The contribution of microtubules and microfilaments to the cytomechanics of transverse nuclear centering were investigated in the charophycean green alga Spirogyracrassa (Zygnematales). Cytoplasmic strands of enhanced rigidity and fasciate appearance radiate from the rim of the lenticular nucleus through the vacuole, frequently split once or twice and are attached to the helical chloroplast bands in the peripheral cytoplasm. The nucleus is encased in tubulin and a web of F-actin. Bundles of microtubules, emerging from the nuclear rim, are organized into dividing fascicles within the strands and reach to the inner surface of the chloroplast envelope. Organelles are translocated in both directions along similarly arranged fascicles of microfilament bundles which extend from the nucleus to the peripheral actin cytoskeleton. Application of microtubule- and/or microfilament-depolymerizing drugs affected the position of the nucleus only slowly, but in distinct ways. The differential effects suggest that nuclear centering depends on the tensional integrity of the perinuclear scaffold, with microfilaments conveying tension along stabilized microtubules and the actin cytoskeleton integrating the translocation forces generated within the scaffold. Summary A plexus of nerve fibers encompassing neuronal perikarya is present within the gill filament; it surrounds the proximal portion of the efferent filament artery and the efferent lamellar arterioles. This innervation resembles the pattern described for the area around the sphincter of the efferent filament artery: acetylcholinesterase-positive neurons and fibers, fast-fading yellow-fluorescent neurons and fibers, long-lasting green-fluorescent fibers. In addition, synaptic contacts between the different components suggest functional interrelationships. Nerves evidently control the efferent limb of the filament circulation including the sphincter of the efferent filament arteries, the proximal portion of the efferent filament arteries proper, and their corresponding efferent lamellar arterioles. Summary The paired external glomus of the fully developed pronephros has been studied in early larvae (ammocoetes) of 2 lamprey species, Lampetra fluviatilis and Petromyzon marinus, several weeks after hatching and newly hatched, by use of light-, scanning (SEM) and transmission (TEM) electron microscopy. Three weeks after hatching the glomus is a complex of capillary loops supplied by a single arteriole branching from the aorta. The glomus consists of 3 cell types: podocytes, fenestrated endothelium, and mesangial cells. A basement membrane, which has a close contact to the podocytes, is the only continuous barrier between blood and the coelomic cavity. The glomus exhibits all fine-structural elements known to be essential for function in the glomeruli of other vertebrates. We therefore assume the pronephric glomus of lampreys to be functional in ultrafiltration, with the ultrafiltrate released into the coelomic cavity. In newly hatched larvae, the structure of the glomus is not fully developed. In this earlier stage several afferent arterioles supply each glomus. The endothelial cells in the glomar capillaries still lack regular epithelial organization and resemble mesenchymal cells. However, the presence of typical podocytes stretching over a continuous basement membrane suggests that the tissue is already capable of ultrafiltration.This paper is dedicated to the memory of Professor W. A phragmosome-like perinuclear scaffold attaches to helical chloroplast bands in the peripheral cytoplasm of Spirogyra crassa Kützing by rigid, stalk-like cytoplasmic strands. Balanced integration of forces within this scaffold by microfilament-based tension conveyed along stabilized microtubules (MTs) within the stalks controls transverse centering of the lenticular nucleus in the cylindrical cell prior to mitosis. Nuclei with their associated scaffolds were released from rapidly frozen, shattered cells upon thawing in a MT-stabilizing buffer and could be purified by a procedure combining filtration, differential centrifugation, and isopyknic centrifugation in a linear density gradient. In the osmotically indifferent gradient medium Nycodenz™, nuclei with adhering scaffolds attained a buoyant density of 1.26–1.27 g·cm−3. The nuclei maintained their energetically unfavorable, lenticular shape in vitro and appeared structurally preserved as judged by video-enhanced differential interference contrast and by fluorescent staining of DNA with 4'-6'-diamidino-2-phenylindole. The stalks of the scaffold, though still radiating from the nuclear rim, lost their straightness and were often severed during the isolation process. The distal ends of many unsevered stalks and of stalk fragments were still associated with chloroplast fragments. Persistent flexural rigidity of the scaffold stalks and indirect immunofluorescence indicated preservation of MTs. In addition to tubulin, both actin and an intermediate filament antigen of M r ≈ 68 kDa were detected in the isolated nuclei with associated scaffolds by immunoblot with monoclonal antibodies.  相似文献

The influence of small-scale differences in resource origin on trophic position estimates was evaluated with the stable isotopes method. Perch Perca fluviatilis , a widespread freshwater predator, was used as a model organism. High individual variability was found in the contribution of benthic resources to the isotopic signatures of perch caught in the littoral zone, suggesting substantial within-habitat individual specialization. Small-scale resource origin and δ13C enrichment should be accounted for to avoid misleading estimates of both the absolute values and the ontogenetic trajectories of trophic position. A conceptual framework using end-member pathways is proposed to estimate trophic position with the stable isotope technique, particularly when marked ontogenetic niche shifts are expected.  相似文献

1.  Conservation of freshwater animal populations requires their access to, as well as sufficient availability of, critical habitats, such as those for reproduction. Abundant small-scale barriers may cause extensive fragmentation of freshwater habitat but, by comparison to larger structures their effects are rarely considered by catchment managers. The relationship between the distribution of, and access to, spawning habitat in a regulated river, characterized by abundant small barriers, was examined for river lamprey Lampetra fluviatilis , a threatened migratory fish.
2.  Telemetry of adult lamprey in the River Derwent, North East England was used to quantify upriver migration and access to spawning habitat, together with surveys of spawning habitat availability and spawning activity between 2002 and 2007.
3.  Access in to the Derwent appeared severely restricted by a tidal barrage, beyond which lamprey migrated rapidly in unobstructed reaches. Of all lamprey tagged in the lower 4 km of river, or ascending the barrage, 64% and 17% passed the first and second weirs respectively, with high flows crucial for this. Although over 98% of lamprey spawning habitat occurred more than 51 km upstream, on average just 1.8% of river lamprey spawners were recorded there.
4.  In order to protect or rehabilitate species or species assemblages, greater attention needs to be paid to the relative spatial distribution of low-head barriers and the resultant availability of key habitats within individual catchments. This is particularly important given the renewed emphasis internationally on low-head hydropower solutions as a source of renewable energy, and the rapid growth in numbers of low-head barriers in many catchments.  相似文献

We developed and characterized 13 microsatellite markers from the water mite Hygrobates fluviatilis. The genotypes of 32 diploid females were assessed, and all of the loci were polymorphic. The number of alleles per locus ranged from two to 21, and the observed heterozygosity ranged from 0.063 to 0.813. These microsatellite markers are the first published for water mites and will contribute to research on the population structure of this widespread species.  相似文献

The life-history characteristics of the river blenny Salaria fluviatilis in a Mediterranean stream in north-east Spain have been studied. The maximum age observed was 4+ years in both sexes, although only age groups 0+ and 1+ years were well represented. For both sexes, growth was rapid between April and June (before the summer drought) and in October. The instantaneous rate of monthly growth (April to October) was positively correlated with water depth. All fish attained sexual maturity during their first year of life. Females spawned multiple times. Spawning occurred mainly from June to August, although older females (2+ years old) began in May. The relationship between fecundity and total length recorded for older females in the spring was not significantly different from that obtained for 1 year-old females in the summer. The mean diameter of ripe oocytes, however, was greater in older females. Larger eggs that were laid during a period of good food supply (spring) might have a higher probability of survival. The results suggest that female fecundity is affected by environmental conditions. The species is characterized by fast growth, early maturity, multiple spawning and limited longevity. This may be a good life-history strategy for small fish species in unstable environments, where adult mortality is high, variable or unpredictable. Some life-history traits of the river blenny suggest that this species might be vulnerable to more severe drought conditions than those associated with the Mediterranean climate.  相似文献

Summary.  Some species of Spirogyra form rosette-shaped or rod-shaped rhizoids in the terminal cell of the filaments. In the present study, we analyzed an involvement of microtubules (MTs) in rhizoid differentiation. Before rhizoid differentiation, cortical MTs were arranged transversely to the long axis of cylindrical cells, reflecting the diffuse growth. At the beginning of rhizoid differentiation, MTs were absent from the extreme tip of the terminal cell. In the other area of the cell, however, MTs were arranged transversely to the long axis of the cell. In the fully differentiated rosette-shaped rhizoid, MTs were randomly organized. However, at a younger stage of rosette-shaped rhizoids, MTs were sometimes arranged almost transversely in the lobes of the rosette. In the rod-shaped rhizoid, MTs were arranged almost transversely. MT-destabilizing drugs (oryzalin and propyzamide) induced swelling of rhizoids, and neither rosette-shaped nor rod-shaped rhizoids were formed. The role of MTs in rhizoid differentiation was discussed.
