Modern chemotherapy has significantly improved patient outcomes against drug-sensitive tuberculosis. However, the rapid emergence of drug-resistant tuberculosis, together with the bacterium’s ability to persist and remain latent present a major public health challenge. To overcome this problem, research into novel anti-tuberculosis targets and drug candidates is thus of paramount importance. This review article provides an overview of tuberculosis highlighting the recent advances and tools that are employed in the field of anti-tuberculosis drug discovery. The predominant focus is on anti-tuberculosis agents that are currently in the pipeline, i.e. clinical trials.  相似文献

In this study, the thermoluminescence (TL) characteristics of Ag‐doped and undoped lithium tetraborate (Li2B4O7, LTB) materials, grown using the Czochralski method, were reported. The TL properties of LTB:Ag, such as glow curve structure, dose response, fading and reproducibility, were investigated. The glow curve of the Li2B4O7:Ag single crystal consists of four peaks located at approximately 75, 130, 190 and 275°C; in undoped LTB, the single crystal shows a broad glow curve with peaks at 65, 90, 125, 160 and 190°C using a heating rate of 5°C/s in the 50–350°C temperature region. The high temperature peak of Ag‐doped sample at 275°C has a nonlinear dose response within the range from 33 mGy to 9 Gy. There is a linear response in the range of 33–800 mGy; after which, a sublinear region appears up to 9 Gy for Ag‐doped LTB single crystal. For undoped single crystal, the dose response is supralinear for low doses and linear for the region between 1 and 9 Gy. The thermal fading ratio of the undoped material is almost 60% for the high temperature peak after 7 days. Ag‐doped LTB single crystal exhibits different behaviour over a period of 7 days.  相似文献

TL1A, a tumor necrosis factor-like cytokine, is a ligand for the death domain receptor DR3. TL1A, upon binding to DR3, can stimulate lymphocytes and trigger secretion of proinflammatory cytokines. Therefore, blockade of TL1A/DR3 interaction may be a potential therapeutic strategy for autoimmune and inflammatory diseases. Recently, the anti-TL1A monoclonal antibody 1 (mAb1) with a strong potency in blocking the TL1A/DR3 interaction was identified. Here, we report on the use of hydrogen/deuterium exchange mass spectrometry (HDX-MS) to obtain molecular-level details of mAb1′s binding epitope on TL1A. HDX coupled with electron-transfer dissociation MS provided residue-level epitope information. The HDX dataset, in combination with solvent accessible surface area (SASA) analysis and computational modeling, revealed a discontinuous epitope within the predicted interaction interface of TL1A and DR3. The epitope regions span a distance within the approximate size of the variable domains of mAb1′s heavy and light chains, indicating it uses a unique mechanism of action to block the TL1A/DR3 interaction.  相似文献

A comparison of the chemical composition and physical states of chloroplast lipids, of atrazine-resistant (R) and sensitive (S) biotypes of Conyza canadensis L. (horseweed), in the rosetta stage showed: (1) the R biotype contains lower amounts of polar lipids in its thylakoids, as expressed on a chlorophyll basis, than the S biotype. (2) The chloroplasts of the R biotype have higher contents of monogalactosyl diacylglycerol (MGDG) and lower contents of digalactosyl diacylglycerol (DGDG) and phosphatidylglycerol (PG), than those of the S biotype. (3) The chloroplast total lipids exhibit a higher degree of unsaturation in the R biotype. This is due to a higher level of linolenic acid, and a lower level of palmitic acid in the glycolipids. The fatty acid compositions of the phospholipids, except that of PG, do not differ significantly. (4) The lipid matrix of the thylakoid membranes of the R biotype is more fluid than that of the S biotype, as measured by the fluorescence polarization technique. The results are discussed in terms of whether these differences are responsible for the herbicide resistance.  相似文献

We present a new phosphor material, BaAlBO3F2 doped with Eu2+ ions, having emission in the UVA region. The phosphor material is prepared by a simple wet chemical method. Phase confirmation was carried out using the Rietveld refinement program which shows that BaAlBO3F2:Eu2+ has an hexagonal crystal system. Using a Fourier transform infrared spectroscopy graph, we studied the bond stretching present in the phosphor material. Photoluminescence (PL) characterization, carried out using a RF spectrofluorophotometer, shows two types of PL excitation and emission. Before reduction, emission is in the blue region at 431 nm; after reduction, excitation is at 258 nm and emission is at 361 nm, which is in the UVA region. Some thermoluminescence (TL) studies were carried out in this material for the first time, for example, determination of the trapping parameters, linearity, fading, glow curve convolution and deconvolution (GCCD) function for curve fitting and the Tm–Tstop method for confirmation of the trapped centers in the TL glow peak. Cytochrome b559 is an essential component of the photosystem II reaction center in photosynthetic oxygen-evolving organisms, but its function still remains unclear. The use of photosystem II preparations from Thermosynechococcus elongatus of high integrity and activity allowed us to measure for the first time the influence of cytochrome b559 mutations on its midpoint redox potential and on the reduction of the cytochrome b559 by the plastoquinone pool (or QB). In this work, five mutants having a mutation in the α-subunit (I14A, I14S, R18S, I27A and I27T) and one in the β-subunit (F32Y) of cytochrome b559 have been investigated. All the mutations led to a destabilization of the high potential form of the cytochrome b559. The midpoint redox potential of the high potential form was significantly altered in the αR18S and αI27T mutant strains. The αR18S strain also showed a high sensitivity to photoinhibitory illumination and an altered oxidase activity. This was suggested by measurements of light induced oxidation and dark re-reduction of the cytochrome b559 showing that under conditions of a non-functional water oxidation system, once the cytochrome is oxidized by P680+, the yield of its reduction by QB or the PQ pool was smaller and the kinetic slower in the αR18S mutant than in the wild-type strain. Thus, the extremely positive redox potential of the high potential form of cytochrome b559 could be necessary to ensure efficient oxidation of the PQ pool and to function as an electron reservoir replacing the water oxidation system when it is not operating.  相似文献

TL1A, a TNF member implicated in autoimmune diseases, is a transmembrane protein that is processed to release soluble TL1A (TL1A-S). TL1A-S induces a Th1 response, although the functional significance of membrane-bound TL1A (TL1A-M) remains unknown. We generated TL1A-M expression in HEK-293 cells capable of binding DR3-Fc. Co-incubating IL-12/IL-18-primed CD4+ T cells with HEK-293 cells expressing TL1A-M induced 3-fold increase in IFN-γ that was blocked by anti-TL1A Ab. These results demonstrate that TL1A-M can bind death domain receptor 3 (DR3) through cell-cell contact to induce downstream IFN-γ secretion enhancement. Anti-TL1A antibodies designed to treat immune diseases should be verified to block both endogenous TL1A forms.  相似文献

The major light-harvesting chlorophyll a/b complex (LHCIIb) of photosystem (PS) II functions by harvesting light energy and by limiting and balancing the energy flow directed towards the PSI and PSII reaction centers. The complex is predominantly trimeric; however, the monomeric form may play a role in one or several of the regulatory functions of LHCIIb. In this work the dissociation temperature was measured of trimeric LHCIIb isolated from Pisum thylakoids and inserted into liposomes made of various combinations of thylakoid lipids at various protein densities. Dissociation was measured by monitoring a trimer-specific circular dichroism signal in the visible range. The LHCIIb density in the membrane significantly affected the trimer dissociation temperature ranging from 70 °C at an LHCIIb concentration comparable to or higher than the one in thylakoid grana, to 65 °C at the density estimated in stromal lamellae. Omitting one thylakoid lipid from the liposomes had virtually no effect on the thermal trimer stability in most cases except when digalactosyl diacylglycerol (DGDG) was omitted which caused a drop in the apparent dissociation temperature by 2 °C. In liposomes containing only one lipid species, DGDG and, even more so, monogalactosyl diacylglycerol (MGDG) increased the thermal stability of LHCIIb trimers whereas phosphatidyl diacylglycerol (PG) significantly decreased it. The lateral pressure exerted by the non-bilayer lipid MGDG did not significantly influence LHCII trimer stability.  相似文献
