Depth and flipper movements were simultaneously measured during 23 dives for a free-swimming green turtle (Chelonia mydas) at Ascension Island. A few characteristic dive profiles that have been widely reported in hard-shelled turtles were recorded. Flipper movements revealed that, on dives to midwater, there was generally active swimming, compared to long periods of inactivity on dives to the seafloor. During all dives, there were clear changes in the flipper beat frequency during the descent. On leaving the surface, flippers beats occurred quickly (typically 30-40 beats min−1) and then as the descent continued the frequency declined (typically to about 10-14 beats min−1). These observations match the general pattern reported for other air-breathing divers for increased effort at the start of the descent to overcome initial positive buoyancy.  相似文献

Five different methods for measuring transpiration, which include gravimetric analysis (control), heat pulse velocity (HPV), time domain reflectometry (TDR), single leaf and whole plant infrared gas-exchange measurements, have been tested on two cultivars (Redcort and Empire) of young apple trees (Malus communis L.). The objective was to compare these methods and establish the most affordable one to be used in greenhouse conditions in order to determine and/or estimate the amount of water for an efficient irrigation management. Results obtained with TDR were particularly accurate and not statistically different with respect to the control (–4.2 %) and this was supported by the correlation coefficient (r = 0.94) found. The HPV method was sufficiently accurate and reliable for small stems, however, in our conditions this method generally underestimated the transpiration (–32.4 %). Single leaf and particularly whole plant infrared gas-exchange measurements suffered an overestimation of the transpiration with respect to the control.  相似文献

Water deficit and salt accumulation in soil presents serious problems to crop production in semi-arid regions. These problems depend on the active transpiration stream and the selective absorption of ions by crop roots. In this study, a large sized soil column system was used to examine the dynamics of water and ion transport and salt accumulation in soil layers. Special reference was placed on the effects of the active and selective absorption by roots of different crops (i.e., corn plants, sunflower plants and no plants). The column system was equipped with on-line systems for the control of groundwater level. Soil water content sensors enabled time-course evaluations of the volumetric water content and hence upward flux of the groundwater in the soils at different depths. Furthermore, the distribution and accumulation of ions in soil layers, plant organs and xylem sap were analyzed using ion chromatography. In this column experiment, diurnal and longer term changes in water movement and ion accumulation in soil, affected by root absorption characteristics of plants, were evaluated quantitatively. The results demonstrated that the column system was applicable for the quantitative analysis of the effects of root absorption by different crops on water deficit and salinization in soils.  相似文献

Water is a key resource in commercial wine production and both large excesses and deficits have undesirable effects upon the amount and quality of the wine produced. A balance between the water requirements of a fully developed canopy and the induced stress necessary for the commercial quality of the wine must be reached. Thus we need a physiological indicator that integrates both soil and climatic conditions to use as a management tool. An experimental field was established in the eastern part of the Demarcated Region of Douro – Portugal, to study the effect of water supply on the quality of the musts produced and we need a physiological indicator that relates to the water use and stress of the grapevines (Vitis vinifera L.) and to the later evaluation of the effect of irrigation practices upon the quality of the musts. We chose as indicators sap flow, leaf water potential at pre-dawn (0600 h), mid-morning (1000 h), solar noon (1400 h) and sunset (1900 h), stomatal conductance and leaf transpiration both measured at mid-morning and at solar noon, and related them to our experimental treatments that induce differences in soil water content, evaluated with time-domain reflectometry probes, with the objective of selecting the indicator that best describes the plant water status under different amounts of available water. Sap flow, leaf water potential and leaf transpiration rate measured at solar noon had highly significant correlations with soil water content and their regression on soil water content was also highly significant. Each of these parameters has shortcomings and none has a clear advantage over the other two as an integrator of the environmental conditions under these experimental conditions. Further studies of the parameters and their relationship with the quality characteristics of the produced musts are needed to achieve the ultimate objective of manipulating the soil water content.  相似文献

Knowledge of the diving behaviour of aquatic animals expanded considerably with the invention of time-depth recorders (TDRs) in the 1960s. The large volume of data acquired from TDRs can be analyzed using dive analysis software, however, the application of the software has received relatively little attention. We present an empirical procedure to select optimum values that are critical to obtaining reliable results: the zero-offset correction (ZOC) and the dive threshold. We used dive data from shallow-diving coastal dugongs (Dugong dugon) and visual observations from an independent study to develop and test a procedure that minimizes errors in characterizing dives. We initially corrected the surface level using custom software. We then determined the optimum values for each parameter by classifying dives identified by an open-source dive analysis software into Plausible and Implausible dives based on the duration of dives. The Plausible dives were further classified as Unrecognized dives if they were not identified by the software but were of realistic dive duration. The comparison of these dive types indicated that a ZOC of 1 m and a dive threshold of 0.75 m were the optimum values for our dugong data as they gave the largest number of Plausible dives and smaller numbers of other dive types. Frequency distributions of dive durations from TDRs and independent visual observations supported the selection. Our procedure could be applied to other shallow-diving animals such as coastal dolphins and turtles.  相似文献

A potential relationship between transposon-derived repeats (TDR) and human germline methylation is of biological importance since many genes are flanked by TDR and methylation could affect the expression of nearby genes. Furthermore, DNA methylation has been suggested as a global defense mechanism against genome instability threatened by TDR. We studied the correlation between the density of HapMap methyl-associated SNPs (mSNPs), a marker of germline methylation, and proportion of TDR.After correcting for confounding variables, we found a negative correlation between proportion of Alu repeats and mSNP density for 125-1000 kb windows. Similar results were found for the most active subgroup of repeats. In contrast, a negative correlation between proportion of L1 repeats and mSNP density was found only in the larger 1000 kb windows.Using methylation data on germ cells (sperm) from the Human Epigenome Project, we found a lower proportion of Alu repeats adjacent (3-15 kb) to hypermethylated amplicons. On the contrary, there was a higher proportion of L1 repeats in the 3-5 kb of sequence flanking hypermethylated amplicons but not in the 10-15 kb flanks.Our data indicate a differential response to the major repeat families and that DNA methylation is unlikely to be a uniform global defense system against all TDR. It appears to play a role for the L1 subgroup, with sequences adjacent to L1 repeats methylated in response to their proximity. In contrast, sequences adjacent to Alu repeats appear to be hypomethylated, arguing against a role of methylation in germline defense against those elements.  相似文献

Soil moisture and suction head in an irrigated orchard were continuously monitored by time domain reflectometry (TDR) probes and gypsum blocks, respectively, during and between successive irrigation events. On each side of the trees in the plot, two 30-cm long probes were installed vertically 10 cm below the soil surface (denoted as shallow) and another two probes were installed vertically 40 cm below the soil surface (denoted as deep). The variation in moisture content measured by the TDR probes between successive irrigation events was qualitatively divided into four stages: the first was during water application; the second initiated when irrigation stopped and the moisture content in the layer sharply decreased, mainly due to free drainage. The succeeding moderate soil-moisture decrease, caused by the simultaneous diminishing free drainage and root uptake, was the third stage. During the fourth stage, moisture depletion from the layer was solely by root uptake. The slopes of moisture content variation with time throughout this stage enabled the monitoring of water availability to the plant. The range of moisture content variations and moisture depletion rates between subsequent irrigation events was higher in the shallow (10–40 cm) than in the deeper (40–70 cm) layer. Irrigation nonuniformity and spatial variability of soil hydraulic properties contributed to the unevenness of the moisture distribution in the soil profile. However, as soon as moisture content within a layer reached field capacity, namely the free drainage had stopped, irrigation uniformity had a negligible effect on water flux to the plant roots. The measured data indicate that soil moisture is fully available to the plant as long as the momentary moisture flux from the soil bulk to the soil–root interface can replenish the moisture being depleted to supply, under non-stressed conditions, the atmospheric water demand. This flux is dominated by the local momentary value of the soil's bulk hydraulic conductivity, K r, and it stays constant for a certain range of K r values, controlled by the increasing root suction. A decrease in water availability to the plant appears for longer irrigation intervals as a break in the constant soil-moisture depletion rate during stage 4. This break is better correlated to a threshold K r value than to threshold values of moisture content or suction. Therefore, it is suggested that moisture content or suction used to measure water availability or to control irrigation first be alibrated by K r() or K r() curves, respectively.  相似文献

Background

Trypanosomatids are early-diverging eukaryotes devoid of the major disulfide reductases – glutathione reductase and thioredoxin reductase – that control thiol-redox homeostasis in most organisms. These protozoans have evolved a unique thiol-redox system centered on trypanothione, a bis-glutathionyl conjugate of spermidine. Notably, the trypanothione system is capable to sustain several cellular functions mediated by thiol-dependent (redox) processes.

Scope of review

This review provides a summary of some historical and evolutionary aspects related to the discovery and appearance of trypanothione in trypanosomatids. It also addresses trypanothione's biosynthesis, physicochemical properties and reactivity towards biologically-relevant oxidants as well as its participation as a cofactor for metal binding. In addition, the role of the second most abundant thiol of trypanosomatids, glutathione, is revisited in light of the putative glutathione-dependent activities identified in these organisms.

Major conclusions

Based on biochemical and genome data, the occurrence of a thiol-redox system that is strictly dependent on trypanothione appears to be a feature unique to the order Kinetoplastida. The properties of trypanothione, a dithiol, are the basis for its unique reactivity towards a wide diversity of oxidized and/or electrophilic moieties in proteins and low molecular weight compounds from endogenous or exogenous sources. Novel functions have emerged for trypanothione as a potential cofactor in iron metabolism.

General significance

The minimalist thiol-redox system, developed by trypanosomatids, is an example of metabolic fitness driven by the remarkable physicochemical properties of a glutathione derivative. From a pharmacological point of view, such specialization is the Achilles' heel of these ancient and deadly parasites.
