In order to confirm that mechanosensitive Ca2+ channels are activated by membrane stretching, we stretched or compressed the plasma membrane of Chara by applying osmotic shrinkage or swelling of the cell by varying the osmotic potential of the bathing medium. Aequorin studies revealed that treatments causing membrane stretching induced a transient but large increase in cytoplasmic concentration of Ca2+ (Δ[Ca2+]c). However, the observed Δ[Ca2+]c decreased during the treatments, resulting in membrane compression. A second experiment was carried out to study the relationship between changes in membrane potential (ΔE m) and stretching or compression of the plasma membrane. Significant ΔE m values, often accompanied by an action potential, were observed during the initial exchange of the bathing medium from a hypotonic medium to a hypertonic one (plasmolysis). ΔE m appears to be triggered by a partial stretching of the membrane as it was peeled from the cell wall. After plasmolysis, other exchanges from hypertonic to hypotonic media, with their accompanying membrane stretching, always induced large ΔE m values and were often accompanied by an action potential. By contrast, action potentials were scarcely observed during other exchanges from hypotonic to hypertonic solutions (=membrane compression). Thus, we concluded that activation of the mechanosensitive channels is triggered by membrane stretching in Chara.  相似文献

The drop vertical jump is a popular plyometric exercise. Two distinct techniques are commonly used to initiate the drop vertical jump. With the ‘step-off’ technique, athletes step off a raised platform with their dominant limb, while their non-dominant limb remains on the platform. In contrast, with the ‘drop-off’ technique, athletes lean forward and drop off the platform, with both feet leaving the platform more simultaneously. The purpose of this study was to compare landing and jumping kinetics, inter-limb kinetic symmetry, and jump performance when individuals used the step-off and drop-off techniques, and to examine whether potential differences between these techniques are affected by platform height. Sixteen subjects completed drop vertical jumps with the drop-off and step-off techniques, from relatively low and high platform heights. Ground reactions forces were recorded for the dominant and non-dominant limbs during the land-and-jump phase of the drop vertical jump. Subjects demonstrated greater inter-limb asymmetry in peak impact forces when using the step-off technique, vs. the drop-off technique. This difference between the techniques was consistent across platform heights. The step-off technique appears to result in greater asymmetry in limb loading, which could contribute to the development of neuromuscular asymmetries between the limbs and/or asymmetric landing patterns.  相似文献

目的:本研究在成功构建面颌肌细胞体外培养一力学刺激模型的基础上,探讨机械张应力对细胞内Na+水平和面颌肌细胞Na+/K+-ATPase功能活性的影响。方法:本实验采用Blua法,对SD大鼠乳鼠面颌肌细胞进行体外原代培养、鉴定并绘制生长曲线;取第3代细胞接种于细胞加力板上,采用Forcel四点弯曲加力装置,对细胞分别施加1 h、2 h、4 h、8 h、12 h、16 h、24 h和48 h的张应力刺激,后测定细胞内Na+水平变化和Na+/K+-ATPase功能活性改变。结果:(1)Na+水平变化:与对照(不加力)组相比,加力1h细胞内Na+显著增加(P〈0.05),并随加力时间延长逐渐降低,加力至12h时降到正常水平,再延长加力时间Na+无明显变化。(2)Na+/K+-ATPase功能活性变化:分别施加不同时间的周期性张应力后,Na泵的活性在加力8小时后才开始增加(0.5725mmolPi/mgPr.hr),随加力时间延长进一步增加,48小时后达到最大值(0.8963mmolPi/mgPr.h)。结论:周期性张应力刺激会引起细胞内Na+的改变,并可以增加骨骼肌细胞Na+/K+-ATPase的活性。  相似文献

目的:研究周期性张应变对面颌肌细胞Na+/K+-ATPaseα亚单位蛋白表达的影响以确定其作用及机制。方法:在建立面颌肌细胞的力学刺激--细胞体外培养模型的基础上,采用Western blot法分析周期性张应变对Na+/K+-ATPaseα1和α2亚单位蛋白表达的影响,加力组分别给予1、2、12、24和48h的力学刺激,施加力值为15%的细胞形变,频率为10cycles/min。以静态组为对照组。对照组及实验组各包含4个实验样本。Western blot检测Na+/K+-ATPaseα1和α2亚单位蛋白的表达。结果:α1亚单位的蛋白表达量除加力1h组与对照组之间、加力24 h与48 h之间无统计学差异外,其余各组之间以及各组与对照组之间均有显著的统计学意义。α2亚单位蛋白表达量除加力24 h与48 h组之间无统计学差异外,其余各组之间以及各组与对照组之间均有显著的统计学意义。结论:在一定时间范围内,周期性张应变可刺激α1和α2亚单位蛋白表达增加,随作用时间的延长蛋白表达受抑制。提示在肌能力的刺激下,面颌肌细胞的相关酶蛋白的功能及表达将发生适应性改建,但其功能亚基的调控机制可能不同。这为选择不同的方法和手段进行临床干预提供了理论依据,因而具有重要的参考意义。  相似文献

Objective

To determine protein kinase C (PKC), c-Jun NH2-Terminal Kinase (JNK) and P38 mitogen-activated protein kinases (p38MAPK) expression levels and effects of their respective inhibitors on proliferation of human bladder smooth muscle cells (HBSMCs) when physiologically stretched in vitro.

Materials and methods

HBSMCs were grown on silicone membrane and stretch was applied under varying conditions; (equibiaxial elongation: 2.5%, 5%, 10%, 15%, 20%, 25%), (frequency: 0.05, 0.1, 0.2, 0.5, 1 Hz). Optimal physiological stretch was established by assessing proliferation with 5-Bromo-2-deoxyuridine (BrdU) assay and flow cytometry. PKC, JNK and p38 expression levels were analyzed by Western blot. Specificity was maintained by employing specific inhibitors; (GF109203X for PKC, SP600125 for JNK and SB203580 for p38MAPK), in some experiments.

Results

Optimum proliferation was observed at 5% equibiaxial stretch (BrdU: 0.837 ± 0.026 (control) to 1.462 ± 0.023)%, (P < 0.05) and apoptotic cell death rate decreased from 16.4 ± 0.21% (control) to 4.5 ± 0.13% (P < 0.05) applied at 0.1 Hz. Expression of PKC was upregulated with slight increase in JNK and no change in p38MAPK after application of stretch. Inhibition had effects on proliferation (1.075 ± 0.024, P < 0.05 GF109203X); (1.418 ± 0.021, P > 0.05 SP600125) and (1.461 ± 0.01, P > 0.05 SB203580). These findings show that mechanical stretch can promote magnitude-dependent proliferative modulation through PKC and possibly JNK but not via p38MAPK in hBSMCs.  相似文献

Previous studies have shown that the innate immune stimulant LPS augments mechanical ventilation-induced pulmonary coagulation and inflammation. Whether these effects are mediated by alveolar epithelial cells is unclear. The alveolar epithelium is a key regulator of the innate immune reaction to pathogens and can modulate both intra-alveolar inflammation and coagulation through up-regulation of proinflammatory cytokines and tissue factor (TF), the principal initiator of the extrinsic coagulation pathway. We hypothesized that cyclic mechanical stretch (MS) potentiates LPS-mediated alveolar epithelial cell (MLE-12) expression of the chemokine keratinocyte-derived cytokine (KC) and TF. Contrary to our hypothesis, MS significantly decreased LPS-induced KC and TF mRNA and protein expression. Investigation into potential mechanisms showed that stretch significantly reduced LPS-induced surface expression of TLR4 that was not a result of increased degradation. Decreased cell surface TLR4 expression was concomitant with reduced LPS-mediated NF-κB activation. Immunofluorescence staining showed that cyclic MS markedly altered LPS-induced organization of actin filaments. In contrast to expression, MS significantly increased LPS-induced cell surface TF activity independent of calcium signaling. These findings suggest that cyclic MS of lung epithelial cells down-regulates LPS-mediated inflammatory and procoagulant expression by modulating actin organization and reducing cell surface TLR4 expression and signaling. However, because LPS-induced surface TF activity was enhanced by stretch, these data demonstrate differential pathways regulating TF expression and activity. Ultimately, loss of LPS responsiveness in the epithelium induced by MS could result in increased susceptibility of the lung to bacterial infections in the setting of mechanical ventilation.  相似文献

ABSTRACT

What produces and sustains inequality in socioeconomic position and integration across immigrant groups? Luthra, Soehl and Waldinger engage with this core sociological question in their book “Origins and Destinations”. They argue that nominal approaches, which compare and contrast whole national origin groups, are insufficient. Rather, they implement a variable approach. They use data on second generation immigrants living in New York and Los Angeles in the late 1990s to explore empirically what it is about immigrant groups that confers advantages and disadvantages to their offspring in the United States. Rather than comparing whole groups, they use measured data about each group’s characteristics and circumstances to help explain group differences. I describe their innovative approach and provide suggestions for how future research might build on it, such as by accounting for more variation in the context of reception, or by extending the temporal dimensions of inquiry across three or more generations.  相似文献

The purpose of this study was to examine the effects of aging on posture-related changes of the stretch reflex excitability in the ankle extensor, soleus (SOL), and flexor, tibialis anterior (TA) muscles. Fourteen neurologically normal elderly (mean 68 ± 6 years) and 12 young (mean 27 ± 3 years) subjects participated. Under two postural conditions, upright standing (STD) and sitting (SIT), stretch reflex electromyographic (EMG) responses in the SOL/TA muscle were elicited by imposing rapid ankle dorsi-/plantar-flexion. Under the SIT condition, subjects were asked to keep the SOL background EMG level, which is identical to that under the STD condition. In the SOL muscle, both groups showed significant enhancement of the short-latency stretch reflex (SLR) response when the posture changed from SIT to STD. In the TA muscle, the young group showed significant enhancement of the middle- (MLR) and long-latency stretch reflex (LLR) when the posture changed from SIT to STD; no such modulation was observed in the elderly group. Since the TA stretch reflex responses under the STD condition were comparable in the young and elderly groups, the lack of posture-related modulation of the TA muscle in the elderly group might be explained by augmented stretch reflex excitability under the SIT condition. The present results suggest that the (1) SOL SLR responses are modulated both in the young and elderly subjects when the posture is changed from SIT to STD, (2) TA MLR and LLR responses are not modulated in the elderly subjects when the posture is changed from SIT to STD, while each response is same between the young and elderly in STD, and (3) the effect of aging on the posture-related stretch reflex differs in the SOL and TA muscles.  相似文献

Following a series of eccentric contractions, that is stretching of the muscle while generating active tension, the length-tension relationship of isolated amphibian muscle has been shown to shift towards longer muscle lengths (Katz 1939; Wood et al. 1993). Here we report observations of electrically stimulated ankle extensor muscles of nine human subjects, demonstrating a similar shift in optimum angle for torque generation [3.9 (1.5)°] following exercise on an inclined treadmill that involved eccentric contractions in one leg. (All values are means with the SEMs in parentheses.) The shift in the unexercised, control leg was significantly less [mean 0.4 (0.7)°P < 0.05]. Correlated with this shift was a drop in torque [25.1 (5.6)% for the experimental leg; 1.6 (0.7)% for the control leg, P < 0.002]. Optimum angles returned to pre-exercise values by 2 days post-exercise, while torque took a week to recover. A similar shift in optimum length [12 (1.3)% of rest length] was obtained for five toad (Bufo marinus) sartorius muscles subjected to 25 eccentric contractions. Isometrically contracted control muscles showed a smaller shift [3.5 (1.6)%, n = 5]. Accompanying the shift was a drop in tension of 46 (3)% after the eccentric contractions [control isometric, 23 (6)%, P < 0.0001]. By 5 h after the eccentric contractions the shift had returned to control values, while tension had not recovered. When viewed with an electron microscope, sartorius muscles fixed immediately after the eccentric contractions exhibited many small, and a few larger, regions of myofilament disruption. In muscles fixed 5 h after the contractions, no small regions of disruption were visible, and the number of large regions was no greater than in those muscles fixed immediately after the eccentric contractions.
