The loading of amino acids and nitrate into the xylem was investigated by collection and analysis of root-pressure exudate from the cut hypocotyl stumps of seedlings of Ricinus communis L. Glutamine was found to be the dominant amino acid in the exudate and also to be the amino acid which is transferred to the xylem most rapidly and accumulated to the greatest extent. The comparison between uptake and xylem loading showed significant differences in specificity between these two transport reactions, indicating a different set of transport systems. Nitrate is transferred to the xylem at a higher relative rate than any amino acid despite the great nitrate-storage capacity of the root system. Thus the supply of nitrate to Ricinus plants leads to enhanced nitrogen allocation to the shoots.  相似文献

As fresh human cadaveric spine specimens for in vitro testing are hard to obtain and carry a potential risk of infection, the possibility of using embalmed spine specimens has been considered. The cross-linking effect of formalin fixation, however, raises uncertainties regarding the biomechanical likeness of preserved specimens. They have been reported to be stiffer, but no quantitative data exist.

The purpose of this study was to determine the biomechanical differences between fresh and formalin-fixed spine specimens, using L1–2 motion segments from six 16-week-old calf spines. The range of motion and neutral zone were determined in flexion-/extension, left/right axial rotation, and right/left lateral bending.

The range of motion decreased in the formalin fixed specimens by as much as 80%, and the neutral zone by as much as 96%. The results of this study therefore imply that, for biomechanical testing, formalin-fixed specimens are not representative of the in vivo conditions.  相似文献

The ecosystem response model described in this paper combines an ecosystem model and a three-dimensional circulation model of Lake Ladoga developed earlier by the authors. The ecosystem model describes the process of Lake Ladoga eutrophication, and its biological submodel describes changes in the phyto- and zooplankton. In the earlier model version, lake circulation was determined using a two-dimensional hydrodynamical model which was not completely adequate. The present model allows calculation of the distributions of phyto- and zooplankton and mineral phosphorus and nitrogen. One of its main advantages is that reliable computations of the ecosystem dynamics over an extended period of time are possible. The response of the ecosystem to different levels of phosphorus pollution loading and to weather conditions is studied.  相似文献

Barley leaf protoplasts were incubated in light or darkness in the presence of various inhibitors, metabolites or weak acids/bases. Nitrate reductase (NR) and phosphoenolpyruvate carboxylase (PEPCase) were rapidly extracted from the protoplasts and assayed under sub-optimal conditions, i.e. in the presence of Mg2+ and malate, respectively. Under these conditions changes in activities are thought to reflect changes in the phosphorylation states of the enzymes. The NR was activated by illumination to 90% of its maximal activity within 10 min. Photosynthetic electron transport appeared necessary for light activation of NR since activation was inhibited by the photosynthetic electron-transport inhibitor 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), and, additionally, an electron acceptor (HCO 3 - ) was required. The PEPCase was also activated by light. However, this activation was not prevented by DCMU or lack of HCO 3 - . Loading of protoplasts in the dark with a weak acid resulted in activation of both NR and PEPCase. For NR, full activation was completed within 5 min, whereas for PEPCase a slower, modest activation continued for at least 40 min. Incubation of protoplasts with a weak base also gave activation of PEPCase, but not of NR. On the contrary, base loading counteracted light activation of NR. Large, intermediate, and small bundles and contiguous tissues of the leaf blade of Hordeum tvulgare L. ‘Morex’ were examined with the transmission electron microscope to determine their cellular composition and the distribution and frequency of the plasmodesmata between the various cell combinations. Plasmodesmata are abundant at the mesophyll/parenchymatous bundle sheath, parenchymatous bundle sheath/mestome sheath, and mestome sheath/vascular parenchyma cell interfaces. Within the bundles, plasmodesmata are also abundant between vascular parenchyma cells, which occupy most of the interface between the sieve tube-companion cell complexes and the mestome sheath. Other vascular parenchyma cells commonly separate the thick-walled sieve tubes from the sieve tube-companion cell complexes. Plasmodesmatal frequencies between all remaining cell combinations of the vascular tissues are very low, even between the thin-walled sieve tubes and their associated companion cells. Both the sieve tube-companion cell complexes and the thick-walled sieve tubes, which lack companion cells, are virtually isolated symplastically from the rest of the leaf. Data on plamodesmatal frequency between protophloem sieve tubes and other cell types in intermediate and large bundles indicate that they (and their associated companion cells, when present) are also isolated symplastically from the rest of the leaf. Collectively, these data indicate that both phloem loading and unloading in the barley leaf involve apoplastic mechanisms.  相似文献

Spinach plants (Spinacea oleracea L. cv. Estivato) were grown on nutrient solutions under deficient, normal and excess sulfate supply. In both young and mature plants net uptake of sulfate and its transport to the shoot increased with increasing sulfate supply, but both processes proceeded at a higher rate in young as compared to mature plants. The relative sulfate transport, i.e. the relative amount of the sulfate taken up that is transported to the shoot, decreased with increasing sulfate supply. Apparently, net uptake of sulfate is not strictly controlled by the sulfur demand of the shoot, but xylem loading appears to counteract excess transport of sulfate to the shoot. Fumigation with H2S or SO2 reduced net uptake of sulfate by the roots in sulfur-deficient plants and absolute as well as relative sulfate transport to the shoot independent of the three sulfate levels supplied to the plant. At the same time thiol contents of the shoot and the root were enhanced by fumigation with H2S and SO2. These findings are consistent with the idea that thiols produced in the leaves can mediate demand-driven control of sulfate uptake by the roots and its transport to the shoot.  相似文献

Lakes and reservoirs provide water for human needs and habitat for aquatic birds. Managers of such waters may ask whether nutrients added by waterfowl degrade water quality. For lakes and reservoirs where primary productivity is limited by phosphorus (P), we developed a procedure that integrates annual P loads from waterfowl and other external sources, applies a nutrient load-response model, and determines whether waterfowl that used the lake or reservoir degraded water quality. Annual P loading by waterfowl can be derived from a figure in this report, using the days per year that each kind spent on any lake or reservoir. In our example, over 6500 Canada geese (Branta canadensis) and 4200 ducks (mostly mallards, Anas platyrhynchos) added 4462 kg of carbon (C), 280 kg of nitrogen (N), and 88 kg of P y–1 to Wintergreen Lake in southwestern Michigan, mostly during their migration. These amounts were 69% of all C, 27% of all N, and 70% of all P that entered the lake from external sources. Loads from all external sources totaled 840 mg P m–2 y–1. Application of a nutrient load-response model to this concentration, the hydraulic load (0.25 m y–1), and the water residence time (9.7 y) of Wintergreen Lake yielded an average annual concentration of total P in the lake of 818 mg m–3 that classified the lake as hypertrophic. This trophic classification agreed with independent measures of primary productivity, chlorophyll-a, total P, total N, and Secchi disk transparency made in Wintergreen Lake. The impact of agriculture was estimated on two shallow, eutrophic lakes, Lake Kotojärvi and Lake Villikkalanjärvi in southern Finland. The main emphasis was on phosphorus and nitrogen budgets and on the phytoplankton dynamics. Special attention was paid to internal P loading and blue-green algal blooms. The mean Tot-P load from agricultural land was 1.2 kg ha-1 a-1 in both basins and Tot-N loads were 19 kg ha-1 a-1 in L. Villikkalanjärvi and 12 kg ha-1 a-1 in L. Kotojärvi. The Tot-P input to L. Kotojärvi was on an average 0.62 g m-2 a-1 (per lake surface area), and the Tot-N input 9.1 g m-2 a-1. The corresponding inputs to L. Villikkalanjärvi were 3.1 and 57 g m-2 a-1, respectively. The annual variation followed the runoff volumes. About half of the Tot-P and one third of the Tot-N load was retained in L. Kotojärvi. In L. Villikkalanjärvi the retention was only 24% for Tot-P and 19% for Tot-N. The difference was very probably due to a longer theoretical retention time in L. Kotojärvi. In L. Villikkalanjärvi the mean concentration of Tot-P was 120 µg 1-1 and that of Tot-N 1700 µg 1-1 and the corresponding figures in L. Kotojärvi 67 and 990 µg 1-1, respectively. The mean chlorophyll a concentration was, however, higher in L. Kotojärvi (26 µg 1-1) than in L. Villikkalanjärvi (20 µg 1-1). This was probably due to an internal P load in L. Kotojärvi: in 1988 the internal load of dissolved P was estimated to be as much as twofold the external load. In L. Villikkalanjärvi the internal dissolved P load was only up to 50% of the external input. In L. Kotojärvi the high internal P load coupled with a low DIN:DIP ratio resulted in a strong blue-green algal bloom in the summer of 1988. In L. Villikkalanjärvi blue-green algae were observed only in small amounts. Even in August 1990, when the DIN:DIP ratio was low enough to favor the occurrence of blue-green algae, they contributed only up to 10–15% of the total phytoplankton biomass.  相似文献

Summary InChaetoceros peruvianus, the two very long, delicately tapered setae (spine-like processes) of the lower valve curve downwards gently until they are often almost parallel, while those emerging from the upper valve curve sharply downwards until oriented almost in the same direction as the setae of the lower valve. This curvature creates a deep pit between the bases of the upper valve's setae, where they emerge from the valve. In live cells, extension of setae is rapid and very sensitive to disturbance. After cleavage the new silica deposition vesicle (SDV) appears in the centre of the furrow and expands outwards over it. A distinct microtubule centre (MC) appears directly on top of the SDV. Microtubules (MTs) from the MC ensheath the nucleus, and others fan out over the SDV and plasmalemma. A little later, the MC in the lower daughter cell moves off the SDV, and its MTs now appear to mould the plasmalemma/ SDV into the deep pit between the base of the setae. In the upper daughter cell, the MC remains on the SDV. Initiation of setae is first observed as protuberances of bare cytoplasm growing from the sides of the daughter cells, through gaps in the parental valve. Many MTs initially line the plasmalemma of these protuberances as they grow outwards and the SDV also expands over the new surface. As the setae get longer, a unique complex of three organelles appears. Just behind the naked cytoplasm at the tip of the seta, a thin flat layer of fibrous material lines the plasmalemma. This, the first of the complex, is called the thin band. Immediately behind this is the second, a much thicker, denser fibrous band, the thick band. At the front edge of the SDV, 5–6 finger-like outgrowths of silicified wall grow forwards. These are interconnected by the elements of the thick band which thus apparently dictate the polygonal profile of the seta. These also appear to generate the spinules (tiny spines) that adorn the surface of the seta; the spiral pattern of the spinules indicates that this whole complex might differentiate one after the next, in order. Further back from the tip, evenly spaced transverse ribs are formed. These are connected to the third organelle in the complex, the striated band; our interpretation is that the striated band sets up the spacing of the ridges that regularly line the inner surface of the setae. During seta growth, this complex is apparently responsible for controlling the delicate tapering curvature of the very fine silica processes. Since the complex is always seen near the tip of the seta, we conclude that it migrates forwards steadily as the tip grows. While the thin and thick bands could slide continuously over the cell membrane, the striated band must be disassembled and then recycled forward during extension if it is indeed connected to the ridges lining the inside of the setae. We could find no indication that turgor pressure drives extension of the setae, in which event the activity of these organelles is responsible for growth using the justformed silica tube as the base from which extension is generated.  相似文献

The effect of shock-loading of zinc, copper and cadmium ions on the removal of total organic carbon (TOC) and phosphate in an anaerobic-aerobic activated sludge process was investigated. TOC removal was not sensitive to shock-loading of Zn2+ and Cd2+ ions, and complete removal was achieved even at 20 mg Zn2+/l and 20 mg Cd2+/l. However, with over 1 mg Cu2+/1 TOC removal efficiency decreased. PO inf4 sup3- removal, in contrast, was extremely sensitive to these metal ions, with the threshold being 1 mg Zn2+/l and 1 mg Cd2+/l. Higher concentrations adversely affected PO inf4 sup3- removal. Copper again proved detrimental; no PO inf4 sup3- removal was achieved even at 1 mg Cu/l. These results highlight the sensitivity of the removal efficiencies of TOC and PO inf4 sup3- to shock loadings of these heavy metals.Y.P.
