~(14)C 追踪试验结果表明,白兰瓜幼果中输入的~(14)C-葡萄糖,50%以上转化为稀酸水解和稀酸不水解的结构物质;果实发育后期,输入后48小时,在果肉和种子中分别只有18%和32%的~(14)C 参入结构物质。根据醇溶性糖的纸层析鉴定,幼果薄片渗入的~(14)C-葡萄糖仅转化为果糖,而发育后期果实则更多转化为蔗糖。显然,幼果的代谢模式是使物质和能量导向结构物质的形成;而后期果实生长已基本停止,物质代谢的方向又转向蔗糖合成的轨道上来。蔗糖合成底物试验结果表明,供给幼果不同底物都只有很低的蔗糖合成活性;发育后期果实供给UDPG+F-6-P 底物时可测出较高的蔗糖合成活性,初步推测白兰瓜中蔗糖合成主要是通过蔗糖磷酸酯合成酶来实现的。  相似文献

Behavioral effects of somatostatin-14, and some of its fragments [somatostatin(3–8), somatostatin(9–14), somatostatin(7–10)] after intracerebroventricular (ICV) administration have been investigated in male rats. In a passive avoidance learning test, somatostatin-14 (0.6 nM) given immediately after the learning session increased the avoidance latency at 24 hr after the injection, when compared to a somatostatin(3–8) (0.6 nM)-treated group. However, compared to a saline-treated group, the peptides did not significantly influence the avoidance latency. Somatostatin-14 administered in higher dose (6.0 nM) decreased the avoidance latency compared to the saline-treated group, while its fragments did not influence it. In an open field behavioral test, immediately after the 24-hr passive avoidance test, 6 nM of somatostatin-14 decreased the rearing activity, while the fragments did not influence this behavior. Somatostatin-14 produced barrel rotation in a dose-related manner, but after the injection of a high dose of the peptide (12 nM) all of the animals died in cardiorespiratory failure (apnea, pulmonary oedema). The intracellular pH of the halotolerant green algae Dunaliella tertiolecta, was determined by the distribution of 5,5-dimethyl-2(14C)-oxalolidine-2,5-dione (DMO) between the cell and the surrounding medium. 5,5-dimethyl-2(14C)oxalolidine-2,4-dione was not metabolized by the algal cells. The intracellular pH of Dunaliella tertiolecta was 6.8 in the dark and 7.4 in the light. During a salt stress, after two hours, the intracellular pH was increased by 0.2 pH units in both light and dark. The salt stressed cells maintained a constant pH of about 7.5 over the pH range of 6.5 to 8.5. Because of the relatively low permeability coefficient of the plasma membrane for DMO, this technique does not permit rapid pH determinations during the induction period after a salt stress. The magnitude of the salt induced pH changes measured 2 h after the salt stress implies a minor importance of this alkalization in this time range, but does not exclude a larger importance of pH changes for osmoregulation during the induction period.Abbreviations Chl chlorophyll - DMO 5,5-dimethyl-2(14C)oxalolidine-2,4-dione - PCV packed cell volume - SDS sodium dodecyl sulfate  相似文献

The parasporal body of the mosquitocidal isolate (PG-14) of Bacillus thuringiensis subsp. morrisoni (BTM) contains five major proteins with molecular masses of, respectively, 27.3, 65, 128, 135, and 144 kDa. Proteins corresponding in mass to the first four of these also occur in the mosquitocidal strain, B. thuringiensis subsp. israelensis (BTI), and it is thought therefore that the mosquitocidal activity of both strains is due to these four proteins. In other studies it has been shown that each of these proteins exhibits from moderate to high toxicity to mosquitoes, though the specific toxicity of the 144 kDa protein in PG-14 to mosquitoes remains unknown. In the present study, two parasporal body mutants (M146 and M242) of PG-14 were developed growing the wild-type strain at 42 degrees C. The parasporal body of M146 contained less of the 65-kDa protein and was less toxic (LC50 = 108 ng/ml) to mosquitoes than the wild-type strain (LC50 = 8.3 ng/ml). The parasporal body of M242 consisted of a bipyramidal crystal composed of a 144-kDa protein that was not toxic to the mosquito, Aedes aegypti, but exhibited substantial toxicity (LC50 = 2.5 micrograms/ml) to the lepidopteran. Trichoplusia ni. Because the parasporal bodies of BTI and BTM PG-14 are similar in mosquitocidal toxicity on a weight basis, the latter results suggest the 144-kDa protein, though not mosquitocidal alone, can contribute to mosquitocidal, activity when in the presence of other mosquitocidal proteins.  相似文献

Summary The toxicity of chromium and tin on growth, photosynthetic carbon-fixation, oxygen evolution, heterocyst differentiation and nitrogenase activity ofAnabaena doliolum and its interaction with bivalent cations has been studied. Some interacting cations, viz. Ca2+, Mg2+ and Mn2+, substantially antagonised the toxic effects of chromium and tin with reference to growth, heterocyst differentiation and nitrogenase activity in the following hierarchal sequence: Ca2+ > Mg2+ > Mn2+. However, the sequence of hierarchy was Mg2+ > Ca2+ > Mn2+ for carbon fixation and Mn2+ > Mg2+ > Ca2+ for photosynthetic oxygen evolution. Synergistically inhibitory patterns were noticed for all the parameters, viz. growth,14CO2 uptake, oxygen evolution, heterocyst differentiation and nitrogenase activity ofA. doliolum when Ni2+, Co2+ and Zn2+ were combined with the test metals in the growth medium. These cations followed the following sequence of synergistic inhibition: Ni2+ > Co2+ > Zn2+. Among all the interacting cations, Ca2+, Mg2+ and Mn2+ exhibited antagonistic effects which relieved the test cyanobacterium from metal toxicity. In contrast to this, Ni2+, CO2+ and Zn2+ showed synergistic inhibition which potentiating the toxicity of test metals in the N2-fixing cyanobacteriumA. doliolum. It is evident from the present study that bivalent cations, viz. Ca2+, Mg2+, Mn2+, Ni2+, Co2+ and Zn2+, may appreciably regulate the toxicity of heavy metals in N2-fixing cyanobacteria if present in aquatic media.  相似文献

The error inin vivo 14C incubator measurements of primary production in the Eastern Scheldt when neutral density filters were used and the error obtained when no account was taken of the spectral changes in submarine irradiance that occur with increasing depth, were evaluated theoretically. By multiplying the photosynthetic action spectra of two marine algae by calculated irradiance in the euphotic layer using Kd and Kd() respectively, the gross primary production P[Ed(400–700)] and P[Ed()] was computed. In the green-brown waters of the Eastern Scheldt estuary the use of neutral density filters was sufficient to simulate the underwater light conditions. In clear waters it can cause an overestimation of the gross production.  相似文献

L. Naranbagh (alt. 1587 m) is a polymictic, shallow marl lake in the flood-plain valley of Kashmir, India. Macrofloral affinities resemble Potamogeton Type of Forsberg (1965) with alkaline waters, not rich in phosphorus. CaCO3 precipitation coupled with decline in Ca2+ and alkalinity values are characteristic of the lake. Fluctuations in Mg2+, Na+, K+, and Cl were relatively conservative. The levels of PO inf4 sup3– -P and NO inf3 sup– -N indicate moderate fertility of the lake water.Persistence of a summer-autumn planktonic algal pulse is related to favourable irradiance, high water temperatures, and increased photosynthetic efficiency values. The most striking seasonality in photosynthetic rates (m–2 h–1) between winter minimum (3 mg Cassim) and summer maximum (75.4 mg Cassim) is determined by mainly climatic changes. Energy flow gave annual phytoplankton production of 51.95 × 102 KJ m–2 for the ecosystem.The nutrient levels and productivity rates suggest mesotrophic status of L. Naranbagh in classic oligoeutrophic classification of lake types.  相似文献

Fruits of tomato incorporated [2-14C]mevalonic acid lactone into the steroidal glycoalkaloid α-tomatine. Young fruits showed the greatest alkaloid-synthesizing ability but this decreased as the fruits developed. Analysis of sap exuded from fruit stalks and also application of[4-14C]cholesterol to leaves confirmed that tomatine is not transported into fruits from vegetative organs. Accumulation of this alkaloid in fruits thus appears entirely due to synthesis. Excised fruits of all developmental stages degraded injected [14C]tomatine and rates were directly related to fruit age. The pattern of accumulation/decline in fruit tomatine may be explicable on the basis of changing capacity for synthesis/degradation during development. Label from injected [14C]tomatine was present mainly in chlorophylls and carotenoids where it increased with time as that in tomatine decreased. The significance of the relationship between tomatine disappearance and carotenoid development is briefly discussed. The aglycone tomatidine was not detected in green fruits but a Δ16-5α-pregnenolone-like compound was.  相似文献
