The goal of the present study was to obtain new insights into the mechanisms underlying drought stress adaptations in barley plants. For this purpose we evaluated changes in endogenous abscisic acid (ABA) and its glucose ester conjugate (ABAGE), as well as changes in proline content, water relations and growth parameters of five barley genotypes with different drought resistance characteristics. Different responses among the five genotypes studied (Ardahoui, Pakistan, Rihane, Manel ad Roho) led to changes in their pattern of growth and development under drought conditions. Water stress induced a reduction in relative water content, as well as an increase in proline content and endogenous ABA concentrations in all tested genotypes. The lack of water led to a 2-fold increase in proline content for var. Rihane and to a 5-fold increase in endogenous ABA for cv. Ardhaoui. Also, increases in endogenous ABAGE in all genotypes except for cv. Ardhaoui were observed. Our results show that changes in ABA and ABAGE correlated with variations in proline content and growth parameters of these genotypes which present different mechanisms to cope with water stress. We also suggest that new regulatory mechanisms implying ABA mobilization can be of great importance in adaptation of barley to drought.  相似文献

本文以分株相连的美丽箬竹(Indocalamus decorus)盆栽苗为试材,设置3个盆栽基质相对含水率(90%±5%高水势、60%±5%中水势、30%±5%低水势)和3个水势差处理(90%~60%、60%~30%、90%~30%),研究美丽箬竹克隆分株抗氧化系统对异质水分环境的响应规律,分析水分生理整合发生条件、方向和强度。结果表明:随处理时间的延长,美丽箬竹叶片MDA、可溶性蛋白质含量和CAT、POD活性总体上呈升高趋势;SOD活性呈降低趋势;随分株间水势差的增大,低水势处理分株叶片CAT、SOD活性和可溶性蛋白质含量升高,POD活性、MDA含量降低,高水势处理分株的变化规律相反,中水势处理分株叶片抗氧化生理指标总体上变化不明显;在异质水分环境下美丽箬竹克隆系统存在水分生理整合作用,分株首先保障自身的水分需求,当克隆分株同时存在水分丰盈、亏缺时,会发生强烈的从高水势向低水势分株的水分转移,分株间水势差越大,整合强度越高;克隆分株在水分生理整合上存在"利己"和"利他"行为。本研究为商品竹林水分人工供应提供了理论依据。  相似文献

Modular design is at the foundation of contemporary engineering, enabling rapid, efficient, and reproducible construction and maintenance of complex systems across applications. Remarkably, modularity has recently been discovered as a governing principle in natural biological systems from genes to proteins to complex networks within a cell and organism communities. The convergent knowledge of natural and engineered modular systems provides a key to drive modern biotechnology to address emergent challenges associated with health, food, energy, and the environment. Here, we first present the theory and application of modular design in traditional engineering fields. We then discuss the significance and impact of modular architectures on systems biology and biotechnology. Next, we focus on the very recent theoretical and experimental advances in modular cell engineering that seeks to enable rapid and systematic development of microbial catalysts capable of efficiently synthesizing a large space of useful chemicals. We conclude with an outlook towards theoretical and practical opportunities for a more systematic and effective application of modular cell engineering in biotechnology.  相似文献

Headbob displays play an important role in the social behaviour of male green iguanas (Iguana iguana). Four types of headbobs were identified, and the variability in form and context were determined from quantitative analyses of displays filmed in the field. The Roll is a conspicuous advertisement display. The Shudder, the most variable in form, occurred during close contact between males and females. The Roll-shudder is intermediate between the Roll and Shudder in its form and use. The Signature Bob, the most stereotyped display in form, punctuates much activity and is the most variable in contextual use. This display encodes species identity, and possibly individual identity. The conspicuous nature of the headbob displays was enhanced by the intensified colour of territorial males, and by the selection of highly visible display posts. This system of visual displays is viewed as the product of related sets of constraints, including the iguanas' physical capacities, the forest environment, and the social environment.  相似文献

Simultaneous measurements were made by radioimmunoassay of testosterone, 17β-hydroxy-5 a -androstan-3-one (DHT), 5 a -androstane-3 a, 17 β -diol (3a-diol), and 5 a-androstane-3 β, 17β-diol (3β-diol) in the testicular venous plasma (TVP) and peripheral plasma (PP) of 30, 45, and 55 day old rats. At 30 days of age, the preponderant androgen in both plasmas was 3a-diol but testosterone predominated by day 55. Testosterone levels increased with age in both TVP (6.39, 15.08, and 54.93 ng/ml on days 30, 45, and 55 respectively) and PP (0.13, 0.56, and 1.02 ng/ml on days 30, 45 and 55 respectively) whereas 3a-diol concentrations decreased in TVP (48.07 ng/ml, day 30; 24.85 ng/ml, day 55) though not peripherally (range: 0.41–0.52 ng/ml). DHT was low in both TVP and PP and appeared to rise only slightly although the increase was not statistically significant. Levels of 3β-diol remained low and unchanged. These observations suggest that the total androgen content of the venous effluent from the prepubertal rat test is is quite high and that significant changes in peripheral interconversions of androgens are occurring during sexual maturation.  相似文献

In phragmoplast-assisted cytokinesis of somatic cells, vesicle fusion generates a cell plate that matures into a new cell wall and its flanking plasma membranes. Insight into this dynamic process has been gained in the past few years and additional molecular components of the basic machinery of cytokinesis have been identified. Specialized modes of cytokinesis occur in meiosis and gametophyte development, and recent studies indicate that they are genetically distinct from somatic cytokinesis.  相似文献

The vertebrate cranial base is a complex structure composed of bone, cartilage and other connective tissues underlying the brain; it is intimately connected with development of the face and cranial vault. Despite its central importance in craniofacial development, morphogenesis and tissue origins of the cranial base have not been studied in detail in the mouse, an important model organism. We describe here the location and time of appearance of the cartilages of the chondrocranium. We also examine the tissue origins of the mouse cranial base using a neural crest cell lineage cell marker, Wnt1-Cre/R26R, and a mesoderm lineage cell marker, Mesp1-Cre/R26R. The chondrocranium develops between E11 and E16 in the mouse, beginning with development of the caudal (occipital) chondrocranium, followed by chondrogenesis rostrally to form the nasal capsule, and finally fusion of these two parts via the midline central stem and the lateral struts of the vault cartilages. X-Gal staining of transgenic mice from E8.0 to 10 days post-natal showed that neural crest cells contribute to all of the cartilages that form the ethmoid, presphenoid, and basisphenoid bones with the exception of the hypochiasmatic cartilages. The basioccipital bone and non-squamous parts of the temporal bones are mesoderm derived. Therefore the prechordal head is mostly composed of neural crest-derived tissues, as predicted by the New Head Hypothesis. However, the anterior location of the mesoderm-derived hypochiasmatic cartilages, which are closely linked with the extra-ocular muscles, suggests that some tissues associated with the visual apparatus may have evolved independently of the rest of the “New Head”.  相似文献

Interferon gamma (IFN gamma) induces the expression of three catalytic subunits of the 20S proteasome that can replace their constitutive homologues to form the "immunoproteasome," named to reflect its antigen presentation function. However, immunoproteasome levels and their modulation in nonimmune tissues remain unknown. A disrupted lens differentiation program observed in transgenic mice that constitutively express IFN gamma in the immune-privileged lens tissue suggests a role for this cytokine in differentiation. We have developed a competitive RT-PCR assay that demonstrates substantially increased levels of immuno subunits and unchanged levels of constitutive subunits in transgenic compared to wild-type lenses. Similar results were observed with IFN gamma treated alpha TN4-1 lens epithelial cells. A comparison of these subunits in different immune and nonimmune mouse tissues revealed unique expression patterns. The presence of immuno subunits in nonimmune tissues such as lens suggests that the immunoproteasome may also have nonimmune functions, such as that in lens differentiation.  相似文献

Drinking water reservoirs in agricultural dominated watersheds are particularly vulnerable to cyanobacterial blooms. A major byproduct of cyanobacteria is the production of objectionable taste and odor compounds such as geosmin. During May 1997 to September 1998, we studied spatial and temporal patterns of cyanobacterial abundance and composition with respect to a series of physical and chemical properties in Clinton Lake, located in east central Kansas, USA. Our results suggest that nutrients (in particular TN, NO3–N concentrations), turbidity, and hydrologic regime all played potentially important roles in regulating cyanobacterial production. Specifically, low levels of nitrogen coupled with the internal release of phosphorus from the lake sediment under brief periods of anoxia may have helped promote cyanobacterial blooms. There was also a strong association between cyanobacterial blooms, geosmin production, and most taste and odor events in Clinton Lake. Anabaena circinalis appeared to be the source for geosmin production as a result of senescing algal cells just after the primary die-off of cyanobacteria.  相似文献

旨在探讨体外培养条件下HBx蛋白对TTRAP(TRAF and TNF receptor-associated protein)基因转录水平表达的影响。用RT-PCR及Real-time PCR检测TTRAP在HepG2细胞和HepG2.2.15细胞中的表达;构建TTRAP启动子虫荧光素酶报告质粒;分别与HBV、HBs、HBp、HBc、HBx表达质粒共转染HepG2细胞,比较虫荧光素酶活性。RT-PCR和Real-time PCR结果显示,TTRAP在HepG2.2.15细胞中的表达量分别是其在HepG2细胞中表达量的44.9%和27.8%(P0.05)。TTRAP启动子虫荧光素酶报告质粒与HBV表达质粒共转染组的相对荧光素酶活性,与对照组相比下降了43.8%。转染HBx表达质粒组的相对荧光素酶活性与其对照组相比下降了35%,而转染HBc、HBs及HBp表达质粒组对相对荧光素酶活性没有影响。因此证实HBx蛋白能抑制TTRAP启动子活性。  相似文献
