Pentachlorophenol (PCP) was a commonly used fungicide, herbicide, insecticide, and bactericide in industrial, agricultural, and domestic settings; however, it was also contaminated with polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs). It has been reported that technical grade PCP had immunosuppressive effects and that the immune system was the major target of PCDD/PCDFs toxicity. Although the immune response after exposure to PCP or 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) has been studied, the toxic effects of exposure to both PCP and TCDD have not yet been reported. The aim of this study was to evaluate the effects on immune cells from mice intraperitoneally immunized with OVA and subsequently treated with PCP or TCDD alone or in combination by gavage. The animals were terminated on day 7 and 14, and the spleen and plasma samples were collected for immunotoxicity evaluation. The numbers and populations of splenocytes, T cell-derived cytokines produced by splenocytes, splenocyte-generated cytotoxicity and OVA-specific antibodies in plasma were investigated. Our results indicate that the spleen/body weight ratio and splenocyte number was reduced by TCDD alone; in addition, this reduction was enhanced when TCDD was combined with PCP. Exposure to TCDD alone or in conjunction with PCP suppressed many ovalbumin (OVA)-stimulated cytokines, including IL-2, IFN-γ, IL-4, IL-5, and IL-10. Furthermore, the immunoglobulins IgG and IgM were suppressed in mice administered by PCP alone, but the suppressive effects were greater in mice treated with TCDD alone or in combination with PCP. Co-exposure to PCP and TCDD resulted in an antagonistic effect on TCDD-induced suppression of IFN-γ and IL-10. Our results demonstrate that PCP alone is immunotoxic, regardless of the presence of TCDD. PCP led to mild changes in cytokine secretion, and it compromised splenocyte-generated cytotoxicity and IgM and IgG antibody production on day 7. The finding that PCP antagonizes TCDD-induced IFN-γ suppression could be due to the competitive binding of PCP to AhR (aryl hydrocarbon receptor).  相似文献

MicroRNA319 (miR319) family is one of the conserved microRNA (miRNA) families among diverse plant species. It has been reported that miR319 regulates plant development in dicotyledons, but little is known at present about its functions in monocotyledons. In rice (Oryza sativa L.), the MIR319 gene family comprises two members, OsaMIR319a and OsaMIR319b. Here, we report an expression pattern analysis and a functional characterization of the two OsaMIR319 genes in rice. We found that overexpressing OsaMIR319a and OsaMIR319b in rice both resulted in wider leaf blades. Leaves of osa‐miR319 overexpression transgenic plants showed an increased number of longitudinal small veins, which probably accounted for the increased leaf blade width. In addition, we observed that overexpressing osa‐miR319 led to enhanced cold tolerance (4 °C) after chilling acclimation (12 °C) in transgenic rice seedlings. Notably, under both 4 and 12 °C low temperatures, OsaMIR319a and OsaMIR319b were down‐regulated while the expression of miR319‐targeted genes was induced. Furthermore, genetically down‐regulating the expression of either of the two miR319‐targeted genes, OsPCF5 and OsPCF8, in RNA interference (RNAi) plants also resulted in enhanced cold tolerance after chilling acclimation. Our findings in this study demonstrate that miR319 plays important roles in leaf morphogenesis and cold tolerance in rice.  相似文献

The anion–π interactions between Br, Cl, F and H anions and hexafluorobenzene (HFB), 1,2,4,5-tetracyanobenzene (TCB) and tetracyanopyrazine (TCP) have been studied by standard and counterpoise (CP) corrected methods at HF, B3LYP and MP2/6-31+ + G (d,p) levels of theory. The complexation energies were corrected for basis set superposition error (ΔE BSSE) and zero point energy (ΔE BSSE + ZPE). Also, the B3LYP results were corrected by single-point calculation at B3LYP/aug-cc-PVTZ level of theory. Although the CP-corrected method results in higher distances between anions and rings, the standard method gives lower complexation energies. TCP…X series gives higher complexation energies in both CP-corrected and standard methods. Topological analysis of the charge density ρ(r) has been performed by the means of atoms in molecules method on the wave functions obtained at MP2/6-31+ + G (d,p) level of theory. The number and the nature of critical points depend on aromatic ring and anion. Natural bond orbital analysis indicates that nX → π*CC and nX → π*CN interactions are the most important interactions for TCB (and HFB)…X and TCP…X complexes, respectively.  相似文献

The impact of a rectal spacer and an increased near maximum target dose in VMAT prostate SBRT is studied.For a group of 11 patients (35 Gy-in-five-fractions VMAT prostate SBRT) a set of 4 plans were generated, namely two VMAT plans, with D2%  37.5 Gy (Hom) and with D2%  40.2 Gy (Het), were created for each of two CT scans taken before (NoSpc) and after (Spc) transperineal spacer insertion. Consequently the methodology for parameter invariant TCP (tumor control probability) plan ranking was applied for comparison of the plans in terms of tumor control. NTCPs (normal tissue complication probabilities) were calculated for rectum and bladder using Lyman’s model.For all 11 patients the TCP plan ranking has shown that the Het plans would perform considerably better in TCP terms than the Hom ones. The plans without rectal spacer were ranked worse compared to those with rectal spacer except for one set of Hom plans. The calculated NTCPs for rectum produced by the Het plans were quite similar to the NTCPs of the Hom ones. The rectal NTCPs of the Hom Spc plans were always lower than the NTCPs of the Hom NoSpc plans. The NTCP values for bladder were extremely low in all cases.The use of rectal spacer leads in general to lower risk of rectal complications, as expected, and even to better tumor control. Plans with increased near maximum target dose (D2%  40.2 Gy) are expected to perform much better in terms of tumor control than those with D2%  37.5 Gy.  相似文献

FPP and adenosine modulate the adenylyl cyclase (AC)/cAMP signal transduction pathway in mammalian spermatozoa to elicit a biphasic response, initially stimulating capacitation and then inhibiting spontaneous acrosome loss. This study addressed the hypothesis that responses to FPP involve interactions between receptors for FPP and adenosine, the biphasic responses involving stimulatory and inhibitory adenosine receptors. Gln‐FPP, a competitive inhibitor of FPP, significantly inhibited binding of an adenosine analogue and responses to adenosine, especially in capacitated suspensions, consistent with interaction between FPP and adenosine receptors. CGS‐21680 (1 μM), a stimulatory A2a adenosine receptor agonist, significantly stimulated capacitation and cAMP in uncapacitated cells, while cyclopentyl adenosine (1 μM), an inhibitory A1 adenosine receptor agonist only affected capacitated cells, inhibiting spontaneous acrosome loss. Responses to FPP and adenosine were inhibited in uncapacitated cells by a selective A2a antagonist and in capacitated cells by a selective A1 antagonist; subsequent investigations indicated possible involvement of G proteins. Like FPP, cholera toxin stimulated capacitation and cAMP production in uncapacitated cells, suggesting involvement of a G protein with a Gαs subunit. In contrast, pertussis toxin prevented FPP's inhibition of both spontaneous acrosome loss and cAMP production, suggesting involvement of a Gαi/o subunit. Immunoblotting evidence revealed the presence of proteins of the appropriate molecular weights for Gαs, Gαi2, i3, and Gαo subunits. This study provides the first direct evidence suggesting the involvement of two different types of adenosine receptors and both Gαs and Gαi/o subunits in the regulation of capacitation, resulting in modulation of AC activity and availability of cAMP. 为明确TCP转录因子家族在景宁木兰(Magnolia sinostellata)遮阴胁迫中的作用,通过模拟自然生境对景宁木兰3 a生嫁接苗进行遮阴处理,基于转录组分析TCP转录因子相关信息,利用转录组表达量和qRT-PCR分析遮阴胁迫下的表达模式。结果表明,从景宁木兰转录组中筛选出12个TCPs基因,编码蛋白长度为228~558个氨基酸,分子量为23.7~52.7 kDa,TCPs蛋白均为不稳定蛋白、亲水性蛋白;TCPs均定位于细胞核,MsTCP20-a/b蛋白还定位于叶绿体。TCPs成员分为2大类,Class Ⅰ包含10个TCPs成员,Class Ⅱ包含2个TCPs成员。所有成员均含有TCP保守结构域,亲缘关系较近的成员具有相似的保守基序;克隆得到TCPs启动子序列,其序列含有大量光响应和激素响应元件。在遮阴处理后景宁木兰TCPs基因存在差异表达,其中MsTCP9-aMsTCP2在遮阴后显著上升,MsTCP8MsTCP9-bMsTCP7-aMsTCP15在遮阴后显著下降。因此,景宁木兰TCPs可能在响应遮阴胁迫过程中发挥重要功能。  相似文献
