Summary Following fusion of protoplasts from crown gall tumour calli, characterized by hormone independent growth, and protoplasts from normal tissues of a streptomycin-resistant mutant, SR1, we selected hormone independent streptomycin-resistant calli in Nicotiana tabacum. The tumour line, B6S3, lost the ability to form shoots. Some of the selected lines, similar to SR1, however, are morphogenic. Both calli and shoots contained the tumour specific enzyme lysopinedehydrogenase. The hybrid shoots are resistant to Agrobacterium infection and do not root. These tumorous properties are dominantly expressed in the somatic hybrids.  相似文献

Three chrysanthemum (Dendranthema grandiflora) cultivars were cocultivated with 2 Agrobacterium tumefaciens strains in combination with 4 pBIN19 derived binary plasmids, all carrying the Nosnptll selection gene and 35Sgus(intron) reporter gene. All binary plasmids transferred DNA to chrysanthemum explants but only pMOG410 gave good stable expression of GUS. This plasmid differs from the other plasmids in 2 aspects: 1) It carries a restored nptll gene and 2) the selection gene is positioned at the left border side of the reporter gene. Cocultivation with AGLO(pMOG410) yielded up to 13 GUS positive shoots per 100 explants. The presence of the gus and nptll gene in recovered shoots was confirmed by PCR and Southern blot analysis.  相似文献

The morphological integration of the hind wings of the western corn rootworm Diabrotica virgifera virgifera LeConte was investigated to get a better insight of the undergone by this invasive species. Geometric morphometric methods were used to test two modularity hypotheses associated with the wing development and function (hypothesis H1: anterior/posterior or H2: distal/proximal wing parts). Both hypotheses were rejected and the results showed the integrated behavior of the hind wings of D. v. virgifera. The hypothesized modules do not represent separate units of variation, so in a similar fashion as exhibited by the model species Drosophila melanogaster, the hind wings of D. v. virgifera act as a single functional unit. The moderate covariation strength found between anterior and posterior and distal and proximal parts of the hind wing of D. v. virgifera confirms its integrated behavior. We conclude that the wing shape shows internal integration, which could enable flexibility and thus enhance flight maneuverability. This study contributes to the understanding of morphological integration and modularity on a non-model organism. Additionally, these findings lay the groundwork for future flight performance and biogeographical studies on how wing shape and size vary across the endemic and expanded/invaded range in the USA and Europe infested with D. v. virgifera.  相似文献

Computational modeling of biological networks permits the comprehensive analysis of cells and tissues to define molecular phenotypes and novel hypotheses. Although a large number of software tools have been developed, the versatility of these tools is limited by mathematical complexities that prevent their broad adoption and effective use by molecular biologists. This study clarifies the basic aspects of molecular modeling, how to convert data into useful input, as well as the number of time points and molecular parameters that should be considered for molecular regulatory models with both explanatory and predictive potential. We illustrate the necessary experimental preconditions for converting data into a computational model of network dynamics. This model requires neither a thorough background in mathematics nor precise data on intracellular concentrations, binding affinities or reaction kinetics. Finally, we show how an interactive model of crosstalk between signal transduction pathways in primary human articular chondrocytes allows insight into processes that regulate gene expression.  相似文献

How the brain combines information from different sensory modalities and of differing reliability is an important and still-unanswered question. Using the head direction (HD) system as a model, we explored the resolution of conflicts between landmarks and background cues. Sensory cue integration models predict averaging of the two cues, whereas attractor models predict capture of the signal by the dominant cue. We found that a visual landmark mostly captured the HD signal at low conflicts: however, there was an increasing propensity for the cells to integrate the cues thereafter. A large conflict presented to naive rats resulted in greater visual cue capture (less integration) than in experienced rats, revealing an effect of experience. We propose that weighted cue integration in HD cells arises from dynamic plasticity of the feed-forward inputs to the network, causing within-trial spatial redistribution of the visual inputs onto the ring. This suggests that an attractor network can implement decision processes about cue reliability using simple architecture and learning rules, thus providing a potential neural substrate for weighted cue integration.  相似文献

Although most studies on integration and modularity have focused on variation among individuals within populations or species, this is not the only level of variation for which integration and modularity exist. Multiple levels of biological variation originate from distinct sources: genetic variation, phenotypic plasticity resulting from environmental heterogeneity, fluctuating asymmetry from random developmental variation and, at the interpopulation or interspecific levels, evolutionary change. The processes that produce variation at all these levels can impart integration or modularity on the covariance structure among morphological traits. In turn, studies of the patterns of integration and modularity can inform about the underlying processes. In particular, the methods of geometric morphometrics offer many advantages for such studies because they can characterize the patterns of morphological variation in great detail and maintain the anatomical context of the structures under study. This paper reviews biological concepts and analytical methods for characterizing patterns of variation and for comparing across levels. Because research comparing patterns across level has only just begun, there are relatively few results, generalizations are difficult and many biological and statistical questions remain unanswered. Nevertheless, it is clear that research using this approach can take advantage of an abundance of new possibilities that are so far largely unexplored.  相似文献

Flowers of animal-pollinated plants are integrated structures shaped by the action of pollinator-mediated selection. It is widely assumed that pollination specialization increases the magnitude of floral integration. However, empirical evidence is still inconclusive. In this study, we explored the role of pollinator diversity in shaping the evolution of corolla-shape integration in Erysimum, a plant genus with generalized pollination systems. We quantified floral integration in Erysimum using geometric morphometrics and explored its evolution using phylogenetic comparative methods. Corolla-shape integration was low but significantly different from zero in all study species. Spatial autocorrelation and phylogenetic signal in corolla-shape integration were not detected. In addition, integration in Erysimum seems to have evolved in a way that is consistent with Brownian motion, but with frequent convergent evolution. Corolla-shape integration was negatively associated with the number of pollinators visiting the flowers of each Erysimum species. That is, it was lower in those species having a more generalized pollination system. This negative association may occur because the co-occurrence of many pollinators imposes conflicting selection and cancels out any consistent selection on specific floral traits, preventing the evolution of highly integrated flowers.  相似文献

生物膜研究是现代生物学研究的前沿方向之一,本文对文革结束以后生物膜研究在中国科学院生物物理研究所(下称生物物理所)的兴起与发展进行了系统回顾.文革结束后,中国科学院领导了解到国外生物膜研究迅速发展的情况,迅即派遣以生物物理所杨福愉为组长,包括生物化学研究所、动物研究所、植物研究所、上海实验生物研究所等六人的代表团前往联邦德国进行考察.考察结束后院领导根据多学科交叉对生物膜研究的重要作用,组织了生物物理所、植物所、中国医学科学院、北京医科大学等单位联合申报国家自然科学基金委员会重大项目"膜脂-膜蛋白的相互作用及其在医学和农业上的应用",并获得立项.与此同时,院领导建议由中国生物物理学会、中国生化学会和中国细胞生物学会共同组织生物膜学术讨论会.首次会议于1979年3月在北京友谊宾馆举行,以后每3年召开1次,从未中断,有力地促进了生物膜研究的交流与发展.2003年举行的第200届香山会议专门组织讨论21世纪生物膜研究在中国的布局,进一步推动了生物膜研究的发展.本文还重点阐述了中国科学院生物物理研究所在生物膜研究方面所取得的代表性成果:a.金属离子通过膜脂-膜蛋白相互作用调控生物膜能量转换、物质运输及信号转导的分子机制;b.提出"克山病是一种心肌线粒体病"的重要观点;c.发现溶酶体内含有为量甚微、一般认为是消化酶的胰凝乳蛋白酶,并阐明了它通过溶酶体膜外泄后参与细胞凋亡的作用机制;d.确定了通过调控线粒体动态变化而干预肿瘤细胞迁移侵袭的新靶标.最后,特别值得一提的是,2004年,常文瑞与植物研究所匡廷云等在《自然》(Nature)发表了《菠菜中主要捕光色素复合体2.72魡分辨率的晶体结构分析》的研究论文,2005年饶子和与徐建兴等在《细胞》(Cell)发表了《猪心线粒体呼吸链复合体Ⅱ的晶体结构》的研究论文,充分标志着我国生物膜研究已在国际上占据一席之地.2004年,徐涛研究员因其在囊泡转运方面的丰厚学术积淀,作为首席科学家组织一批生物膜专家承担了国内首个生物膜973项目,这标志着国内生物膜研究开始进入一个新的发展时期.  相似文献
