Fluorescence of the marine alga Thalassiosira weissflogii (Grunow) Fryxell et Hasle with open (F o ) and closed (F m ) reaction centers of photosystem 2 (PS 2) and its relative variable fluorescence (F v/F m ) were measured at various levels of inorganic nitrogen. A significant heterogeneity of the population in terms of these parameters was revealed. Some cells within the population were more sensitive to nitrogen deficiency, and their photosynthetic apparatus was disrupted to a greater extent. The cells within a population also differed in terms of their ability to recover after incubation at low nitrogen levels. Enhancement of nitrogen deficiency resulted in an increase in the variability of the F o and F v/F m values of the cells. Fluorescence variability decreased at a less pronounced deficiency. Fluorescence variability should be taken into consideration in the studies concerning responses of algae to changes in nutrient contents.  相似文献

Concentrated stock solutions of sodium metasilicate (Na2SiO3.9H2O), prepared in deionized-distilled water and acidified to pH 2 as suggested by McLachlan (1973), were found to be polymerized; stocks kept at their alkaline pH of dissolution were not polymerized. Depolymerization studies using the acidified stock added to deionized-distilled water, 3% NaCl or artificial saltwater medium showed that complete depolymerization took longer than 2 days. Studies of Si uptake, using the marine diatom Thalassiosira pseudonana (Hust.) Hasle and Heimdal, showed that use of an acidified Si stock for experiments can lead to anomolous results. It is recommended that concentrated Si stocks be stored at their pH of dissolution and not be acidified prior to addition to media. In instances where the pH must be adjusted prior to addition (e.g. in some Si-uptake experiments), it may be necessary to take into account polymerization / depolymerization effects.  相似文献

The morphology of the valve of Coscinodiscus wailesii and the development of its siliceous architecture, studied in the SEM and TEM, is compared with valve formation in Thalassiosira eccentrica (Ehrenberg.) Cleve. Though the areolae-architecture of these two species differs in such that the cribrum is proximal and the foramen distal in T. eccentrica, and in C. wailesii the cribrum is distal and the foramen proximal, the formation of their complex loculate system is similar, displacing a centrifugal growth pattern with respect to the valve, and a proximal to distal, sequentially. During base layer formation a hitherto undescribed rib system outlines the prospective areolae. The vertical differentiation is in principle the same as in T. eccentrica and also the cribra are formed centripetally in relation to the areolae in both species. The location of the cribra at the proximal or distal side, therefore, seems to be of minor importance for the sequence of silica deposition. Variation in girdle bands is discussed with respect to cell division. The prophasic nuclear migration from the interphase position to the girdle bands, where mitosis is performed, seems to be necessary for triggering the formation of the unilateral cleavage furrow that later forms a cleavage ring with excentric position. The divided nuclei migrate with the ingrowing cleavage furrow to the center of the newly created protoplasmic surfaces to initiate valve formation.  相似文献

The feasibility of estimating phytoplankton carbon and RNA concentrations from measurements of ATP and chlorophyll a (chl a) concentrations was studied using chemostat populations of the marine diatom Thalassiosira weissflogii (Grunow) Fryxell & Hasle (= T. fluviatilis Hustedt). C:ATP and RNA:ATP ratios were studied for six additional marine species in batch culture representing five classes of phytoplankton. Statistical analyses revealed that both the growth rate and the factor limiting growth (NO3-, NH4+, PO43- or light) could alter C:ATP, RNA: ATP, C:chl a and RNA:chl a ratios by amounts which were large compared to measurement error. An analysis of variance of the batch culture results indicated that both species and the source of inorganic nitrogen (NO3-, or NH4+) had a significant effect on C:ATP and RNA:ATP ratios. Light had less of an influence on C:ATP and RNA:ATP ratios than on C:chl a and RNA:chl a ratios, and for this reason we feel that phytoplankton C and RNA concentrations can be estimated with greater reliability from ATP than from chl a measurements. The range of C:ATP and RNA:ATP values found for T. weissflogii under a variety of growth conditions was similar to that for the six additional species grown in batch culture, suggesting that this range of values is indicative of the extremes likely to occur in living cells. Our results and additional data in the literature indicate that phytoplankton C and RNA concentrations can be estimated to within a factor of two by multiplying ATP concentrations by 311 and 35, respectively, in N limited systems, and by 341 and 36, respectively in PO43- limited systems.  相似文献

Some diatom exudates may remain attached to the exterior cell surface, potentially altering cell stickiness and affecting important aspects of the diatom's ecology such as aggregation rates and grazing rates. We measured the accumulation of cell-surface sugar-containing compounds by labeling cultured marine diatoms with fluorescent-tagged sugar-binding lectins and measuring the fluorescence associated with each cell using flow cytometry. The binding of FITC-labeled concanavalin A (FITC-ConA), a lectin that binds to glucose and mannose residues, varied 5-fold among different diatom species in exponential growth (on a per-cell basis) and 2–3-fold within a given species in different physiological states. Although transparent exopolymer particles followed a simple accumulation curve over time in batch culture, FITC-ConA. cell-1 did not follow the same pattern, suggesting that surface sugar accumulation is not driven simply by the accumulation of such particles in the medium. For Thalassiosira pseudonana (Hustedt) Hasle and Heimdal (3H clone), the amount of sugar-containing compounds on the cell surface increased transiently as growth rate slowed in early stationary phase under both N and Si limitation. For Chaetoceros neogracile (Schuett) van Landingham, FITC-ConA. cell-1had a strong inverse relationship with growth rate across several Si-limited batch culture experiments. Both results suggest some biological mediation of cell-surface sugar-containing compounds. Our study reveals the great potential for using lectin binding to investigate cell-surface sugars on diatoms. Lectins allow us to investigate noninvasively the role of cell-surface sugar-containing compounds in modifying cell stickiness and aggregation, as well as the partitioning of exuded phytoplankton carbon. We suggest that cell-surface sugar accumulation may be related to diatom stickiness, based on a correlation between our FITC-ConA measurements and stickiness estimates in the literature on several of the species we studied.  相似文献

Proliferating cell nuclear antigen (PCNA) is an auxiliary protein for polymerase-δ and therefore is essential for cellular DNA synthesis. The synthesis and abundance of PCNA in the cell are cell-cycle-dependent, both increasing markedly during the S phase. Such a protein could be a useful cell cycle marker, which is required for estimating algal species-specific growth rates via the cell cycle approach. By using commercially available monoclonal anti-rat-PCNA antibody and an enhanced chemiluminescence technique, PCNA-like proteins were detected in four species of marine phytoplankton. The strong single band detected on western blots of Isochrysis galbana Parke, Thalassiosira weissflogii Cleve, and Dunaliella tertiolecta Butcher had an apparent molecular weight of 33–36 kDa. This molecular weight is within the range as observed for PCNA in a wide phylogenetic array of organisms (33–36 kDa). In the diatom Skeletonema costatum (Grev.) Cleve, the PCNA antibody detected a major band of about 19 kDa as well as a minor band of 38 kDa. The detected proteins were specifically recognized by the monoclonal anti-rat-PCNA antibody. The PCNA-like proteins in I. galbana, T. weissflogii, and D. tertiolecta were more abundant in the exponential growth stage and then decreased and became undetectable in the late stationary stage. Our results show that the detected antigens appear to be algal analogs of PCNA.  相似文献

Experiments on a marine diatom, Thalassiosira pseudonana (Hustedt) clone 3H, demonstrate that under moderate photon flux densities (75 μmol quanta·m?2·s?1) of visible light the inhibition of photosynthesis by supplemental ultraviolet (UV) radiation (UV-B: 280–320 nm) is well described as a hyperbolic function of UV-B irradiance for time scales of 0.5–4 h. Results are consistent with predictions of a recently developed model of photosynthesis under the influence of UV and visible irradiance. Although net destruction of chlorophyll occurs during a 4-h exposure to UV-B, and the effect is a function of exposure, the principal effect of UV-B is a decrease in chlorophyll-specific photosynthetic rate. The dependence of photoinhibition on dosage rate, rather than cumulative dose, and the hyperbolic shape of the relationship are consistent with net photoinhibition being an equilibrium between damage and repair. The ratio of damage to repair is estimated by a mathematical analysis of the inhibition of photosynthesis during exposures to UV-B. A nitrate-limited culture was much more sensitive to UV-B than were the nutrient-replete cultures, but the kinetics of photoinhibition were similar. The analysis suggests that the nutrient-limited culture was more sensitive than the nutrient-replete cultures because repair or turnover of critical proteins associated with photosynthesis is inhibited. An inhibitor of chloroplast protein synthesis was used to suppress repair processes. Photoinhibition by UV-B was enhanced, and inhibition was a function of cumulative dose, as would be expected if damage were not countered by repair. The fundamental importance of repair processes should be considered in the design of field experiments and models of UV-B effects in the environment, especially in the context of vertical mixing. Repair processes must also be considered whenever biological weighting functions are developed.  相似文献
