Abstract: State wildlife agencies often use input obtained through public meetings to develop management policies. Because public meetings can be dominated by single stakeholder groups, these policies may not reflect the attitudes of new wildlife stakeholders. In 2000 the Utah Wildlife Board, after a series of public meetings, adopted a statewide policy for winter-feeding mule deer (Odocoileus hemionus). The policy was implemented by the Utah Division of Wildlife Resources from 2001 to 2007 in Cache County of northern Utah, USA. In 2007, we surveyed Utah households representing metropolitan, nonmetropolitan, and Cache County residents (n = 1,800) to evaluate whether the winter-feeding policy reflected the attitudes of all wildlife stakeholders. Survey respondents, regardless of residence strata, believed winterfeeding programs were essential for managing mule deer in Utah (χ26 = 7.02, P = 0.32). However, most respondents were reluctant to support feeding programs at the expense of habitat restoration projects (χ26 = 11.64, P = 0.07). Our results suggest that the winter-feeding policy represented the attitudes of the Utah residents surveyed, though few had participated in its development. Respondents' strong utilitarian attitudes toward wildlife (e.g., strong support for hunting and feeding) influenced those respondents' perceptions of the policy. Given the effects of increased urbanization on utilitarian attitudes toward wildlife in many parts of the United States, coupled with decreasing numbers of traditional wildlife stakeholders, state wildlife agencies should continually reevaluate their public involvement processes to ensure new wildlife stakeholders' attitudes and concerns are represented.  相似文献

ABSTRACT River otter (Lontra canadensis) populations have been difficult to monitor and information on densities is lacking throughout their range. To obtain DNA-based population estimates of river otters we developed 2 traps to capture hair; a modified body-snare and a modified foot-hold trap. Of 82 traps activated 77 captured hairs (94%). Traps snagged 3–20 guard hairs per capture. Our capture rates of otter hair ranged from one capture per 3.6 trap nights to one capture per 156.6 trap-nights. Our traps provide an effective, noninvasive technique for obtaining hair DNA from individual river otters.  相似文献

adhB和pdc是运动发酵单胞菌产乙醇途径的关键基因,分别编码乙醇脱氢酶和丙酮酸脱羧酶,将添加有聚球藻PCC7942rbcLS基因RBS序列的adhB和pdc基因插入pUC18载体,经双重菌液PCR检验和酶切检验得到分别含有pUC-adhB、pUC-pdc和pUC-adhB-pdc载体的3个重组菌株。活性检测实验表明聚球藻PCC7942的rbcLS基因的RBS序列能有效介导运动发酵单胞菌的adhB和pdc基因在大肠杆菌中表达,摇瓶发酵实验表明重组大肠杆菌的产乙醇能力较出发菌株大幅提升。鉴于乙醛指示平板法存在着对希夫试剂的要求较高、易产生较强的背景色等缺点,对定性检测丙酮酸脱羧酶和乙醇脱氢酶表达菌株的方法做了改进,即:将菌液诱导表达,然后分别添加对应于两种酶的底物,让酶与底物反应0.5至1小时,之后再加希夫试剂进行显色反应,结果表明改进后的方法比乙醛指示平板法更加简便、快速、可靠。  相似文献

In Synechococcus PCC7942 cells grown in the dark, the concentrations of NAD(H) and NADP(H) were 128+/-2.5 and 483+/-4.0 microm, respectively, while those in the cells under light conditions were 100+/-5.0 and 649+/-7.0 microm, respectively. Analysis of gel filtration indicated that the change of the ratio of NADP(H) to NAD(H) in cyanobacterial cells under light/dark conditions controls the reversible dissociation of the PRK/CP12/GAPDH complex (approximately 520 kDa) consisting of phosphoribulokinase (PRK), CP12, and glyceraldehyde-3-phosphate dehydrogenase (GAPDH). S. 7942 CP12 lacked the two Cys residues essential for formation of the N-terminal peptide loop in the CP12 of higher plants, but the N-terminal region of S. 7942 CP12 had the ability to be associated with PRK. The growth of mutant cells in which the CP12 gene was disrupted by a kanamycin resistance cartridge gene was almost the same as that of wild-type cells under continuous light conditions. However, under the light/dark cycle (12 h/12 h), the growth of CP12-disrupted mutant cells was significantly inhibited compared with that of wild-type cells. The mutant cells showed a decreased rate of O2 consumption and an increased level of ribulose 1,5-bisphosphate compared with wild-type cells in the dark. These data suggest that under light and dark conditions, the oligomerization of CP12 with PRK and GAPDH regulates the activities of both enzymes and thus the carbon flow from the Calvin cycle to the oxidative pentose phosphate cycle.  相似文献

Marine Synechococcus is ubiquitous in aquatic environments. However, distinct phylogenetic lineages of this genus have a complex ecological distribution that is not fully explained. Here, we undertook a broad study of the phosphorus (P)–related behavior of marine Synechococcus isolates from all previously described ribotypes (sensu Fuller et al. 2003 ). A wide variability in P‐related physiology was noted among members of this genus, particularly in the utilization of organic P sources. However, some characteristics (e.g., cell size change during P limitation and the ability to accumulate polyphosphate) were largely consistent with their phylogenetic lineage and inferred ecology, with clear distinctions between oligotrophic, mesotrophic, and opportunistic lineages. Similarly, the ability to induce protein expression in response to P limitation was consistent with the presence/absence of phoB/R regulatory capacity of the corresponding strain. Taxonomic differences in P uptake, storage, and utilization strategies could explain the ubiquitous distribution of marine Synechococcus throughout the world’s oceans and explain the coexistence and/or ecological partitioning of multiple phototrophic taxa in the photic zone of tropical and subtropical oligotrophic oceans.  相似文献

 利用荧光显微技术(Epifluorescence microscopy, EFM)对2006年夏季北黄海水域80个站点的微微型浮游植物进行了检测, 并对其在平面和垂直分布上的变化以及昼夜变化情况进行了研究。检测到聚球藻(Synechococcus, Syn)和真核球藻(Picoeukaryotes, Euk)两类微微型浮游植物, 未检测到原绿球藻(Prochlorococcus, Pro), 其中Syn以富含藻红素的Syn细胞(PE细胞)为优势种类, 而富含藻蓝素的Syn细胞(PC细胞)数量较少。在荣城湾以东的水域, 各水层Syn和Euk的丰度都较其它水域的高。Syn在北黄海冷水团及附近水域下30 m深处和底层水体中出现明显的低值区, Euk丰度受冷水团影响不明显。在垂直分布上, 水表层和水下10 m深处Syn的丰度高于水下30 m深处和底层, 而Euk在垂直分布上无明显差异。在昼夜变化上, 各水层Syn、Euk丰度的白昼与夜间变化趋势无明显区别。  相似文献

This study reports the diversity of uncultured environmental viruses harbouring photosynthetic genes (psbA and psbD) in samples from cold seawater (latitude above 60 degrees ). The viral community in coastal Norwegian waters was separated according to genome size using pulse field gel electrophoresis. Viral populations within a wide genome size range (31-380 kb) were investigated for the presence of the psbA and psbD genes using PCR, combined with cloning and sequencing. The results show the presence of photosynthetic genes in viral populations from all size ranges. Thus, valuable information could be obtained about the size class to which viral particles that encode photosynthesis genes belong. The wide genomic size range detected implies that a different cyanophage profile has been observed than has been reported previously. Thus, the method of phage gene detection applied here may represent a truer picture of phage diversity in general or that there is a larger range of size profile for viruses with psbA and psbD in higher latitudes than for the better-studied lower latitudes. Alternatively, a picture of diversity based on a different set of biases than that from either isolation-based research or from conventional metagenomic approaches may be observed.  相似文献

We studied the mechanisms of the relationships between the generation of millisecond delayed fluorescence in photosystem I (DF) and the oxidative destruction of chlorophyll in the membranes of a thermophilic cyanobacteria Synechococcus elongatusin the temperature range 60–80°C at various irradiation levels and in the presence of substances affecting the intensity of DF. Light and temperature dependencies of the chlorophyll oxidation rates were similar to those of the DF of PSI. Anions Cl, Br, and NO 3, which quench the triplet states of chlorophyll, almost completely inhibited the chlorophyll oxidation and reduced the intensity of the DF maximum by 70%. Under anaerobic conditions and in the presence of sodium ascorbate, the rate of chlorophyll oxidation also markedly decreased. We found that the long-wavelength chlorophyll forms were the most susceptible to oxidation and related the temperature-dependent changes in the DF of PSI and in the oxidative processes in the membranes of thermophilic cyanobacteria to an increase in the concentration of the triplet states of P700and other chlorophyll forms. The latter result from the temperature-dependent inactivation of carotenoids and the inhibition of electron transfer to ferredoxin in PSI.  相似文献
