The influence of four non-ionic surfactants (isododecyl and cetyl polyoxyethylene ethers) on aqueous polysaccharide solutions (sodium alginate, guar gum, and sodium carboxymethyl guar), applicable for textile printing pastes, were studied via rheological measurements.

Rheology of polysaccharide–surfactant solutions in aqueous matrices is primarily governed by polymer content, which imparts marked shear-thinning and viscoelastic character to the system. Such properties are modulated in moderate but sensible way by changes in surfactant concentration or type. Above 3% surfactants addition to non-substituted guar gum solutions results in a significant impact leading to phase separation and a particular strongly associated phase is formed due to hydrogen bonds between ethylene oxy units from the surfactant and primary hydroxyl groups in guar.

A satisfactory fitting of viscosity data is obtained with both the Cross equation and the Roberts–Barnes–Carew model. The experimental results of mechanical spectra can be described quite satisfactory with both the Friedrich–Braun and the generalized Maxwell models.  相似文献

The effluent of a pharmaceutical company was examined microbiologically. Its bacterial count was 2.15 × 105 c.f.u./ml and there was evidence of faecal contamination with MPN of > 1800. The organisms encountered included Staphylococcus aureus, Escherichia coli, Proteus vulgaris, Serratia marcescens and Pseudomonas aeruginosa. The resistances of the 25 bacterial strains isolated from the effluent to the commonly used antibiotics were studied. About 80% of the isolates were resistant to Amoxycillin, 76% to Nitrofurantoin, 64% to Cotrimoxazole and Augmentin, 60% were resistant to Nalidixic acid, 52% were resistant to Tetracycline and Ofloxacin, while resistance of 12% was obtained for Gentamicin. Among the eight antibiotics tested, seven patterns of drug resistance were obtained and all of them were multiple-drug resistance with the number of antibiotics ranging from 2–8. All the strains of E. coli and S. aureus had high MIC values for Cloxacillin and Amoxycillin. In all, 13 strains of the bacterial isolates had evidence for the production of -lactamases. The potential of the effluent in spreading drug resistance and the public health implications are discussed.  相似文献

Caffeine (1,3,7-trimethylxanthine), a ubiquitous component of human diet has been suggested as a chemical indicator of ecosystem impacts of sewage spills and treated effluent discharges because it is not sufficiently metabolized by wastewater microorganisms. This study identified enzymes responsible for caffeine metabolism in sewage bacteria. Pseudomonas putida biotype A (ATCC 700097) originally isolated as a rare caffeine-degrading organism in domestic wastewater exhibited diauxic growth on caffeine, concomitant with the expression of a P450-type cytochrome and peroxidase enzyme activities. Initial growth phase lasted 13.8 ± 1.4 h with a growth rate that was five times slower than the secondary growth phase that lasted 5.5 ± 1.2 h. Molecular and enzymatic characteristics of the cytochrome P450-type enzyme differ from the previously described cytochrome P450 (P450cam) of P. putida (ATCC 17453) involved in camphor metabolism. The caffeine-inducible cytochrome P450-type enzyme exhibited a carbon monoxide difference spectrum peak at 450 nm, but does not allow growth on camphor. Caffeine induced production of haem-associated peroxidase activity was confirmed with 3,3, 5,5-tetramethylbenzidine–H2O2 reaction in polyacrylamide gels. Polymerase chain reaction (PCR) primers derived from the gene for cytochrome P450cam (camC) of P. putida (ATCC 17453) did not yield an amplification product when DNA extracted from P. putida strain ATCC 700097 was used as template. The data demonstrate that caffeine is metabolized through a specific biphasic pathway driven by oxygen-demanding enzymes.  相似文献

Selected isolates of fungi were grown on wheat straw and corncob in the presence of different moistening agents such as water, molasses, potato dextrose broth and distillery effluent. All the fungal isolates responded differently with respect to growth and ligninolytic enzyme production. Fungal growth on different substrates was checked by calculating ergosterol content, which varied widely within a single species when grown on different substrates. The maximum laccase production was obtained for Aspergillus flavus TERI DB9 grown on wheat straw with molasses. For manganese peroxidase, highest production was in Aspergillus niger TERI DB20 grown on corncob with effluent. Among the two isolates positive for lignin peroxidase, the highest production was in Fusarium verticillioides ITCC 6140. This immobilized fungal biomass was then used for decolorization of effluent from a cane molasses based distillery. Maximum decolorization (86.33%) was achieved in Pleurotus ostreatus (Florida) Eger EM 1303 immobilized on corncob with molasses in a period of 28 days.  相似文献

Anaerobic pretreated paper process water was characterized interms of readily biodegradable, slowly biodegradable, very slowly biodegradable and inert wastewaterfractions under mesophilic and thermophilic conditions. The anaerobic pretreated paper process water containeda relatively high amount of slowly biodegradable components and few easily biodegradable componentsas indicated by the ratio of short term BOD over the BOD5. Wastewater readily biodegradable COD, determinedas short term BOD, was almost similar when measured under both temperature conditions. Fractions ofslowly biodegradable COD and inert COD of the same wastewater were found to depend on the type of biomassinvolved in the test. Thermophilic aerobic biomass was not able to degrade the wastewater to the sameextent as the mesophilic biomass resulting in higher apparent inert COD levels. Furthermore, wastewater colloidalCOD did not flocculate under thermophilic conditions and was thus not removed from the liquid phase.  相似文献

In a screening of saprotrophic, ectomycorrhizal and plant pathogen ascomycetes a frequent occurrence of laccase was observed. Lamprospora wrightii, the best producing organism, was chosen to elucidate the properties of a laccase from a moss-associated, saprotrophic ascomycete. The expression of laccase by this bryophilic fungus could be increased by the addition of tomato juice or copper sulfate to the medium. The obtained volumetric activity after optimization was 420 U/mL in either shaking flask or bioreactor-based cultivations. The purified laccase has a molecular mass of 68 kDa and an isoelectric point of 3.4. Although of ascomycete origin, its catalytic properties are similar to typical basidiomycte laccases, and an excellent activity and stability was observed at low pH, which makes it suitable for bioremediation in acidic environments. As an example, the decolorization reactions of azo-, anthraquinone-, trimethylmethane- and indigoid dyes at pH 3.0 and 5.0 were investigated. All ten selected dyes were decolorized, five of them very efficiently. Depending on the dye, the decolorization was found to be a combination of two reactions, degradation of the chromophore and formation of polymerized products, which contributed to the overall process in a dye-specific pattern.  相似文献

A pool of hydrolases with 21.4 U g−1 lipase activity was produced through solid-state fermentation of the fungus Penicillium restrictum in waste from the Orbignya oleifera (babassu) oil processing industry. Enzymatic hydrolysis and anaerobic biodegradability tests were conducted on poultry slaughterhouse effluents with varying oil and grease contents (150–1200 mg l−1) and solid enzymatic pool concentrations (0.1–1.0% w/v). Enhanced anaerobic treatment efficiency relative to raw effluent was achieved when a 0.1% concentration of enzymatic pool was used in the pre-hydrolysis stage with 1200 mg oil and grease l−1 (chemical oxygen demand (COD) removal efficiency of 85% vs. 53% and biogas production of 175 ml vs. 37 ml after 4 d).  相似文献

Ischnoderma resinosum produced extracellular ligninolytic enzymes laccase and MnP. The activity of laccase achieved the maximum on day 10 (29.4 U L−1), the MnP on day 14 (34.5 U L−1). Laccase and Mn-peroxidase were purified from the culture liquid using gel permeation and ion-exchange chromatographies. Purified Mn-peroxidase performed decolorization of all textile dyes tested (Reactive Black 5, Reactive Blue 19, Reactive Red 22 and Reactive Yellow 15). Laccase was inactive with Reactive Black 5 and Reactive Red 22, while all dyes were decolorized after addition of the redox mediators violuric acid (VA) and hydroxybenzotriazole (HBT). The culture liquid from I. resinosum cultures was also able to decolorize all dyes as well as the synthetic dyebaths in the presence of VA and HBT. The highest decolorization rates were detected in acidic pH (3–4).  相似文献

The decolorization kinetics of Remazol Brilliant Violet 5R (RBV-5R) and Remazol Black B (RB-B) (mono- and diazo reactive dyes, respectively) was investigated in the first 9 h (anaerobic phase) of a 24-h cycle anaerobic/aerobic sequencing batch reactor (SBR). Two distinct, successive decolorization periods were observed for both dyes, apparently due to different decolorization mechanisms. The apparent first-order rate constants were much lower for the second periods. First-order kinetics were apparently followed for both periods of RBV-5R but not for the first decolorization period of RB-B, possibly due to the occurrence of mass transfer limitations.  相似文献

Lake Myvatn, northern Iceland is unique in that almost all the inflow is supplied through the groundwater by artesian springs. Hardly any surface water is encountered in the area which is covered by young and porous lava fields and transected by faults. The inflow of water and its chemical composition is, therefore, very stable. Geothermal and volcanic activities affect the groundwater system in the Lake Myvatn area and greatly influence the lake's chemistry and thus the biological conditions especially by providing continuous and ample sources of silica and sulphate. Groundwater studies have been intensified in the area during the last years for further developing the Námafjall geothermal field in the area and the Krafla geothermal field about 10 km from Námafjall. Groundwater chemistry was monitored regularly for 2 years at 22 sampling sites and for determining selected indicator constituents in the samples. Concurrently, several tracer tests were performed in the area, the rate of groundwater flow measured, a reservoir model of the groundwater system established. These studies enabled us to divide cold groundwater and geothermal effluent in the Lake Myvatn area into six distinct groups, based on stable isotope ratios, chemical composition and geographical position. The groundwater has separate origins in the local high ground north of Lake Myvatn and the highlands far to the south, possibly as far south as the glacier Vatnajökull. The waters are to a different extent affected by geothermal activity, and effects of volcanic activity were noted during the Krafla fires in 1975–1984. Although these have diminished, they have not completely disappeared. The effluent from Krafla seems to travel to the east of Lake Myvatn and traces of it have not been found to enter the lake. The Námafjall effluent on the other hand travels along fissures to the lake. Attempts made to simulate the evolution of the geothermal water of Krafla and by theoretically titrating local groundwater with rock at elevated temperatures and adding volcanic gas seem promising and result in a composition close to the natural one.  相似文献
