To obtain adhesive and safe lactic acid bacteria (LAB) strains for expressing heterologous antigens, we screened LAB inhabitants in intestine of Tibetan chickens by analyzing their adhesion and safety properties and the selected LAB was engineered to express heterologous antigen (UTEpi C-A) based on chromosomal integration strategy. We demonstrated that a new Lactobacillu salivarius TCMM17 strain is strongly adhesive to chicken intestinal epithelial cells, contains no endogenous plasmids, is susceptible to tested antimicrobials, and shows no toxicities. In order to examine the potential of TCMM17 strain as heterogenous antigen delivering vehicle, we introduced a UTEpi C-A expression cassette in its chromosome by constructing a non-replicative plasmid (pORI280-UUTEpi C-AD). The recombinant TCMM17 strain (?TCMM17) stably was found to keep the gene cassette through 50 generations, and successfully displayed EpiC encoded by the cassette on its surface. This work provides a universal platform for development of novel oral vaccines and expression of further antigens of avian pathogens.  相似文献

Reactive separations using green catalysts offer great opportunities for manufacturing fatty esters, involved in specialty chemicals and biodiesel production. Integrating reaction and separation into one unit provides key benefits such as: simplified operation, no waste, reduced capital investment and low operating costs.This work presents a novel heat-integrated reactive absorption process that eliminates all conventional catalyst related operations, efficiently uses the raw materials and equipment, and considerably reduces the energy requirements for biodiesel production - 85% lower as compared to the base case. Rigorous simulations based on experimental results were carried out using Aspen Plus and Dynamics. Despite the high degree of integration, the process is well controllable using an efficient control structure proposed in this work. The main results are provided for a plant producing 10 ktpy fatty acid methyl esters from methanol and waste vegetable oil with high free fatty acids content, using sulfated zirconia as solid acid catalyst.  相似文献

We have examined the growth and gravitropic response of seedling roots of rapeseed ( Brassica napus . CrGC5–1) transformed by Agrobacterium rhizogenes A4, in order to evaluate if this could constitute a new model system for the study of gravitropism. The transformed clone chosen for study had integrated full-length TL- and TR-DNA from pRi (the root inducing plasmid), and thus included all of the agrobacterial genes potentially involved in the modified phenotype of transformed plants. In the vertical position, the growth rate of transformed roots was higher than controls. During 24 h of continuous stimulation, the optimal angle for gravitropic bending in normal roots was 135° (with respect to the gravity axis), with decreasing response at 90° and 45°. For transformed roots, slight curvature developed at 45° and at 90°, and stronger curvature was observed at 135°, though transformed roots tips never reached the vertical position. The minimum stimulation time necessary to elicit a response (presentation time) was also determined: it was signficantly shorter in normal roots (80 s) than in transformed ones (120 s). The results show that pRi transformed roots are less sensitive to gravity than normal roots.  相似文献

All natural environments are spatially and temporally heterogeneous. Consequently, their ability to provide essential resources for the growth of plants is variable. Modular plant species produce repeated basic structures which, in the case of clonal species, are called ramets. Ramets belonging to the same clone are distributed throughout the environment in space and time, and therefore they may be located in sites which differ in resource-providing quality. The connections between ramets may allow resources to be shared, enabling the clone to behave as a cooperative system. As a result of such physiological integration, ramets can survive in conditions where there is lethal shortage of a resource because they are connected to, and supported by, ramets located in conditions where there is ample supply of the same resource. Physiological integration between connected ramets presents opportunities for heterogeneous environments to be exploited to an extent that is only just becoming apparent. As heterogeneity is ubiquitous in natural environments, it may be expected that plants, as relatively immobile organisms, will have evolved the capacity to cope with it by making appropriate localized morphological and/or physiological plastic responses. Recent studies suggest that such responses not only enable clonal species to cope with environmental heterogeneity, but that under some circumstances they can benefit more from environments which are heterogeneous rather than homogeneous, even when both types of environment contain the same amount of resources. Studies on Glechoma hederacea (Lamiaceae) that illustrate this phenomenon are described.  相似文献

This article examines the promise and challenge of marine biotelemetry technologies, comparing the case of the Canadian Ocean Tracking Network (OTN) and the Australian Animal Tagging and Monitoring System (AATAMS). These technologies provide increased understanding of the marine environment and promise major advances in knowledge of species at risk. However, that utilization of marine biotelemetry raises challenges, including ensuring that knowledge obtained is translated into forms useful to and usable by decision makers. Knowledge brokering is a key to ensure that the advances seen in marine biotelemetry technologies lead to improved gover-nance of species at risk.  相似文献

Abstract Plasmid-curing studies suggest that acidocin B production is encoded by the 14-kb plasmid pCV461 in Lactobacillus acidophilus M46. Loss of pCV461 from the original producer strain M46 did not coincide with loss of immunity to acidocin B. Bacteriocin activity determination after SDS-PAGE showed that a substance of 2.4 kDa, absent in the culture supernatant of the mutant strain M46A2, lacking pCV461, represented acidocin B activity. In order to introduce a positive selection criterion, pCV461 was marked in vivo by the erythromycin resistance marker of pE194, present on pUC19 containing a 1.4-kb Hin dIII fragment of pCV461, after plasmid integration. Introduction of this recombinant plasmid into the mutant strain M46A2 or Lactobacillus plantarum resulted in erythromycin-resistant, acidocin B-producing transformants, showing unambiguously that acidocin B is encoded by pCV461.  相似文献

Summary Following fusion of protoplasts from crown gall tumour calli, characterized by hormone independent growth, and protoplasts from normal tissues of a streptomycin-resistant mutant, SR1, we selected hormone independent streptomycin-resistant calli in Nicotiana tabacum. The tumour line, B6S3, lost the ability to form shoots. Some of the selected lines, similar to SR1, however, are morphogenic. Both calli and shoots contained the tumour specific enzyme lysopinedehydrogenase. The hybrid shoots are resistant to Agrobacterium infection and do not root. These tumorous properties are dominantly expressed in the somatic hybrids.  相似文献
