temperature for maximum rotting caused byG. candidum, A. niger andA. flavus was 30°C. The relative humidity for maximum rot ranged from 70–90%. Tomato fruits stored well at 0–10°C and rather poorly at 20–30°C. Fruits stored at 35°C showed blemishes. The best RH for storage ranged between 60 and 90%.  相似文献

The Ventilago Gaertn. (Rhamnaceae) is widely distributed in pantropical areas of Africa, Asia, and Australia. However, fossil records of this taxon are sparse, which limits understanding of the evolution and biogeographic history of the genus. In the present study, we report and describe two new fossil species of Ventilago, V. siwalika sp. nov. from the Miocene sediments of Himachal Pradesh, western Himalaya, and V. pliocenica sp. nov. from the Pliocene sediments of Jharkhand, eastern India based on single-winged samaras. Ventilago pliocenica is characterized by a prominent midvein, obtuse to sub-round apex with mucronate tip, longitudinal secondary veins extending the full length of the fruit, and reticulate nature of higher-order veins, the presence of equatorial rim, the hypanthium, and short pedicel. On the other hand, V. siwalika is characterized by a prominent midvein, obtuse to sub-round apex with mucronate tip, longitudinal secondary veins extending the full length of the fruit, and reticulate nature of higher-order veins. Our discovery represents the first unambiguous fossil record of single-winged samara of Ventilago from India and provides valuable insights into the evolution of this genus. In this paper, we also review its biogeographic history and add new information to understand its hypothetical migration route. Present and earlier records of Ventilago also suggest that this genus was a common forest element during Neogene (Miocene time) in Asia.  相似文献

本文报道苦楝果实中的生物活性物质。发现果实的抽提物和单体化合物不仅具有防治作物害虫的作用,而且,还对植物有生长调节作用。通过生物活性鉴定结果表明:果实种仁的水抽提物对水稻早期萌动、发芽与秧苗的生长均有促进作用。其中,处理稻种第2天(48h),其发芽率比对照高出24.67%~42.86%;第4天秧苗的根长在1cm以上占56.5%~65.0%,而对照组只有15.5%。尤其是抽提液浓度在6000ppm时处理的稻种,测得生长10d的秧苗总长为对照的120.27%,胚根长为对照的121.54%;以及叶片中的叶绿素总含量比对照高8.5%。还证明了从果实中提取分离的几种三萜类化合物和在果实中发现的一种新化合物对苯二甲酸二甲酯,均对菜青虫有拒食和胃毒活性。  相似文献
