We isolated and characterized eight polymorphic microsatellite loci for a Texas population of three-toed box turtle, Terrapene carolina triunguis, using a refined hybridization capture procedure. All eight primer pairs amplified successfully at all loci in seven Texas ornate box turtles (T. ornata ornata). Due to the decline and conservation concerns of North American box turtles, these isolated microsatellites may be a most valuable tool for evaluating baseline population genetic structure for threatened box turtle populations.  相似文献

Phase diagrams of 3-component lipid bilayer mixtures containing cholesterol reveal major differences among the different types of lipids. Here we report that mixtures of cholesterol together with POPC and a high-melting temperature PC or sphingomyelin show different phase behavior from similar mixtures that contain DOPC or di-phytanoyl-PC instead of POPC. In particular, only one region of macroscopic phase coexistence occurs with POPC, a region of coexisting liquid disordered and solid phases, {Lα + Lβ}. Fluorescence microscopy imaging is useful for these studies, but is subject to artifactual light-induced domain formation, as reported by Ayuyan and Cohen [A.G. Ayuyan, F.S. Cohen, Lipid peroxides promote large rafts: Effects of excitation of probes in fluorescence microscopy and electrochemical reactions during vesicle formation, Biophys. J. 91 (2006) 2172-2183.]. This artifact can be attenuated by decreased illumination and low dye concentration. The use of the free radical scavenger n-propyl gallate can reduce the artifact, but this molecule enters the bilayer and itself perturbs the phase behavior. We suggest that the light-induced domain separation artifact might actually arise from pre-existing lipid clusters that are induced to coalesce, and therefore indicates highly nonrandom mixing of the lipid components.  相似文献

A standardized sampling method was used to evaluate turnover (β diversity) among cactus species assemblages along a 798 km long latitudinal megatransect across the Chihuahuan Desert Region, from north-central Mexico to southern Texas. A total of 71 cactus species were found along the megatransect, 66.2% of which appeared at low frequencies, mostly as a consequence of their highly discontinuous distribution pattern. At the scale the study was conducted, there was always species turnover among cactus assemblages. The rate of turnover among contiguous sites primarily fluctuated from low to medium, but when all site combinations were considered (contiguous and non-contiguous), medium β diversity values were predominant (β = 0.331–0.66); however, 25.4% of the site pair combinations registered high values (β = 0.661–1.0). Our results showed that turnover among cactus species assemblages in the CDR does not consist for the most part of a process of species succession in the geographic space. Instead, we concluded that the continuous spatial changes in cactus species composition are primarily explained by the commonly intermittent distribution patterns of the species, by the presence in the megatransect of species at the margin of their distribution range, and, to a lesser extent, by the existence of narrowly endemic species.  相似文献

Abstract: Survival and cause-specific mortality estimates are needed to develop effective conservation strategies for the endangered ocelot (Leopardus pardalis) in the United States. We radiomonitored 80 ocelots (36 F, 44 M) from 1983 to 2002 and analyzed survival and cause-specific mortality rates. Pooled estimates of annual survival rates differed between resident (Ŝ = 0.87) and transient (Ŝ = 0.57) ocelots (P = 0.02); therefore, survival and cause-specific mortality analyses were partitioned for resident and transient ocelots. Sex-specific annual survival was similar between resident ocelots (M = 0.92, F = 0.83, P = 0.16) and transient ocelots (M = 0.53, F = 0.63, P = 0.75). Most mortalities were from human (e.g., ocelot-vehicle collisions; M = 45%) and natural (e.g., animal attack, disease; M = 35%) sources. Transient ocelots had higher natural mortality rates (disease, intraspecific mortality; M = 0.26) than resident ocelots (M = 0.04, P = 0.03). Other sources of mortality did not differ (P ≥ 0.10) between resident or transient ocelots or male and female resident or transient ocelots (P ≥ 0.08). Human population expansion within the Lower Rio Grande Valley of southern Texas, USA, will increase transportation-related problems and decrease the quantity of ocelot habitat, leading to increased ocelot-vehicle collisions and possibly cause more transient behavior, thus significantly lowering ocelot survival. Research and development of ocelot road underpasses should be conducted to mitigate ocelot-vehicle collisions.  相似文献

It has been established that Molpadia hemoglobin tends to dissociate into subunits as oxygen is bound. The kinetics and equilibria of carbon monoxide and ehtylisocyanide binding reported here show a dependence on protein concentration that supports the conclusions that the aggregated hemoglobin has a lower ligand affinity than the dissociated subunits. This is true for the isolated D-chain as well as for the unfractionated hemolysate that contains four distinct polypeptide chains (A-D). This indicates that even homopolymers of Molpadia hemoglobin have lower ligand affinity than the dissociated subunits. At high protein concentration hemolysates of Molpadia hemoglobin show slight cooperativity. The time course of ligand binding to the deoxy D-chain also suggests cooperative interactions, The low affinity of the aggregated state may have a different molecular explanation than in human hemoglobin were tetramers of identical subunits (as in Hb H) show high oxygen affinity. The absence of tyrosine and histidine at the C-tremini of the Molpadia D-chains also suggests a different stabilization of the low affinity deoxy state. An additional functional difference between Molpadia hemoglobin and human hemoglobin is that organic phosphate do not alter the ligand affinity of the sea cucumber hemoglobin.  相似文献

Laboratory experiments with batch cultures showed that the Texas brown tide alga, Aureoumbra lagunensis Stockwell, DeYoe, Hargraves et Johnson, produced a large amount of extracellular polymeric substance (EPS) that formed an extracellular polysaccharide mucus layer. Both dissolved and particulate carbohydrate and EPS concentrations increased as A. lagunensis cultures progressed from exponential growth phase, through stationary phase, to declining phase. Particulate carbohydrate and EPS concentrations per cell were more than three times higher during the declining phase than that in exponential growth phase, reflecting a large increase in the EPS mucus layer. The amounts of carbohydrate and EPS produced by A. lagunensis were significantly higher under hypersaline conditions. The thicker EPS mucus layer surrounding A. lagunensis cells under hypersaline conditions might be a protective adaptation that permits it to bloom under hypersaline conditions that most other phytoplankton cannot survive. This could be one of the reasons why the Texas brown tide persisted in the Laguna Madre, an often hypersaline coastal lagoon, for 7 years.  相似文献

Mottled ducks (Anas fulvigula) are endemic to the Gulf Coast of North America, and their range stretches from Alabama to the Laguna Madre of Mexico, with a distinct population in peninsular Florida and an introduced population in South Carolina. As one of the few non-migratory ducks in North America, mottled ducks depend on a variety of locally available habitat throughout the annual cycle, and threats to these landscapes may affect mottled ducks more acutely than migratory species. Annual population monitoring has revealed declines in mottled duck populations in Texas and Louisiana since 2008, and the genetic integrity of the Florida population has been muddled by the presence of large numbers of feral mallards (Anas platyrhynchos) resulting in hybridization. Similar to other closely related dabbling ducks, mottled duck populations are influenced by recruitment and breeding season survival, so changes in these factors may contribute to population decline. Accordingly, researchers have attempted to address various aspects of mottled duck breeding season ecology and population dynamics since the 1950s. We conducted a literature review on this topic by searching a combination of key terms using Google Scholar, including mottled duck, nesting ecology, habitat use, breeding incidence, nest success, brood, and breeding season survival, and followed citation trees to eventually aggregate information from nearly 50 publications on mottled duck breeding ecology. Our review concluded that mottled ducks use brackish and intermediate coastal marsh, including managed impoundments, and agricultural land during the breeding season. Their nests can be found in pastures, levees, dry cordgrass marsh, cutgrass marsh, spoil banks, and small islands. Nesting propensity and nest success estimates are often lower than other waterfowl species that are characterized by stable or increasing populations. Broods use wetlands composed of a mix of open water with submerged and emergent vegetation. Breeding season survival is higher for the Florida population than the western Gulf Coast population, but adult survival in both geographies is comparable to (or higher than) that of other dabbling duck species. Breeding habitat use, breeding season survival, and nest-site selection and success have been studied extensively in mottled ducks, whereas information on nesting propensity, renesting intensity, and post-hatch ecology is lacking. © 2021 The Wildlife Society.  相似文献

The northern bobwhite (Colinus virginianus; bobwhite) exists at the edge of its thermal tolerance in southern Texas, USA, a region characterized by extreme heat and periodic drought. Extreme heat and drought events are predicted to increase in frequency and intensity in semi-arid regions, leading biologists to emphasize management of thermal cover. The degree to which temperatures within patches of woody vegetation vary and the specific vegetation structural characteristics that create cooler microclimates within these patches are unknown. We evaluated temperature variation between selected and available sites, vegetation characteristics facilitating cooler microsites, and whether these characteristics in addition to temperature were important predictors of microsite selection within woody patches. We radio-tracked 83 bobwhites 2–3 times/week during April–August 2017–2018. We recorded operative and ground surface temperatures and measured woody and herbaceous vegetation height, canopy density, and overlapping woody and herbaceous cover at used and paired random locations. Within the same woody patch, 80% of used locations contained operative temperatures lower than or equal to random locations. There was a weak relationship between the vegetation variables we measured and temperature at paired locations, indicating that none of the vegetation variables alone can effectively reduce temperatures. Temperatures within woody patches are likely strongly tied to diurnal variation in solar angle and ability to create shade. Cooler temperatures, dense canopies, and overlapping woody vegetation had the greatest effects on relative probability of microsite use during the middle of the day, whereas warmer temperatures and taller woody vegetation had the greatest relative effects during the morning and evening. Our results suggest that temperature was influential across diurnal periods but foraging and predation avoidance may have also been important. Maintenance and preservation of dense woody vegetation and diverse shrub understories must be prioritized in habitat management for bobwhites in semi-arid regions. © 2021 The Wildlife Society.  相似文献

Laboratory microcosms were used to assess whether tadpole shrimp, Triops sp., affect community structure of other native macroinvertebrates in playa lakes of the Southern High Plains of Texas. Removal of tadpole shrimp shortly after hatching reduced abundances of many taxa, and decreased subsequent taxonomic richness and diversity. For many invertebrates, the presence of tadpole shrimp in low numbers had a positive effect on mean abundance. Direct effects of tadpole shrimp include the reduction of prey species abundance, which in turn may alter biotic interactions among other taxa. Indirect effects include physical modification of the environment during foraging through surface sediments. Results suggest that tadpole shrimp may be a key species controlling structure of macroinvertebrate communities in playa lakes.  相似文献
