Characterization of populations of aerobic hydrogen-oxidizing soil bacteria   总被引:2,自引:0,他引:2  
Abstract Freshly isolated soil bacteria were screened for different characteristics of the H2 metabolism without prior selection for growth on H2. The bacteria were isolated from different grain size fractions of a neutral meadow cambisol and an acidic forest cambisol, and then tested (1) for the ability to oxidize H2, (2) for chemolithoautotrophic growth on H2 as sole electron donor and energy source, (3) for DNA-DNA-hybridization with two hydrogenase gene fragments from Alcaligenes eutrophus and Rhizobium leguminosarum , and (4) for reduction of 2,3,5-triphenyl-2H-tetrazoliumchloride (TTC) in the presence of H2. Many (65–90%) of the isolates were able to reduce TTC, but only 30–65% were actually able to oxidize H2 indicating that the TTC test was not a specific characteristic for H2 oxidation ability. The TTC test was only reliable in pure cultures of known bacteria with optimized test conditions, here shown for Alcaligenes eutrophus, Bradyrhizobium japonicum and Nocardia opaca , but not in mixed cultures of unknown bacteria. Still less (< 30%) of the isolates were able to grow chemolithoautotrophically indicating that culturable aerobic bacteria with the ability for H2 oxidation are more abundant than bacteria with the ability for chemolithoautotrophic growth. The DNA-DNA-hybridization test failed to detect many of the bacteria with H2 oxidation activity, probably since the hydrogenase genes present in the isolates were too diverse to be all detected by the DNA probes applied.  相似文献

Mucociliary epithelia are essential for homeostasis of many organs and consist of mucus-secreting goblet cells and ciliated cells. Here, we present the ciliated epidermis of Xenopus embryos as a facile model system for in vivo molecular studies of mucociliary epithelial development. Using an in situ hybridization-based approach, we identified numerous genes expressed differentially in mucus-secreting cells or in ciliated cells. Focusing on genes expressed in ciliated cells, we have identified new candidate ciliogenesis factors, including several not present in the current ciliome. We find that TTC25-GFP is localized to the base of cilia and to ciliary axonemes, and disruption of TTC25 function disrupts ciliogenesis. Mig12-GFP localizes very strongly to the base of cilia and confocal imaging of this construct allows for simple visualization of the planar polarity of basal bodies that underlies polarized ciliary beating. Knockdown of Mig12 disrupts ciliogenesis. Finally, we show that ciliogenesis factors identified in the Xenopus epidermis are required in the midline to facilitate neural tube closure. These results provide further evidence of a requirement for cilia in neural tube morphogenesis and suggest that genes identified in the Xenopus epidermis play broad roles in ciliogenesis. The suites of genes identified here will provide a foundation for future studies, and may also contribute to our understanding of pathological changes in mucociliary epithelia that accompany diseases such as asthma.  相似文献

Defining root death in studies of root dynamics is problematic because cell death occurs gradually and the resulting effects on root function are not well understood. In this study, metabolic activity of grape roots of different ages was assessed by excised root respiration and tetrazolium chloride reduction. We investigated changes in metabolic activity and patterns of cell death occurring with root age and changes in root pigmentation. Tetrazolium chloride reduction of roots of different ages was strongly correlated to respiration ( R 2 = 0.786). As roots aged, respiration and tetrazolium chloride reduction declined similarly, with minimum metabolic activity reached at six weeks. Tetrazolium chloride reduction indicated that the onset of root browning corresponded to a 77% reduction in metabolic activity ( P < 0.001). Anatomical examination of roots at each pigmentation stage showed that even though some cells in brown roots were still alive, these roots were functionally dead. The effect of using different definitions of root death in relation to root survivorship was determined in a study of 'Concord' grapes with two pruning treatments, using three criteria for root death: browning, blackening or shriveling, and disappearance. There was no effect of vine pruning on root life span when life span was defined as the time from first appearance to the onset of browning. However, if death was judged as the point when roots either became black or shriveled or disappeared, vine pruning decreased root life span by 34% and 40%, respectively ( P < 0.001), and also increased the decay constant for root decomposition by about 45% ( P < 0.001). We conclude that the discrepancy among determinations of root life span assessed with different definitions of death might be partly caused by the latter evaluations of root life span incorporating a portion of root decomposition in definitions of root death.  相似文献

以马铃薯栽培种呼自83-213无菌试管苗茎尖为材料,通过开展2,3,5-氯化三苯基四氮唑(TTC,2,3,5-Triphenyl tetrazolium chloride)茎尖活力染色关键因素研究,优化了马铃薯茎尖TTC活力染色条件,确定了适合的染色温度为40℃,染色时间为2 h。利用优化的TTC活力染色条件,对马铃薯茎尖小滴玻璃化超低温保存关键步骤处理茎尖进行TTC活力观察。研究发现:经蔗糖预培养(MS培养液添加0.3 mol/L和0.5 mol/L蔗糖)的茎尖与新鲜茎尖均保持高活力;经PVS2处理后茎尖表现时空特异性活力丧失和存活,分生组织和叶原基中间区域仍保持较高活力。通过对茎尖TTC活力染色面积测定,发现当茎尖TTC活力染色面积比≥0.4时,TTC活力染色与恢复培养存活率呈极显著正相关。  相似文献

Previous studies about the effects of experimental warming on tree species have focused primarily on response of morphology and physiology in leaf and biomass allocation in the growing season, and a few studies considered the importance of roots. Based on the available evidence, it is unclear whether photosynthesis rate is enhanced by night warming in late autumn an issue that deserves further investigation. Thus, we exposed two coniferous species, Picea asperata and Abies faxoniana, to night warming continued throughout the year to investigate morphological and physiological responses of roots and leaves in the autumn. The results showed that night warming caused significant increases in net influxes of NH4+ and NO3 in P. asperata seedlings corresponding well with net H+ efflux and net influx of O2. Meanwhile, night warming had a positive effect on foliar gas exchange such as net photosynthesis rate, apparent quantum efficiency, dark respiration rate and maximum quantum efficiency of PS II, and nitrate reductase activity of roots. Additionally, root morphology such as total roots length, surface area, specific root area and specific root length was also stimulated by night warming. In contrast, night warming decreased concentrations of non-structural carbohydrate in leaves and roots of both species in autumn. The present study demonstrates that night warming would enhance late autumn leaf photosynthetic rate, and increase N uptake capacity of roots.  相似文献

受一定浓度SO_2熏气的大豆幼苗出现可见伤害以后,在继续熏气的过程中可见伤害程度不再进一步发展,表现出一定的适应性。与此相联系,膜透性增加和TTC还原力下降这两个SO_2伤害指标也得到一定程度的恢复,SO_2熏气使游离氨基酸含量增加,随着熏气时间延长,增加的游离氨基酸含量回到对照水平,提示受扰乱的代谢过程有所恢复。低浓度SO_2预处理提高了大豆对高浓度SO_2的抗性,与抗氧化有关的巯基(-SH)含量显著增加,超氧物歧化酶(SOD)活性也有所增强,同工酶谱分析显示有SOD同工酶带的酶量增加或新带出现。  相似文献

Summary The heat shock (HS) response in callus cultures of the ornamental plant Gerbera jamesonii H. Bolus var. hybrida was analyzed. A HS at 35° C or 40° C for 4 h induced (a) the synthesis of several heat shock proteins (HSPs), especially in the small molecular weight range and some spots corresponding to HSP70 components, and (b) an increase in the steady state levels of some specific mRNAs. At the nonstressing temperature (26° C), a sustainable level of translation for HSP70 was indeed carried out, as confirmed by immunological analysis with a monoclonal antibody against cotton HSP70. The steady state levels of mRNAs measured before and after a HS by Northern hybridization showed an increase with the heterologous probes HSP17.4, HSP17.6, and HSP21, whereas the probes HSC70 and HSP70 did not show any difference between the levels of control and HS-mRNAs. A pretreatment at 35° C, which induced a set of HSPs in the callus cultures, decreased the cell damage upon exposure to a temperature of 45° C as determined either with a regrowth test or by the tetrazolium reduction assay. Typically, as with the whole plants, callus of Gerbera jamesonii possessed the ability to respond to HS both by inducing HSPs and by developing an acquired thermotolerance.  相似文献

Aims:  To identify lactic acid bacteria (LAB) colonies isolated from sediments of a coastal marsh by the reduction of 2,3,5‐triphenyltetrazolium chloride (TTC) in MRS medium. Methods and Results:  Single colonies isolated from sediments of a coastal marsh by enrichment in MRS broth were selected from MRS‐TTC plates and classified according to colony phenotype based on TTC reduction. A total of 37 colonies grouped in seven different phenotypes were identified by analysis of its 16S ribosomal gene sequence. Most isolates belonged to the Firmicutes phylum, mainly to orders Bacillales and Lactobacillales. LAB were represented by 20 isolates, 15 of which belong to the genus Weissella. Conclusions:  Enrichment in MRS was highly selective for the isolation of bacteria belonging to phylum Firmicutes. Several different phenotypes were developed by LAB and must be considered during LAB isolation based on TTC reduction. Significance and Impact of the Study:  To our knowledge, this is the first study aimed at determining a relationship between colony phenotype from TTC reduction and a partial identification of isolates based on 16S ribosomal gene sequence similarities. Besides, this is the first report of isolation of W. cibaria from environmental samples.  相似文献
