Compromised TLR-mediated chronic inflammation contributes to bacterial infection-caused chronic suppurative otitis media, but the mechanisms are unclear. The present study examined the expression status of nuclear erythroid 2-related factor 2 (Nrf2) and TLRs in human middle-ear mucosae tissues collected from patients with chronic suppurative otitis media, chronic otitis media and non-otitis media, and found that Nrf2 was high-expressed, whereas TLR4, instead of other TLRs, was low expressed in chronic suppurative otitis media compared to chronic otitis media and non-chronic otitis media groups. Consistently, inflammatory cytokines were significantly up-regulated in the chronic suppurative otitis media group, instead of the chronic otitis media and non-chronic otitis media groups. Next, LPS-induced acute otitis media and chronic suppurative otitis media models in mice were established, and high levels of inflammatory cytokines were sustained in the mucosae tissues of chronic suppurative otitis media mice compared to the non-otitis media and acute otitis media groups. Interestingly, continuous low-dose LPS stimulation promoted Nrf2 expression, but decreased TLR4 levels in chronic suppurative otitis media mice mucosae. In addition, knock-down of Nrf2 increased TLR4 expression levels in chronic suppurative otitis media mice, and both Nrf2 ablation and TLR4 overexpression inhibited the pro-inflammatory cytokine expression in chronic suppurative otitis media. Finally, we found that both Nrf2 overexpression and TLR4 deficiency promoted chronic inflammation in LPS-induced acute otitis media mice models. Taken together, knock-down of Nrf2 reversed chronic inflammation to attenuate chronic suppurative otitis media by up-regulating TLR4.  相似文献

繁缕(Stellaria media)和小繁缕(S.apetala)是两个形态相似的近缘种,有人把后者作为前者的亚种或变种来处理。本文通过对南京地区不同生境的三个自然群体和三个人工控制栽培群体的取样,以群体为单位,分别测算了叶、萼片、雄蕊、花瓣、果实和种子的8个数量性状的变异,绘制了多角形图;对花粉粒和种子进行了扫描;还通过花蕾套袋试验对种子活力作了检查。结果发现繁缕和小繁缕都是近亲繁殖植物,在形态上区别明显,对生态环境的要求基本相同,但小繁缕似更能耐受人为的践踏和刈割。  相似文献

Monoliths represent a special class of chromatographic supports. In contrast to other stationary phases, they consist of a single piece of highly porous material through which a sample is mainly transported by convection. As a consequence, monoliths enable fast separations and exhibit flow-unaffected properties, which make them attractive for purification of macromolecules like proteins or DNA. In this work, methacrylate-based monolithic columns with the bed volume up to 8000 ml are characterized. They perform high-resolution separations of several hundreds of grams of proteins per hour by utilizing liter per minute flow rates. They are incompressible under these operating conditions and resistant to strong alkaline conditions.  相似文献

将城市污泥应用于生态恢复因能避开粮食作物食物链而具有良好前景,但其对地表水环境影响仍不十分清楚.本文将城市污泥与建筑垃圾按1∶1体积比混合为生长基质,覆盖在模拟的煤矸石边坡上,播种8种乡土木本植物,对生长季植物生长情况以及坡面地表水和下渗水的电导率、p H、氮磷钾、重金属、多环芳烃等含量进行分析.结果表明:坡面植物生长良好,平均覆盖度达60%;地表水和下渗水的p H值近中性,变化不大,而电导率、氮磷钾、重金属和多环芳烃含量均较高,其中,氮、磷含量在整个生长季超过国家地表水环境质量Ⅴ类水质标准;7月重金属含量最高,其中,As含量只达地表水环境质量Ⅳ~Ⅴ类标准,其余重金属含量多达Ⅱ~Ⅳ标准;随着夏季雨水淋洗增加以及植物-土壤系统对化学物质的吸收、降解和固着作用,地表水和下渗水的电导率、氮磷钾、重金属和多环芳烃含量均显著下降,生长季后期重金属含量达到地表水环境质量Ⅱ~Ⅲ类标准,多环芳烃含量减少约一半.将城市污泥直接应用在煤矸石边坡生态恢复中有利于植物生长,植物-土壤系统使得生长基质中的有害物含量逐渐降低,对地表水环境的负影响主要表现为氮、磷的富营养化,但总体上其环境安全性可控.  相似文献

建立Trametes hirsuta的生长繁殖和对模式染料比布列希猩红脱色降解的反应体系,研究表明:菌的生长与降解活动的适宜温度为30℃,静培养;培养基组分对脱色降解的影响不大;从便于观察和缩短反应周期考虑,土豆液体培养基有明显的优点,可作为建立Trametes hirsuta反应体系的首选培养基。菌对比布列希猩红、直接深蓝L-3RB、活性艳蓝X-BR、碱性紫5BN和亚甲基蓝等均有较好的脱色降解效果。  相似文献

This study aimed to investigate the effect of conditioned media (CM) from osteo‐differentiating and adipo‐differentiating human mesenchymal stem cells (MSCs) isolated from lipoaspirates of healthy female donors on the viability of triple‐negative breast cancer cells MDA‐MB231. The CM of undifferentiated and differentiating MSCs were collected after 7, 14, 21 and 28 days of culture. The effects of MSC CM on cell proliferation were assessed using an 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay after 24 h. The effects of osteo‐differentiating cell CM on apoptotic promotion, cell cycle impairment, mitochondrial transmembrane potential dissipation, production of reactive oxygen species and autophagosome accumulation were analysed by flow cytometry and Western blot. MTT assay showed that only CM collected from osteo‐induced cells at day 28 (d28O‐CM) reduced tumour cell viability. Treatment with d28O‐CM restrained cell cycle progression through G2 phase, elicited a caspase‐8‐driven apoptotic effect already after 5 h of culture, and down‐regulated autophagosome accumulation and beclin‐1 expression. The finding that factor(s) secreted by osteo‐differentiating MSCs shows properties of an apoptotic inducer and autophagy inhibitor on triple‐negative breast cancer cells may have an important applicative potential that deserves further investigation. VIA medium for Stenotrophomonas maltophilia was modified by substituting meropenem (16 mg/L) for imipenem. S. maltophilia grew from 40% of drains sampled within a hospital and surrounding locations in Perth, Western Australia. The specificity of the new medium for S. maltophilia was 62%, and all contaminating bacteria were easily distinguishable by phenotypic tests and PCR.  相似文献

Aims:  To develop a selective medium for isolation of F. tularensis, F. novicida and F. philomiragia from environmental samples.
Methods and Results:  A selective media, cysteine heart agar with 9% chocolatized sheep blood, containing polymyxin B, amphotericin B, cyclohexamide, cefepime and vancomycin (CHAB-PACCV) was developed and evaluated for growth of Francisella spp. No differences were observed in recovered colony forming units (CFUs) for F. tularensis , F. novicida and F. philomiragia on CHAB-PACCV vs nonselective CHAB. Growth of non- Francisella species was inhibited on CHAB-PACCV. When environmental samples were cultured on CHAB and CHAB-PACCV, only CHAB-PACCV allowed isolation of Francisella spp. Three new Francisella strains were isolated directly from seawater and seaweed samples by culture on CHAB-PACCV.
Conclusions:  CHAB-PACCV can be used for direct isolation of Francisella spp from environmental samples.
Significance and Impact of the Study:  Francisella spp. show a close association with environmental sources. Future utilization of CHAB-PACCV for isolation of Francisella spp. directly from environmental samples should prove valuable for investigating outbreaks and human infections attributed to environmental exposure.  相似文献

对繁缕(Stellaria media)和无瓣繁缕(S.apetala)的6个居群的57个性状进行Q-聚类和R-聚类的研究。结果表明:(1)Q-聚类中,用一条结合线,可以把繁缕的4个居群聚为一类,无瓣繁缕的2个居群聚为一类。这一结果支持肥繁缕和无瓣繁缕划分为两个物种;(2)R-聚类中,发现了呈现完全正相关、极大正相关和极大负相关的性状,并根据R-聚类的结果,运用一条适当的结合线,把繁缕和无瓣繁缕的57个性状划为5个类群,并分析了各性状的分类学意义。  相似文献
