A seasonal study of two spring-fed stream systems in south-central Texas was undertaken over a 15-month period from June 1996 to September 1997. Relative abundance of the epilithic diatom flora, percent cover of macroalgae, and several physical and chemical conditions were monitored in one 20-m stretch in each of the streams at approximately two-month intervals. Six additional spring-fed stream segments were sampled in May 1996 for macroalgae and measured for the same conditions, in order to determine how representative the macroalgal floras of the study sites were of south-central and western Texas. Conditions in the two stream segments were relatively constant over the 15 months, except for maximum depth; this finding is similar those reported for other spring-fed systems worldwide. Sixty-eight diatom taxa and nine macroalgae taxa were identified from the San Marcos River, which compares to 46 diatom taxa and six macroalgae taxa from the Comal River. Several diatom and macroalgae species were significantly correlated to stream conditions (e.g. Dichotomosiphon tuberosus and Hildenbrandia angolensis with daylength, and Amphora pediculus with maximum depth). Twenty-one macroalgal taxa were identified from the six additional spring-fed steam segments. Conditions in these streams were very similar to those measured in the San Marcos River and Comal River, except that they had a higher average pH x= 8.1 versus x= 7.6). The epilithic diatom flora of the San Marcos River and the Comal River contains many of the diatom and some macroalgal taxa commonly reported from springs globally. The cell wall of the red microalgae Porphyridium sp. (UTEX 637) comprises a complex amorphous polysaccharide (6–7 × 106 Da). The polysaccharide is made up of xylose, glucose, and galactose as the main sugars, as well as some minor sugars, protein, sulfate, and glucuronic acid, the latter two conferring a negative charge on the polysaccharide. In this study, we used synchronized cultures as one of the ways of unraveling the mechanism of biosynthesis of this complex polysaccharide by following cell-wall formation during the cell cycle. Synchronization of Porphyridium sp. was achieved with an alternating light:dark regime of 12:12 h LD and dilution of the culture at the end of the cycle. Under these conditions, cell duplication occurred between the 12th and 14th hours of the cycle. The following order of building toward formation of the final polysaccharide appeared to take place: Intermediate polysaccharides with molecular masses ranging from 0.5 × 106 to 2 × 106 Da appeared in succession during hours 2–6 of the cycle, and the full-sized polysaccharide was detected by the 8th hour. At the beginning of the cycle, xylose was the predominant sugar. Sulfur peaked at hours 2–4; glucose, galactose, and glucuronic acid at hours 8–12; and the minor sugars at hours 12–14. Upon incubation of low molecular mass polymer (0.5 × 106 Da) collected from the 4th hour with cellular crude extract from cells of the 6th hour of the cycle, two intermediates were formed (0.8 × 106 Da and 2 × 106 Da). We suggest that the 0.5 × 106 Da polymer intermediate, which is composed mainly of xylose, is the first polymer secreted into the medium, where it is further polymerized enzymatically to produce the 2 × 106 Da polymer via an intermediate 0.8 × 106 Da polymer. Later, the full-size polysaccharide is produced.  相似文献

Interactions between bacteria and species of harmful and/or toxic algae are potentially important factors affecting both the population dynamics and the toxicity of these algae. Recent reports of bacteria lethal to certain harmful algal bloom (HAB) species, coupled with a rapidly evolving interest in attempting to minimize the adverse effects of HABs through various prevention, control, and mitigation strategies, have focused attention on defining the role of algicidal bacteria in bloom termination. The aim of the present study was to determine whether algicidal bacteria active against Gymnodinium breve Davis, a dinoflagellate responsible for frequent and protracted red tides in the Gulf of Mexico, are present in the waters of the west Florida shelf. To date, we have isolated two bacterial strains from this region lethal to G. breve and have begun to characterize the algicidal activity of one of these strains, 41-DBG2. This bacterium, a yellow-pigmented, gram-negative rod, was isolated from waters containing no detectable G. breve cells, suggesting that such bacteria are part of the ambient microbial community and are not restricted to areas of high G. breve abundance. Strain 41-DBG2 produced a dissolved algicidal compound(s) that was released into the growth medium, and the algicide was effective against the four Gulf of Mexico G. breve isolates tested as well as a closely related HAB species that also occurs in this region, Gymnodinium mikimotoi Miyake et Kominami ex Oda. Nonetheless, data showing that a nontoxic isolate of Gymnodinium sanguineum Hirasaka from Florida Bay was not affected indicate that the algicidal activity of this bacterium does exhibit a degree of taxonomic specificity. Our efforts are currently being directed at resolving several critical issues, including the identity of the algicide(s), the mechanisms regulating its production and ability to discriminate between target algal species, and how the growth rate of 41-DBG2 is affected by the presence of G. breve cells. We have also proposed a conceptual model for interactions between algicidal bacteria and their target species to serve as a testable framework for ensuing field studies.  相似文献

Mean d2 is a recently devised microsatellite-based measure that is hypothesised to allow the detection of inbreeding depression and heterosis in free-living populations. Two studies that have investigated the measure have both demonstrated an association between mean d2 and traits related to fitness. Here we present an association between mean d1 and an important component of fitness, first-year overwinter survival, in a population of red deer on the Isle of Rum, Scotland. The association between survival and mean d2 differed between males and females. As predicted, outbred female calves (high mean d2) survived better than those that were inbred (low mean d2). However, the association was in the opposite direction in male calves. We suggest that this difference is due to different early growth strategies between the sexes. The association between mean d2 and survival was not significantly influenced by any single locus. Decomposition of mean d2 into a recent inbreeding component and an outbreeding component showed that it was the degree of outbreeding that influenced survival in males and both the degree of outbreeding and recent inbreeding that influenced survival in females. Our analyses suggest that mean d2 is an easy-to-calculate measure of inbreeding and degree of outbreeding that can reveal interesting interactions between genetics and ecology.  相似文献

A male done of the red alga Gracilaria tikvahiae McLachlan spontaneously produced a bisexual frond which remained bisexual in subsequent subcultures. Both male and female components of bisexual fronds were functional; however, some unusual results were obtained in crosses. When bisexual fronds were crossed with a normal haploid male, the resulting carpospores all developed into diploid male gametophytes. When bisexual plants were self fertilized, all the carpospores yielded diploid bisexual gametophytes. Only when bisexual plants were crossed to normal haploid females, did carpospores develop into diploid tetrasporophytes as they normally do. The F1 gametophyte generation obtained from these tetrasporophytes, however, included not only females and males but also bisexual plants, in a 2:1:1 ratio. These results are consistent with the interpretation that bisexual plants have a recessive mutation of a gene other than the primary sex determining locus, and that this mutation is expressed only in male plants. It is suggested that the altered gene may ordinarily have a regulatory function in the maintenance of the dioecious condition.  相似文献

The presence of the “symbiotic” red alga Hypneocolax stellaris Borgesen was found to substantially reduce the growth rate of Hypnea musciformis (Wulfen) Lamouroux. The growth rate of injected field material was 40% less than unifected thalli. Likewise, the growth rate of host thalli infected in the laboratory was reduced 70% compared to uninfected thalli. Neither the quantity nor quality of carrageenan extracted from Hypnea musciformis was directly affected by the presence of Hypneocolax stellaris. In a mariculture system the presence of a symbiotic alga on its host would be expected to reduce the host's growth rate and thereby lower its economic potential.  相似文献

The effect of different light wavelengths on the development of lesions induced by Alternaria tenuissima in broad bean leaves was investigated. Lesion development was completely suppressed in red‐light‐irradiated broad bean leaflets, irrespective of isolate or spore concentration. Pre‐treatment of leaflets with red light for 24 h before inoculation also suppressed lesion development. Alternaria tenuissima failed to produce infection hyphae in red‐light‐irradiated broad bean leaflets. These results indicate that disease suppression in broad bean leaflets is due to light‐induced resistance. Spore germination fluid (SGF) of A. tenuissima allowed non‐pathogenic Alternaria alternata to infect wounded and unwounded broad bean leaflets kept in the dark, results suggesting that SGF induced susceptibility. Red light suppressed susceptibility induced by A. tenuissima SGF; thus, lesion formation and development were suppressed when leaflets inoculated with the spores of A. alternata suspended in A. tenuissima SGF were kept under red light. From these results, we conclude that red light induced resistance in broad bean to leaf spot disease caused by A. tenuissima, and that SGF induced susceptibility of broad bean leaflets to a non‐pathogenic isolate of A. alternata.  相似文献

Although ecological theory exists to predict dynamics in communities with intraguild predation (IGP), few empirical tests have examined this theory. IGP theory, in particular, predicts that when two competitors interact via IGP, with increasing resource productivity: (1) the IG predator will increase in abundance as the IG prey declines, and (2) increasing dominance of the IG predator will cause resource density to increase. Here, we provide a first test of these predictions in a field community consisting of a scale insect and its two specialist parasitoids, Aphytis melinus (the IG predator) and Encarsia perniciosi (the IG prey). The shared resource, California red scale, is a pest of citrus, and its productivity varies across a threefold range among citrus cultivars. We examined both absolute and relative densities of parasitoids along this natural gradient of scale productivity in three citrus cultivars (orange, grapefruit and lemon). Although both parasitoid species were found in all three cultivars, their abundances reflected those predicted by IGP theory: the IG prey species dominated at low productivity and the IG predator dominated at high productivity. This relationship was caused by an increase in Aphytis density with productivity. In addition, the density of scale increased with the dominance of the IG predator. These results from a field system demonstrate the important dynamic outcomes for food webs with IGP.  相似文献

Diagnostic photopigment analysis is a useful tool for determining the presence and relative abundance of algal groups in natural phytoplankton assemblages. This approach is especially useful when a genus has a unique photopigment composition. The toxic dinoflagellate Karenia brevis (Davis) G. Hansen & Moestrup comb. nov. shares the diagnostic pigment gyroxanthin‐diester with only a few other dinoflagellates and lacks peridinin, one of the major diagnostic pigments of most dinoflagellate species. In this study, measurements of gyroxanthin‐diester and other diagnostic pigments of K. brevis were incorporated into the initial pigment ratio matrix of the chemical taxonomy program (CHEMTAX) to resolve the relative contribution of K. brevis biomass in mixed estuarine phytoplankton assemblages from Florida and Galveston Bay, Texas. The phytoplankton community composition of the bloom in Galveston Bay was calculated based on cell enumerations and biovolumetric measurements in addition to chl a‐specific photopigment estimates of biomass (HPLC and CHEMTAX). The CHEMTAX and biovolume estimates of the phytoplankton community structure were not significantly different and suggest that the HPLC–CHEMTAX approach provides reasonable estimates of K. brevis biomass in natural assemblages. The gyroxanthin‐diester content per cell of K. brevis from Galveston Bay was significantly higher than in K. brevis collected from the west coast of Florida. This pigment‐based approach provides a useful tool for resolving spatiotemporal distributions of phytoplankton in the presence of K. brevis blooms, when an appropriate initial ratio matrix is applied.  相似文献

Regulatory volume decrease, following physical swelling of red cells from brown trout Salmo trutta , was almost complete in oxygenated cells but much less in deoxygenated cells. There was a small, insignificant regulatory volume increase, following physical shrinkage. Amiloride had no effect on this response, indicating that hypertonic shrinkage did not activate the Na+/H+ exchanger. However, cell volume was increased markedly in shrunken cells by addition of noradrenaline, with deoxygenated cells showing complete recovery. These data show that the previously reported differences in volume regulation between the red cells of brown trout and rainbow trout Oncorhynchus mykiss are not present and that both species appear to have lost volume sensitivity of the Na+/H+ exchanger.  相似文献
