Electron crystallography of membrane proteins determines the structure of membrane-reconstituted and two-dimensionally (2D) crystallized membrane proteins by low-dose imaging with the transmission electron microscope, and computer image processing. We have previously presented the software system 2dx, for user-friendly image processing of 2D crystal images. Its central component 2dx_image is based on the MRC program suite, and allows the optionally fully automatic processing of one 2D crystal image. We present here the program 2dx_merge, which assists the user in the management of a 2D crystal image processing project, and facilitates the merging of the data from multiple images. The merged dataset can be used as a reference to re-process all images, which usually improves the resolution of the final reconstruction. The threespine stickleback Gasterosteus aculeatus is known to include several morphologically and ecologically divergent forms. Its phenotypic traits related to feeding vary among forms, and are considered to be a result of adaptations to various environments to find foods effectively. To examine whether the diversification of feeding modes in the stickleback involves genetic changes of the sense of taste, taste receptor family 1 (T1R) genes in stickleback were analyzed and compared with those in other model fishes. Ten T1R genes and 2 pseudogenes were identified from the stickleback genomic sequences. In particular, putative sweet taste receptors (T1R2s) highly increased in number in stickleback (8 genes and 2 pseudogenes) compared to other fishes (2-3 genes). Maximum likelihood estimations of nonsynonymous-synonymous nucleotide substitution rate have indicated that stickleback T1R2 are under positive selection. Expression analysis by RT-PCR revealed that most stickleback T1R genes were expressed in the taste organs; however, at least two T1R2 genes were not expressed in the taste organs, suggesting that the expression levels of these T1R2 genes may be fluctuated through the life history. In addition, sequencing analysis showed that several T1R2 genes in an anadromous form stickleback individual collected from the western Pacific (Japan) were substantially different from those in genomic data derived from a freshwater form individual collected in North America. This suggested that intra-specific variations of stickleback T1R2 genes were considerably large. Our results imply that, in stickleback, T1R2s have diversified through adaptation to various environments, probably to perceive substances important for its survival and reproduction. 相似文献

Riparian areas contain structurally diverse habitats that are challenging to monitor routinely and accurately over broad areas.
As the structural variability within riparian areas is often indiscernible using moderate-scale satellite imagery, new mapping
techniques are needed. We used high spatial resolution satellite imagery from the QuickBird satellite to map harvested and
intact forests in coastal British Columbia, Canada. We distinguished forest structural classes used in riparian restoration
planning, each with different restoration costs. To assess the accuracy of high spatial resolution imagery relative to coarser
imagery, we coarsened the pixel resolution of the image, repeated the classifications, and compared results. Accuracy assessments
produced individual class accuracies ranging from 70 to 90% for most classes; whilst accuracies obtained using coarser scale
imagery were lower. We also examined the implications of map error on riparian restoration budgets derived from our classified
maps. To do so, we modified the confusion matrix to create a cost error matrix quantifying costs associated with misclassification.
High spatial resolution satellite imagery can be useful for riparian mapping; however, errors in restoration budgets attributable
to misclassification error can be significant, even when using highly accurate maps. As the spatial resolution of imagery
increases, it will be used more routinely in ecosystem ecology. Thus, our ability to evaluate map accuracy in practical, meaningful
ways must develop further. The cost error matrix is one method that can be adapted for conservation and planning decisions

应用2001年8月和2010年8月Landsat TM/ETM+数据,计算沈阳市区及三环内各区的地表热岛强度(SUHI),根据土地利用数据和热岛源汇特征提取源汇信息,分析SUHI与土地利用类型、热岛源汇面积及边界长度、归一化植被指数(NDVI)、归一化建筑指数(NDBI)、改进的归一化差异水体指数(MNDWI)的关系。结果表明:2001年沈阳市区的中等热岛强度以上区域主要集中在三环内和苏家屯区,强热岛地区主要出现在铁西区和皇姑区;2010年中等以上热岛强度地区,与城区发展相一致,主要向西南,南面扩展,弱热岛、中等强度和强热岛地区有较大幅度的增加;城市热岛强度与NDVI存在显著的负相关,与NDBI存在显著的正相关;热岛源在区域内的面积比重与中等强度以上热岛存在较显著的相关关系,源汇边界长度与弱热岛和中等热岛存在较显著的相关关系。 相似文献

Nilaparvata lugens, the brown planthopper (BPH) feeds on rice phloem sap, containing high amounts of sucrose as a carbon source. Nutrients such as sugars in the digestive tract are incorporated into the body cavity via transporters with substrate selectivity. Eighteen sugar transporter genes of BPH (Nlst) were reported and three transporters have been functionally characterized. However, individual characteristics of NlST members associated with sugar transport remain poorly understood. Comparative gene expression analyses using oligo-microarray and quantitative RT-PCR revealed that the sugar transporter gene Nlst16 was markedly up-regulated during BPH feeding. Expression of Nlst16 was induced 2 h after BPH feeding on rice plants. Nlst16, mainly expressed in the midgut, appears to be involved in carbohydrate incorporation from the gut cavity into the hemolymph. Nlst1 (NlHT1), the most highly expressed sugar transporter gene in the midgut was not up-regulated during BPH feeding. The biochemical function of NlST16 was shown as facilitative glucose transport along gradients. Glucose uptake activity by NlST16 was higher than that of NlST1 in the Xenopus oocyte expression system. At least two NlST members are responsible for glucose uptake in the BPH midgut, suggesting that the midgut of BPH is equipped with various types of transporters having diversified manner for sugar uptake. 相似文献
