本文论述了生物多样性的价值、多样性丧失的严重性和后果,以及多样性保护与持续利用的对策。文中强调指出,生物界中的每片基因,每一个物种,每一类生态系统对人类的持久生存都是无价之宝,任何多样性的丧失都是不可逆、不可再生的,因而对人类的损失是难以估量的。然而目前由于人类剧烈的活动干扰如滥砍滥伐、滥捕滥猎、环境污染、火灾、垦荒等,生物多样性丧失的速率怵目惊心!若不赶快行动起来,人类赖以生存的生物多样性将所剩无几,人类生存的危机也将难以避免。从保护与利用及其协同发展出发,本文呼吁要大力开展生物多样性的研究和开发工作,目的一方面在于进一步加强生物多样性的基础调查和研究工作,另一方面在于使生物多样性资源更好地为人类造福。为减缓目前生物多样性所承受的压力及促进其恢复,本文建议对完全依赖野生生物资源的传统产业征收“资源更新税”;而对开发和利用生物多样性资源,不仅不影响野生资源,而且能替代它,或减缓其压力,或促进其恢复的高新技术产业,在税收上给予特别优惠。  相似文献

我们用免疫胶体金色埋前标记技术和免疫荧光技术研究了人胚肺细胞(HEL)内,人巨细胞病毒(HCMV-AD_(169))对单纯疱疹病毒1型(HSV-ⅠSM_(44))抗原表达的影响,旨在探讨在细胞这一微生境内,一病毒对另一病毒可能发生的影响。电镜下计数HSV-1组和HCMV HSV-1组特异性结合金颗粒数得HSV-1组为657个,HCMV HSV-1组的总数为283个。t检验P<0.01,差别非常显著。并且HSV-1组细胞的胞浆中的病毒颗粒,比HCMV HSV-1组明显多。荧光显微镜下:HSV-1组阳性细胞数为689个HCMV HSV-1组只有484个,经poisson分布u检验,P<0.01,差别非常显著。免疫荧光实验还表明:HSV-1组,抗血清在1:320时仍有荧光清晰的阳性细胞,而HCMV HSV-1组,抗血清在1:160时,却无荧光阳性细胞。细胞病变效应(CPE)动态观察显示:HSV-1组8小时即有细胞病变,24小时蔓延整个单层;而HCMV HSV-1组超感染14小时才有细胞病变。24小时约有75%细胞受累。结果表明HCMV对HSV-1的抗原表达有明显的抑制作用。对抑制作用的可能机理及其在分子生态学中的意义,进行了讨论。  相似文献

This paper was adopting intestinal normal microbiota: Micrococcus tntestinalt, sp. and Beneckea campbell, and were prepared to microecologics, also gave a rebuild of intestinal micro ecospecies for ossified eels, and research effect of increasing growth and rejuvenation to them, by the microecologics. Moreover, specially to set up double-control, including a thyroideum medicament and an empty test group, for the comparison and analysis. Our results show that the intestinal microecologics of eel, for increasing growth and rejuvenation to ossified eels, that are botth achieve striking effect. But looked the thyroideum for increasing growth to ossified eels were be very effective. 本实验采用木瓜酶水解,SPA柱亲合层析等手段得到人IgGFc段及Fab段,以Sigma抗人IgGfFc段和抗人IgG Fab段单抗为标准品,鉴定了细胞库中抗人IgG系列的部分细胞株,得到特异性分泌抗人IgG Fc段和抗人IgG Fab段单抗的细胞各一株。 在上述实验基础上,用抗人IgG Fc及抗人IgG Fab单抗分别制备了Sepharose4B亲合层析柱,提纯了酶解人IgG Fc、Fab片段,经ELISA法鉴定,相互之间无交叉反应。同时用此方法制备了人抗HBe Fab片段,并将该片段进行了过氧化物酶标记,用来配制HBe ELISA诊断盒,证明其生物活性未受影响,而且消除了类风湿因子引起的HBe Ag假阳性现象。因抗HBe单抗来源困难,如采用HBe多抗制备ELISA试剂,本法将是提高质量的一个好方法。  相似文献

Schiff bases (imines or azomethines) are versatile ligands synthesized from the condensation of amino compounds with active carbonyl groups and used for many pharmaceutical and medicinal applications. In our study, we aimed to determine the cytotoxic, antifungal and larvicidal activities of biologically potent bis-sulfonamide Schiff base derivatives that were re-synthesized by us. For this aim, 16 compounds were re-synthesized and tested for their cytotoxic, antifungal and larvicidal properties. Among this series, compounds A1B2 , A1B4 , A4B2 , A4B3 , and A4B4 were shown to have cytotoxic activity against tested cancer lung cell line (A549). The most potent antifungal activity was observed in compounds A2B1 and A2B2 against all fungi. A1B1 showed the strongest larvicidal effect at all concentrations at the 72nd h (100% mortality). These obtained results demonstrate that these type of bis-substituted compounds might be used as biologically potent pharmacophores against different types of diseases.  相似文献

Radiocarbon (14C) has been used to date carbon-rich objects in Earth science, archeology, and history since the 1940s. New methods, using spikes in 14C caused by solar proton events, can be used to annually date wood when crossdating is not possible, such as when sample size is low, samples are floating in time, or external disturbances lead to insecure dates. Here, we use a spike in radiocarbon during a solar energetic particle (SEP) event in 774/775 CE to confirm crossdating of a poorly-replicated King Billy pine (Athrotaxis selaginoides) chronology. Low sample depth between 1498 and 1523 CE (two trees) prevented confident dating of the early period of the chronology. Three core samples with strong correlation with the master chronology that likely included the 774/775 CE Miyake SEP event were identified for radiocarbon isotope analysis. We sectioned segments centered on the estimated 774/775 CE date and then isolated the holocellulose in each sample. Samples were sent to an accelerator mass spectrometry (AMS) for radiocarbon measurements. The AMS data confirmed the crossdating accuracy of the tree ring series and reinforces the applicability of this technique to anchor poorly dated tree ring series in time. In addition, we found sample processing with a microtome proved superior for holocellulose extractions and yielded more accurate 14C measurements. We recommend sampling with a microtome, processing at least three samples per year, and including sample masses greater than 100 ug C to confirm dating using radiocarbon spikes.  相似文献

The increase in frequency and intensity of wildfires is seriously affecting forest ecosystems, especially in drought-prone areas. Trees’ recovery after fire is related to direct tree damage and is influenced by climate conditions, such as warm temperature and water shortage. In this study, we evaluate the post-fire effects on a Pinus pinaster Aiton forest growing in a hot and dry area of the Mediterranean region by comparing burned trees with severe crown reduction against unburned and not-defoliated trees. Inter-annual analyses of dendrochronology and stable isotopes in tree rings were combined with xylogenesis monitoring to investigate the effects of fire on tree growth, ecophysiological processes and wood formation. Tree-ring and isotope data showed a growth reduction and a decrease in photosynthetic activity in the burned trees, compared to control individuals, in the three years after fire. Further, the monitoring of cambial activity demonstrated a negative influence of warm and dry periods on wood formation, low xylem production, a delay in phenology and a reduction in xylem plasticity in burned trees. Our findings suggest that substantial photosynthetic limitations caused by crown defoliation and recurrent drought events could lead to severe growth decrease and reduction of trees ability to regain the pre-disturbance productivity rates.  相似文献

ABSTRACT. The microsporidium Chytridiopsis trichopterae n. sp., a parasite of the midgut epithelium of larvae of the caddis fly Polycentropus flavomaculatus found in southern Sweden, is described based on light microscopic and ultrastructural characteristics. All life cycle stages have isolated nuclei. Merogonial reproduction was not observed. the sporogony comprises two sequences: one with free spores in parasitophorous vacuoles, the other in spherical, 5.6-6.8 μm wide, sporophorous vesicles which lie in the cytoplasm. the free sporogony yields more than 20 spores per sporont. the vesicle-bound sporogony produces 8, 12 or 16 spores. the envelope of the sporophorous vesicle is about 82 nm thick and layered. the internal layer is the plasma membrane of the sporont; the surface layer is electron dense with regularly arranged translucent components. Both spore types are spherical. They have an ~ 35-nm thick spore wall, with a plasma membrane, an electron-lucent endospore, and an ~ 14-nm thick electron-dense exospore. the polar sac is cup-like and lacks a layered anchoring disc. the polar filament is arranged in two to three isofilar coils in the half of the spore opposite the nucleus. the coupling between the polar sac and the polar filament is characteristic. the surface of the polar filament is covered with regularly arranged membraneous chambers resembling a honeycomb. There is no polaroplast of traditional type. the cytoplasm lacks polyribosomes. the nucleus has a prominent, wide nucleolus. the two spore types have identical construction, but differ in dimensions and electron density. Free living spores are about 3.2 μm wide, the diameter of the polar filament proper is 102-187 nm, the chambers of the honeycomb are 70-85 nm high, and the polar sac is up to 425 nm wide. Living spores in the vesicle-bound sporogony are about 2.1 μm wide, the polar filament measures 69-102 nm, the chambers of the honeycomb are about 45 nm high, and these spores are more electron dense. Comparisons of cytology (especially the construction of the spore wall and the polar filament and associated structures) and life cycles reveal prominent differences among the Chytridiopsis-like microsporidia, and close relationships between the families Chytridiopsidae and Metchnikovellidae.  相似文献

The time course and nature of the pulmonary inflammatory and antioxidant responses, both during and after hyperoxic-induced acute lung injury were studied in the preterm guinea pig. Three-day preterm (65 days gestation) guinea pigs were randomly exposed to either 21% O2 (control) or 95% O2 (hyperoxia) for 72 hours. All pups were then maintained in ambient conditions for up to a further 11 days, during which time lung damage was monitored. In animals exposed to hyperoxia, evidence of acute lung injury and inflammation was characterized by a marked increase in microvascular permeability and elevated numbers of neutrophils in bronchoalveolar lavage fluid. Protein concentration, elastase-like activity and elastase-inhibitory capacity in lavage fluid were at a maximum at the end of the 72 hours hyperoxic exposure. Four days later, all values had returned to control levels. In contrast, increased numbers of neutrophils, macrophages and lymphocytes were recovered in the lavage fluid during this early recovery period. Coinciding with the influx of inflammatory cells, there was a significant increase in glutathione peroxidase, manganese superoxide dismutase and catalase activities in immature lung. Lung copper/zinc superoxide dismutase activity remained unchanged during both experimental periods. The strong temporal relationship between the influx of inflammatory cells to the lung and the induction of pulmonary antioxidant enzyme defences suggests that a common mechanism underlies both responses. These findings have led us to regard inflammation in the hyperoxic-injured immature lung as a beneficial event and not, as previously suggested, as part of the injurious process.  相似文献

-phenyl-tert-butyl-nitrone (PBN) a spin adduct forming agent is believed to have a protective action in ischemia-reperfusion injury of brain by forming adducts of oxygen free radicals including ±OH radical. Electron paramagnetic resonance (EPR) has been used to both detect and monitor the time course of oxygen free radical formation in the in vivo rat cerebral cortex. Cortical cups were placed over both cerebral hemispheres of methoxyflurane anesthetized rats prepared for four vessel occlusion-evoked cerebral ischemia. Prior to the onset of sample collection, both cups were perfused with artificial cerebrospinal fluid (aCSF) containing the spin trap agent -(4-pyridyl-1-oxide)-N-tert butylnitrone (POBN 100 mM) for 20 min. In addition 50 mg/kg BW of POBN was administered intraperitoneally (IP) 20 min prior to ischemia in order to improve our ability to detect free radical adducts. Cup fluid was subsequently replaced every 15 min during ischemia and every 10 min during reperfusion with fresh POBN containing CSF and the collected cortical superfusates were analyzed for radical adducts by EPR spectroscopy. After a basal 10 min collection, cerebral ischemia was induced for 15 or 30 min (confirmed by EEG flattening) followed by a 90 min reperfusion. -OH radical adducts (characterized by six line EPR spectra) were detected during ischemia and 90 min reperfusion. No adduct was detected in the basal sample or after 90 min of reperfusion. Similar results were obtained when diethylenetriaminepenta-acetic acid (100 μM; DETAPAC) a chelating agent was included in the artificial CSF. Systemic administration of PBN (100 mg/kg BW) produced a significant attenuation of radical adduct during reperfusion. A combination of systemic and topical PBN (100 mM) was required to suppress -OH radical adduct formation during ischemia as well as reperfusion. PBN free radical adducts were detected in EPR spectra of the lipid extracts of PBN treated rat brains subjected to ischemia/reperfusion. Thus this study suggests that PBN's protective action in cerebral ischemia/reperfusion injury is related to its ability to prevent a cascade of free radical generation by forming spin adducts.  相似文献
