Abstract The microbial ecology of the sulfur cycle in the anaerobic part of Rotsee (Switzerland) was studied. Almost all the sulfate reduction took place at the sediment surface at a rate of 2 mmol SO2−4 reduced m−2 day−1. Approx. 104 sulfate reducers per ml were present in the surface sediments. The sulfide produced was phototrophically consumed mainly by Thiopedia rosea, Lamprocystis roseopersicina and ' Pelochromatium roseum ' consortia. Thiopedia rosea migrated diurnally about one meter. Bacterial photosynthesis was limited by light and sulfide rather than by temperature.  相似文献

Mitochondria prepared in small scale from skeletal muscle were studied with respiration measurements and low temperature spectroscopy. The method of preparation was developed for 25–100 mg tissue with pigeon breast muscle as model organ. The yield was 40%. Data collected during the developmental work were used to evaluate criteria of mitochondrial quality. The cytochrome c conservation, i.e. cytochrome c per mitochondrial quantity in the preparation relative to that in the tissue, is a most useful test parameter. It is bounded between 0–100%. Proportionality between the state 3 rate and the cytochrome c conservation was not rejected by statistical tests. The respiratory control ratio (RCR) was also highly correlated to the cytochrome c conservation. These correlations might be extrapolated to 100% conservation to give hypothetical tissue values. The cause for the correlations is discussed. The P/O ratio showed only weak dependence on the cytochrome c conservation and the state 4 rate s howed no dependence. Other, rather insensitive test parameters are also discussed. The pigeon breast muscle mitochondria isolated by the final method showed cytochrome c conservation of 73 ± 9% (n = 16). They are compared with pig biceps femoris mitochondria prepared by the same method. The two types of mitochondria show many similarities. Some differences may be explained by a different amount of inner mitochondrial membrane relative to mitochondrial protein. Abstract.  Metabolic rate variation with temperature, body mass, gender and feeding status is documented for Glossina morsitans centralis . Metabolic rate [mean ± SE; VCO2= 19.78 ± 3.11 μL CO2 h−1 in males (mean mass = 22.72 ± 1.41 mg) and 27.34 ± 3.86 μL CO2 h−1 in females (mean mass = 29.28 ± 1.96 mg) at 24 °C in fasted individuals] is strongly influenced by temperature, body mass and feeding status, but not by gender once the effects of body mass have been accounted for. A significant interaction between gender and feeding status is seen, similar to patterns of metabolic rate variation documented in Glossina morsitans morsitans . Synthesis of metabolic rate-temperature relationships in G. m. centralis , G. m. morsitans and Glossina pallidipes indicate that biting frequency as well as mortality risks associated with foraging will probably increase with temperature as a consequence of increasing metabolic demands, although there is little evidence for variation among species at present. Furthermore, metabolic rate–body mass relationships appear to be similarly invariant among these species. These data provide important physiological information for bottom-up modelling of tsetse fly population dynamics.  相似文献

Photosynthetic activity and growth physiology of Spirulina platensis (Nordstedt) Geitler cultures maintained at ultrahigh cell densities (i.e. above 100 mg chlorophyll-L?1) in a newly designed photobioreactor were investigated. Nitrogen (NaNO3) in standard Zarouk medium was characterized as a major nutrient-limiting factor in such cultures. The effect of ultrahigh cell density on photoinhibition of photosynthesis, as reflected by chlorophyll fluorescence and photosynthetic oxygen evolution, was studied: elevating the population density may arrest photoinhibition induced by high photon flux density, as well as low temperature. The relationship between incident irradiance and oxygen production rate was linear in situ for cultures at the optimal cell density, indicating that light limitation rather than light saturation or photoinhibition is the dominant condition outdoors in cultures of ultrahigh cell densities. In contrast with other reports, the extent of biomass loss at night due mainly to dark respiration was found to be relatively small when cell density was optimal, exerting only a minor effect on overall net productivity. Measurements of oxygen consumption at night revealed low rates of respiration, which may be explained by the low value of the volumetric mass transfer coefficient (KLa) of oxygen. Hence, reduced oxygen tension may play a role in preventing full expression of the respiratory potential in ultrahigh cell density cultures in which photoadaptive strategy may explain cell composition. Ultrahigh cell densities optimized with respect to the intensity of the light source, the length of the light path, and the extent of stirring represent the key for obtaining high output rates of cell mass and some natural products.  相似文献

Soluble carbohydrate, protein, polysaccharide and cell wall composition were assayed in freshly harvested Pleurotus ostreatus sporophores and those stored for 4 days at 2° or 18°. Mannitol and trehalose were present at 1.8 and 6.5% dry wt respectively in fresh sporophores, and at reduced levels in those stored at 18°. In sporophores stored at 2°, trehalose levels increased by up to 122%. Soluble polysaccharide appeared to be composed of glycogen-like material, which was susceptible to post-harvest breakdown, and components containing mannose and other sugars. The total protein content was 42% dry wt; no protein degradation was seen in sporophores stored at 2°, but about 25% of the protein disappeared during storage at 18°. Cell wall polysaccharide was utilised during storage. Respiration rate was about 8–10 ml CO2/g dry wt/hr at harvest and declined to about 5 ml/g dry wt/hr after 40 hr storage at 18°.  相似文献

The role of the chorion in the mechanism of respiration is examined for the egg of Megaselia imitatrix. An air layer in the chorion is not essential for respiration, and oxygen can be absorbed through a hydrophilic pathway in the chorion by diffusion from the surrounding water. A model based on the diffusion of oxygen through the surface layer of the water down to the depth of the egg is proposed and presented in detail.  相似文献

A bioenergetics model was developed from observed consumption, respiration and growth rates for zebrafish Danio rerio across a range (18–32° C) of water temperatures, and evaluated with a 50 day laboratory trial at 28° C. No significant bias in variable estimates was found during the validation trial; namely, predicted zebrafish mass generally agreed with observed mass.  相似文献

Ca2+-activated K+ channels (KCa) are expressed at the plasma membrane and in cellular organelles. Expression of all KCa channel subtypes (BK, IK and SK) has been detected at the inner mitochondrial membrane of several cell types. Primary functions of these mitochondrial KCa channels include the regulation of mitochondrial ROS production, maintenance of the mitochondrial membrane potential and preservation of mitochondrial calcium homeostasis. These channels are therefore thought to contribute to cellular protection against oxidative stress through mitochondrial mechanisms of preconditioning. In this review, we summarize the current knowledge on mitochondrial KCa channels, and their role in mitochondrial function in relation to cell death and survival pathways. More specifically, we systematically discuss studies on the role of these mitochondrial KCa channels in pharmacological preconditioning, and according protective effects on ischemic insults to the brain and the heart.  相似文献

土壤呼吸组分的区分对于理解地下碳循环过程非常重要。而菌根真菌在地下碳循环过程中扮演着重要的角色, 但是有关菌根呼吸在草原生态系统中的研究相对较少。该研究在内蒙古半干旱草原应用深浅环网孔法, 结合浅环、深环(排除根系)和一个带有40 μm孔径窗口的土壤环(排除根系但是有菌根菌丝体)将根和菌丝物理分离, 来区分不同的呼吸组分。结果表明: 异养呼吸对总呼吸的贡献比例为51%, 根呼吸的贡献比例为26%, 菌根呼吸的贡献比例为23%, 菌根呼吸的比例3年变化范围为21%-26%。与国内外研究相比, 此方法提供了一个相对稳定的菌根呼吸测量精度范围, 在草原生态系统中切实可行。对菌根呼吸的准确定量将有助于预测草原生态系统土壤碳释放过程对未来气候变化的响应。  相似文献
