Two tRNA sequences from Methanobacterium thermoautotrophium are reported. Both tRNAGlyGCC and tRNANUUAsn, the first tRNA sequences from methanogens, were determined by partial hydrolyses (both chemical and enzymatic) and analyzed by gel electrophoresis. The two tRNAs contain the unusual T-loop modifications, Cm and m1I, which are present in other archaebacterial tRNAs. Finally the presence of an unknown modification in the D-loop has been inferred by a large jump in the sequence ladder. Bovine serum albumin conjugates of guanosine prepared by the periodate method was used as immunogen to elicit guanosine antibodies in rabbits. The specificities of the antibodies were studied by the inhibition of their binding to [3H]Gox-red, [32P]DNA and [3H]RNA by related non-radioactive compounds. A population of antibodies is specific to Gox-red with an average association constant of around 10(7) M-1 at 4 degrees C. There are a population of antibodies which bind to [32P]ssDNA and [3H]RNA specifically at guanosine residues. RNA binding antibodies were separated into two populations by affinity chromatography. 相似文献

The low-Mr proteins (tentatively called protein I and II) were purified from 2 M NaCl extracts of the Bacillus stearothermophilus ribosome. Their amino acid sequences have been determined from the peptides obtained by digestion with trypsin, chymotrypsin, and pepsin, and by cleavage with CNBr, using the micro-DABITC/PITC double-coupling method [FEBS Lett. (1978) 93, 205-214]. Protein I contains 56 residues and has an Mr of 6514. Protein II had 37 residues with an Mr of 4361. The amino acid sequence of protein I shows significant similarity to L32 from E. coli, whereas that of protein II is slightly, if at all, related to ribosomal protein L34 from E. coli. 相似文献

Summary Studies were made on the genetic determination of androgenetic plant yield and its two components: embryo production and green plant regeneration. This involved the analysis of a complete 7×7 diallel cross of 4 androgenetic lines and 3 lines obtained by pedigree selection, one of them having the Triticum timopheevi cytoplasm. The three traits analysed are both heritable and environmentally influenced (by season and culture medium composition). The analysis of embryo production shows a mainly nuclear inheritance, with predominantly additive gene action, but also a favourable effect of Triticum timopheevi cytoplasm. Green plant regeneration has a more complex genetic determination, with additive as well as non-additive gene action and cytoplasmic influences. Hybrids appear superior to inbred lines for embryogenesis and green plant yield, but not for green plant regeneration. Androgenetic lines used as parents did not show superiority over other parents either in their own value or in the transmission of androgenetic abilities. Genetic improvement seems to be possible by recombination in progenies of hybrids between lines having complementary abilities. 相似文献

Summary Twenty characters were measured on 60 tomato varieties cultivated in the open-air and in polyethylene plastic-house. Data were analyzed by means of principal components, factorial discriminant methods, Mahalanobis D2 distances and principal coordinate techniques. Factorial discriminant and Mahalanobis D2 distances methods, both of which require collecting data plant by plant, lead to similar conclusions as the principal components method that only requires taking data by plots. Characters that make up the principal components in both environments studied are the same, although the relative importance of each one of them varies within the principal components. By combining information supplied by multivariate analysis with the inheritance mode of characters, crossings among cultivars can be experimented with that will produce heterotic hybrids showing characters within previously established limits. 相似文献

Adult and juvenile common marmosets were introduced to unfamiliar conspecifics individually and in whole groups. In introductions
using animals of the same sex, adults were mutually hostile unless they were related or socially familiar; juveniles behaved
submissively to adults and little aggression was observed between juveniles. In introductions using animals of the opposite
sex, males of all ages solicited females but females did not reciprocate. When whole groups were introduced, in two experiments
most of the elder group members behaved aggressively and younger animals did not interact frequently; but in a third experiment,
in which the adult males were related, little aggression was observed and younger animals behaved amicably. It is argued that
the behavioral reactions shown by individual marmosets are related to territorial hostility and that the behavior of juveniles

Summary Sequences from Petunia hybrida chloroplast DNA which have the property to promote autonomous replication in Saccharomyces cerevisiae were cloned in vector YIp5. Seven cloned chloroplast DNA fragments are localized at one of two different sites on the chloroplast genome. One site, arsA was mapped on a 1.8 Kb fragment at position 27.0–28.8 Kb on the P. hybrida chloroplast genome. The plasmids containing this arsA are stable both in yeast and E. coli. The other site, arsB, was shown to be very unstable and is located either in the small single copy region close to the inverted repeat or just in the inverted repeat. The functioning of these sequences as a possible origin of replication in vivo is discussed. 相似文献
