An improved calcium alginate gel formulation was developed and tested as a carrier for entomopathogenic nematodes against Spodoptera littoralis and Helicoverpa armigera larvae. Mortality of 100% was caused in 4th instar larvae of the two insects by feeding them on 1000 infective juveniles (IJ) g -1 of Steinernema carpocapsae (ALL strain) in the gel for 24 h. Exposing 2nd to 5th instars of H. armigera and 3rd to 6th of S. littoralis to 500 IJ g -1 of S. carpocapsae (ALL strain) resulted in 70-100% larval mortality. Mature larvae were less susceptible to the nematodes. Mortality of larvae exposed to 500 IJg -1 of S. carpocapsae (ALL strain) ranged from about 45-55% at 4 h to 90-95% at 48 h. Fourth instar larvae fed for 24 h with 250 IJ g -1 of nematode strains in gel showed in S. littoralis ranges of susceptibility in the following descending order: S. feltiae (IS -7 strain) = S. carpocapsae (DT strain) = S. feltiae (IS-6 strain) > S. carpocapsae (Mexican strain) = S. carpocapsae (ALL strain) = Heterorhabditis bacteriophora (HP-88 strain) = H sp. (IS-5 strain) > S. riobravae (Texas strain); in H. armigera the rating was: S. feltiae (IS-7 strain) = H. bacteriophora (HP88 strain) > S. carpocapsae (ALL strain) = S. feltiae (IS-6 strain ) = Heterorhabditis sp. (IS5 strain) > S. carpocapsae (Mexican strain) > S. riobravae (Texas strain) . The number of nematodes per larval cadaver increased with mortality rates. In greenhouse tests at 28 &#45 2°C and 90% relative humidity, gel discs containing 500 IJ g -1 of nematodes were pinned to leaves of potted plants of cotton ( Gossypium hirsutum ) (Acala SJ2) and the plants were offered to S. littoralis larvae. Larval mortality of 89 &#45 12.7% was caused by S. feltiae (IS-7 strain) and most of the plant leaves were protected against the larvae by the nematodes. In the control, larval mortality was 3.3 &#45 0.05% and the plants were almost completely defoliated. Possibilities of using the gel-nematode formulation to protect sheltered crops against insect pests are discussed  相似文献

Abstract  1. Chrysoperla carnea is an important predatory insect in maize. To assess the ecological effects of Bt-maize, expressing the Cry1Ab protein, on larvae of this predator, the following factors were examined: (1) the performance of three prey herbivores ( Rhopalosiphum padi , Tetranychus urticae , and Spodoptera littoralis ) on transgenic Bt and non‐transgenic maize plants; (2) the intake of the Cry1Ab toxin by the three herbivores; and (3) the effects on C. carnea when fed each of the prey species.
2. The intrinsic rate of natural increase (rm) was used as a measure of performance for R. padi and T. urticae . No difference in this parameter was observed between herbivores reared on Bt or non‐transgenic plants. In contrast, a higher mortality rate and a delay in development were observed in S. littoralis larvae when fed Bt-maize compared with those fed the control maize plants.
3. The ingestion of Cry1Ab toxin by the different herbivores was measured using an immunological assay (ELISA). Highest amounts of Cry1Ab toxin were detected in T. urticae , followed by S. littoralis , and only trace amounts detected in R. padi .
4. Feeding C. carnea with T. urticae , which were shown to contain the Cry1Ab toxin, or with R. padi , which do not ingest the toxin, did not affect survival, development, or weight of C. carnea . In contrast, a significant increase in mortality and a delay in development were observed when predators were fed S. littoralis larvae reared on Bt-maize.
5. A combined interaction of poor prey quality and Cry1Ab toxin may account for the negative effects observed on C. carnea when fed S. littoralis . The relevance of these findings to the ecological risks of Bt-maize on C. carnea is discussed.  相似文献

Plants employ a variety of defence mechanisms, some of which act directly by having a negative effect on herbivores and others that act indirectly by attracting natural enemies of herbivores. In this study we asked if a common jasmonate‐signalling pathway links the regulation of direct and indirect defences in plants. We examined the performance of herbivores (direct defence) and the attraction of natural enemies of herbivores (indirect defence) to wild‐type tomato plants and mutant plants that are deficient in the production of the signalling hormone jasmonic acid. Wild‐type plants supported lower survivorship of caterpillars compared with jasmonic acid‐deficient plants. Damaged wild‐type plants were more attractive to predaceous mites compared with undamaged wild‐type plants, whereas damaged jasmonate‐deficient plants were not more attractive to predators. Damaged wild‐type plants induced a greater production of volatile compounds (primarily the sesquiterpene β‐caryophyllene and the monoterpenes α‐pinene, β‐pinene, 2‐carene and β‐phellandrene) compared with damaged jasmonate‐deficient plants. Treating jasmonate‐deficient plants with exogenous jasmonic acid restored both the direct and indirect defence capabilities, demonstrating that jasmonic acid is an essential regulatory component for the expression of direct and indirect plant defence.  相似文献

虫酰肼对甜菜夜蛾多巴脱羧酶和酪氨酸羟化酶的抑制作用   总被引:2,自引:0,他引:2  
虫酰肼模拟昆虫蜕皮激素的作用干扰新表皮的形成。为了探讨虫酰肼对昆虫新表皮形成的影响是否与抑制表皮形成相关酶的活性有关, 本研究应用高效液相色谱-荧光检测法(HPLC-RP), 测定了甜菜夜蛾Spodoptera exigua 5龄幼虫用虫酰肼处理不同时间(24, 48和72 h)后多巴脱羧酶和酪氨酸羟化酶的活性。结果表明: 用LC11 (28.41 μmol/L)和LC33 (85.23 μmol/L)两个亚致死剂量的虫酰肼处理5龄幼虫后, 多巴脱羧酶和酪氨酸羟化酶的活性均受到明显抑制, 高浓度的抑制作用大于低浓度的抑制作用。随着处理时间的延长, 同一剂量的抑制作用逐渐增强。进一步测定虫酰肼处理24, 48和72 h后5龄幼虫血淋巴、 脂肪体、 中肠、 表皮和头部的多巴脱羧酶和酪氨酸羟化酶的活性, 可看出虫酰肼对幼虫不同组织的多巴脱羧酶和酪氨酸羟化酶的活性也具有相似的抑制作用。结果提示, 虫酰肼对甜菜夜蛾幼虫多巴脱羧酶和酪氨酸羟化酶活性具有明显抑制作用, 幼虫新表皮形成受阻可能与虫酰肼抑制多巴脱羧酶和酪氨酸羟化酶的活性有关。  相似文献

Fe-S蛋白(rieske iron-sulfur protein, RISP)是线粒体复合物Ⅲ的关键蛋白亚基之一, 在呼吸链电子传递中起着重要作用。本文利用RT-PCR技术和RACE方法获得了斜纹夜蛾Spodoptera litura (Fab.)线粒体复合物ⅢRISP基因, 该基因序列全长4 413 bp, 含有3个内含子; 开放阅读框长816 bp, 编码271个氨基酸。经氨基酸序列比对, 与鳞翅目家蚕Bombyx mori和小菜蛾Plutella xylostella的Fe-S蛋白一致性较高, 分别为83%和79%, 与其他目昆虫的RISP一致性较低。qRT-PCR分析结果表明, SlitRISP基因在斜纹夜蛾不同发育阶段的表达水平存在显著差异, 即幼虫期和成虫期的表达量显著高于卵期和蛹期, 其中幼虫期又以第4和5龄幼虫表达水平显著高于初孵幼虫以及第1, 2, 3和6龄幼虫。斜纹夜蛾SlitRISP基因的克隆成功为今后对其功能研究以及作为靶标设计新型高效杀虫剂提供了基础。  相似文献

喜树碱诱导的草地贪夜蛾Sf9细胞凋亡   总被引:1,自引:0,他引:1  
传统植物源杀虫剂喜树碱具有优异的抑制昆虫生长发育活性, 其诱导昆虫细胞凋亡的作用方式和机制尚不明确, 极大地限制了喜树碱在植物保护领域的应用开发。本研究以1 μmol/L喜树碱诱导草地贪夜蛾Spodoptera frugiperda Sf9细胞呈现细胞皱缩、微绒毛消失和染色质边集等典型细胞凋亡早期超微结构形态特征, 中期凋亡小体逐渐出现并急剧增多, DNA电泳分析可见清晰DNA片段化凋亡特征。流式细胞术分析表明1 μmol/L喜树碱诱导Sf9细胞12 h凋亡率达到最大值39.67%, 是对照的13.13倍, 随后减小。喜树碱诱导Sf9细胞凋亡在12 h和24 h 时Sf caspase-1分别出现两个活性高峰, 表明其作为效应因子在细胞凋亡级联反应过程中具有影响作用。喜树碱显著抑制Sf9细胞拓扑异构酶Ⅰ活性, 阻断解旋负超螺旋pBR322 DNA, 导致DNA损伤进而启动细胞凋亡级联反应使Sf caspase-1活性增加, 提示其信号转导过程是细胞凋亡诱导机制之一。本研究通过分析喜树碱的诱导昆虫Sf9细胞凋亡, 对揭示喜树碱诱导昆虫细胞凋亡的作用机制具有重要启示和帮助。  相似文献

We assessed the effects of the isoflavonoid rotenone, an insecticidal allelochemical occurring in various legumes, on larval performance of three polyphagous noctuid species: the corn earworm (CEW), the fall armyworm (FAW) and the southern armyworm (SAW). As rotenone concentration was increased up to 1% fresh mass in an artificial diet, neither mortality (for all three species) nor food consumption (for SAW and FAW) was significantly affected, but developmental time of the latter two species was prolonged. In contrast, for CEW developmental time was shortened and food consumption declined, especially at the two highest rotenone concentrations. Final biomass of all three species declined as dietary rotenone increased.Analysis of covariance (ANCOVA) was used to evaluate larval food utilization analogous to, but without the potential statistical problems of, the traditional ratio-based indices approximate digestibility (AD) and efficiency of conversion of digested food (ECD). Frass output, statistically adjusted to account for differences in consumption, increased with rotenone concentration for all three species, suggesting that, when ingested, this allelochemical interfered with the digestion and/or absorption of ingested food, especially at the higher concentrations tested.Significant statistical interactions in the ANCOVA necessitated the use of utilization plots to examine the linear regressions between biomass gain and absorption at each rotenone concentration. At the two highest concentrations, rotenone tended to reduce the amount of food absorbed by the larvae, as well as their ability to convert the absorbed food to biomass.When piperonyl butoxide, an inhibitor of the polysubstrate monooxygenase (PSMO) system known to metabolize rotenone, was added to the diet along with 0.5 or 1% rotenone, mortality increased significantly only for FAW, approaching or exceeding 50%. The only apparent effects of dietary rotenone on PSMO (aldrin epoxidase) activity (adjusted by ANCOVA for the covariate, crude homogenate protein) were for CEW, where activity decreased in all rotenone treatments compared to the control, and for SAW, where activity at the 3 lowest rotenone concentrations (0.001–0.1% fm) declined about 50% below that of larvae on the control diet, whereas at 0.5 and 1% rotenone, it increased approximately 2-fold over that on the control diet.  相似文献

This study has demonstrated that low numbers of African armyworms (Spodoptera exempta) (Walker) (Lepidoptera: Noctuidae) can be found throughout the year in parts of the coastal and highland regions of Kenya where there is frequent rainfall. During the study period the numbers of moths caught in pheromone traps in these regions built up during the short and long rains, and decreased dramatically during the intervening dry seasons. There was a lag of one to two months between the peaks for rainfall and moth numbers. This contrasts with the situation at sites of the seasonal outbreaks of armyworm larvae, where a sudden preceding influx of moths coincides with the rainfall. A positive correlation was found, for the long rains seasons only, between the number of armyworm outbreaks throughout the country and the peak numbers of moths in coastal and highland regions.It is suggested therefore that the peak numbers of moths trapped in the eastern highlands and coastal regions during the rainy seasons arise principally from outbreaks in the extensive seasonal grasslands. These grassland areas diminish considerably during the prolonged dry periods between the two rainy seasons, and it is only the eastern highland and coastal regions that are likely to provide suitable habitats for breeding during the dry seasons. The significance of these populations for initiating the first outbreaks of the following season is discussed.  相似文献

We released nearly 332 000 Spodoptera pectinicornis individuals for biological control of the floating aquatic weed Pistia stratiotes (waterlettuce) at 22 sites in Florida during December 1990-December 1997. Predation (by boat-tailed grackles, fire ants, and spiders) and Allee effects (resulting from rapid dispersal of adults) prevented populations from persisting during early attempts. These efforts consisted of small releases at multiple sites, with no nurturing of the introduced populations. Modification of our initial approach resulted in a series of release strategies that consisted of multiple releases at fewer sites and employed progressively more intensive nurturing of the introduced populations. The most successful strategy incorporated: (i) cages to restrict adult dispersal and exclude flying predators; (ii) plant-free zones around the cages to exclude crawling predators; (iii) enhancement of the nutritional quality of waterlettuce in the cages; and (iv) frequent inoculation of the cages with laboratory-reared insects. Use of this strategy resulted in several provisionally-established S. pectinicornis populations, numbering as much as 23.5 S. pectinicornis individuals m -2 , and produced up to seven post-release generations. Unfortunately, all of these incipient populations declined until S. pectinicornis was undetectable. We conclude that field populations of this moth no longer persist in Florida, and offer an introspective assessment of how this project might have been improved.  相似文献
