Populations of rare tree species such as the dioecious and anemophilous yew (Taxus baccata) are especially prone to extinction if they occupy marginal habitats. This is the case of yew populations growing in Mediterranean mountains under dry climate and a severe anthropogenic disturbance regime, which show insufficient regeneration. We examined two of the likely mechanisms driving this population decline. The study was conducted in a central Spanish yew population in 2005 and 2006. On one hand, we tested for the effect of the amount of pollen received by female flowers by means of a pollen addition experiment. On the other hand, we tested for the effect of initial fruit set on abortion and mature fruit production. Finally, we evaluated the effect of the spatial arrangement of male and female yew trees on realized fecundity. The success of fertilization and fruiting in the experimental population was pollen-limited. Female trees aborted ca. 70% of the fruit set in both years and treatments. Fruit set was significantly influenced by the females' neighbourhood, with fecundity decreasing exponentially with distance to the nearest male in both years. Streptozotocin-diabetic rats were maintained on a stock diet for 16 weeks and some were given conduritol A (10 mg/kg/day). The administration of conduritol A, having a hypoglycemic effect, markedly prevented the diabetic rats from getting cataracts. In practice, conduritol A inhibited aldose reductase (EC 1.1.1.21) that is capable of catalyzing the conversion of aldoses to sugar alcohols in the polyol pathway. In an in vitro assay, lens aldose reductase was most effectively inhibited by conduritol A when α,β-d-glucose was used as a substrate. Some enzymes from the rats were not inhibited by conduritol A. Neither an intraperitoneal injection nor oral dosage of conduritol A caused acute toxicity in the rats. These findings suggest that the inhibition of lens aldose reductase by conduritol A may be responsible for its cataract-suppressing effect. 相似文献

The uptake rate of 14C-labelled sucrose, myo-inositol and PC was studied in callus cultures of two oilseed rape cultivars, characterized by different in vitro regeneration ability. Transfer of calli onto regeneration stimulating medium resulted in changes of examined substances uptake
rate, which were depended on tissue morphogenic potential. Non-regenerating calli of both cultivars increased uptake rate
of sucrose whereas changes in incorporation of other compounds were under genome control. Significant increase of uptake rate
of all tested compounds was observed as result of organogenesis initiation. Such differences, in the responses of organogenic
and non-organogenic tissue indicate that this parameter could be useful as marker of organogenesis
A correlation was observed between the rate of sucrose uptake and its concentration in the medium, which suggests an advantage
to passive transport through the callus cell membrane. Lack of such correlation in the case of other labels indicates that

研究了基因型、外植体、植物激素和光暗等因子对山药愈伤组织诱导的影响。结果表明 :(1)不同基因型山药均能诱导形成愈伤组织 ,但出愈率不同 ,其排列顺序是 :“4 7号”山药 >铁棍山药 >太谷山药。 (2 )不同外植体在同一种培养基上的诱导率存在差异。同一外植体在不同植物激素浓度配比中 ,诱导率也不同。叶片、茎段、零余子的最佳激素配比均为 6 - BA2 (mg/ L,以下单位同 ) +NAA2 ,诱导率分别为 53.3%、 6 5.6 %、 10 0 % ,茎尖的最佳激素配比为 6 - BA2 +NAA0 .2 ,诱导率为 93.3%。 (3)植物激素在愈伤组织诱导过程中起关键的作用。细胞分裂素与生长素组合使用优于单一激素。二者的浓度配比不同 ,出愈率也不同。 (4 )光暗条件对不同外植体愈伤组织诱导的影响不同。暗培养有利于零余子的诱导 ,而光培养则有利于叶片的诱导。 相似文献

Summary The present study aimed to evaluate the response to salinity of Populus euphratica, which is more salt-resistant than other poplar cultivars, at the cellular level. To this purpose, callus was induced from
shoot segments of P. euphratica on Murashige and Skoog (MS) medium supplemented with 0.5 mg l−1 (2.2 μM) 6-benzyladenine (BA) and 0.5 mg l−1 (2.7 μM 1-naphthaleneacetic acid (NAA). Callus was transferred to MS medium supplemented with 0.25 mg l−1 (1.1 μM) BA and 0.5 mg l−1 NAA. The relative growth rate of callus reached a maximum in the presence of 50 mmol l−1 NaCl and growth was inhibited with increasing NaCl concentrations. Examination of the changes of osmotic substances under
salt stress showed that accumulation of proline, glycine betaine, and total soluble sugars increased with increasing salt

Patterns of variation in nuclear DNA content and chromosome number were analysed in a temporal sequence, during in vitro growth of calli and cell suspensions in two monohaploids, a dihaploid and a tetraploid of potato (Solanum tuberosum). The results showed that both polyploidization and aneuploidy occurred during the initial stages of callus induction in all the genotypes. With further growth of callus, the frequency and extent of polyploidy and aneuploidy increased. In addition, the patterns of DNA and chromosome variation in cell suspension cultures revealed continued mitotic activity and transmission of cells with higher ploidy levels and aneuploidy. The results suggest that endoreduplication as well as endomitosis are important mechanisms of polyploidization, and that chromosome lagging and non-disjunction contribute to the production of aneuploidy.The various genotypes cultured under the same in vitro growth conditions differed in genetic instability, as assessed from the rate and degree of polyploidization and aneuploidy. Monohaploids showed more rapid rate of polyploidization than the dihaploid and tetraploid potatoes.
