研究了用超滤技术分离大豆乳清中的蛋白质和异黄酮的工艺条件. 结果表明,大豆乳清在超滤之前要进行预处理以减轻膜污染;通过2因素3水平正交试验,确定了最佳的预处理工艺:按大豆乳清中固形物含量的5%,向其中加入CaCl2,并在85 ℃下加热15 min,在此条件下,蛋白质的沉淀率为49.8%,异黄酮的保留率为90.4%;通过单因素试验,确定了比较合适的超滤条件:选择切割分子量(MWCO)为10000的聚醚砜膜,超滤压力选择51~68 kPa,超滤温度选择30 ℃~40 ℃.在此条件下,大豆乳清中蛋白质的截留率为83.9%,而异黄酮的截留率为7.6%. 相似文献

The impact of succinylation on soy proteins as excipients for delayed delivery of drugs in the gastrointestinal tract was studied. Succinylation decreased protein solubility and protein charge density at pH 1.2 and increased solubility and zeta potential at pH above 4.5. Tablet erosion and swelling were decreased at pH 1.2 and increased at pH 7.5. FTIR analysis indicated polypeptide chain unfolding as a result of succinylation. Tablets of protein succinylated 50% or 100% released less than 10% of loaded riboflavin or rifampicin in 2 h at gastric pH in the presence of pepsin but released these compounds rapidly at intestinal pH. Succinylated soy protein tablets were thus gastroresistant, suggesting their use as excipients for controlled release of medicinal or nutraceutical agents. 相似文献

The aim of the present study was to evaluate the protective effects of isoflavone (ISO) against zearalenone (ZEA) residues in the muscle and liver tissues of prepubertal gilts. Seventy 75-day-old, prepubertal, female pigs (Duroc × Landrace × Yorkshire, 26.5 ± 0.60 kg) were allocated randomly to seven diet treatments for 21days as follows: one control group (fed the basal diet) and six groups fed the basal diet with the addition of either 0.5 or 2.0 mg/kg ZEA plus either 0, 300 or 600 mg/kg ISO. The results showed that the diet with 2.0 mg/kg ZEA added caused an increase of ZEA residue level in muscle tissue (P < 0.05), and that the addition of both 0.5 and 2.0 mg/kg ZEA increased the residue level of ZEA in the liver of prepubertal gilts (P < 0.05). Addition of 600 mg/kg ISO to 2.0 mg/kg ZEA-contaminated diet decreased the ZEA residue level in liver tissue (P < 0.05), and the addition of 300 or 600 mg/kg ISO to the 2.0 mg/kg ZEA-contaminated diet decreased the residue levels of ZEA in muscle tissue (P < 0.05). Western blot analysis demonstrated that feeding ZEA to prepubertal gilts increased their protein expression of 3α/3β-hydroxysteroid dehydrogenase (HSD; P < 0.05), and that the addition of 300 or 600 mg/kg ISO to the 2.0 mg/kg ZEA-contaminated diet decreased the protein expression of 3α/3β-HSD (P < 0.05), compared with the addition of 2.0 mg/kg ZEA alone. The results demonstrated that muscle and liver tissues retain residual ZEA when pigs are fed a diet contaminated with high concentrations of ZEA, and that the concentration of ZEA in muscle and liver tissues increased with increased amounts of ZEA in the feed. In diets contaminated with high levels of ZEA, the addition of ISO may accelerate the biotransformation and degradation of ZEA and its metabolites, and reduce the residues of ZEA in liver and muscle tissues of prepubertal gilts. 相似文献

A diet containing a high concentration of soy isoflavone was administered to laying hens and the contents of the isoflavones transferred to the plasma and egg yolk were measured. A method for quantitatively measuring the concentration of isoflavone in the yolk was first established, before a high concentration of soy isoflavone was administered to the laying hens over an 18-day period. The concentrations of isoflavone in the plasma and egg yolk reached their highest on the 12th day of the feeding period, the values being 3,167 nmol/l and 65.29 μg/100g, respectively. The concentration of cholesterol in the yolk was slightly affected during the early stages of the feeding period. These findings clearly demonstrate that soy isoflavone was transferred into the yolk from the feed and that the cholesterol concentration in the yolk was affected by administering the soy isoflavone-enriched feed. 相似文献

Production of polyols such as erythritol, d-mannitol and d-arabitol by citric acid-producing yeasts occurred only when the medium-pH was controlled at acidic pH, as described in the previous papers.In order to elucidate the conversion mechanism of citric acid fermentation to polyol fermentation, the effect of pH on the activities of enzymes involved in polyol synthesis and tricarboxylic acid cycle was studied. Shifting down of the medium-pH from 5.5 to 3.5 led immediately to the change of intracellular pH, from 6.5~6.7 to 5.5~5.7. Such the change affected remarkably on the activities of intracellular enzymes. Citrate synthase was significantly depressed at pH 5.7, but isocitrate lyase and phosphoenolpyruvate carboxykinase were reversely stimulated at this pH.In some yeast strains incapable of polyol production, the change of medium-pH reflected directly on intracellular pH, whereby almost all enzymes were inhibited.From these results, the conversion of citric acid production to polyol production was explained by the change in the enzyme activities caused by lowering of intracellular pH. 相似文献

Bacterial neuraminidase (NA) is one of the key enzymes involved in pathogenesis of inflammation during infection. The organic extract of the roots of Flemingia philippinensis showed high bacterial NA inhibitory activity with an IC50 of around 5 μg/mL. Activity-guided separation of the methanol extract yielded nine prenylated isoflavones together with the novel species isoflavone (2) which was given the name flemingsin. Isolated prenylated isoflavones (1–9) were evaluated for NA inhibition and their IC50 values were determined to range between 0.30 and 56.8 μM. The most potent inhibitor 4 (IC50 = 300 nM, Ki = 130 nM) features a catechol motif in the B-ring and a furan in the A-ring. Structure–activity analysis also showed a 4-hydroxyl group within the B-ring was essential for NA inhibitory activity, because isoflavone (9) having protected 4-hydroxyl group was much less potent than its hydroxylated counterpart. All neuraminidase compounds screened were found to be reversible noncompetitive inhibitors. Furthermore, the most active NA inhibitors (1–9) were proven to be present in the native roots in high quantities by HPLC and LC-DAD-ESI/MS. 相似文献

The ability of a soy-based high-phytoestrogen diet (nutritional intervention) or genistein (pharmacological intervention), to limit ischemic brain damage in Wistar, Wistar-Kyoto (WKY) and spontaneously hypertensive (SHR) rats, has been assessed. As to the nutritional intervention, two groups from each strain received either a phytoestrogen-free (PE-0) or a high-phytoestrogen (PE-600) diet from weaning to adulthood. As to the pharmacological intervention, all animals were fed the standard soy-free AIN-93G diet and subsequently separated into two groups from each strain to receive either pure genistein (aglycone form, 1 mg/kg/day intraperitoneal) or vehicle at 30 min reperfusion. After an episode of 90 min ischemia (intraluminal thread procedure) followed by 3 days reperfusion, cerebral infarct volume was measured. Arterial blood pressure (ABP) was significantly higher at the basal stage (just before ischemia) in SHR (140 ± 7 mmHg, n = 17, p < 0.05) than in Wistar (113 ± 4 mmHg, n = 23) and WKY (111 ± 6 mmHg, n = 14) rats. No significant differences were shown among the three stages (basal, ischemia, reperfusion) within each rat strain for both PE-0 and PE-600 diets. Wistar, but not WKY or SHR, rats fed the PE-600 diet showed significantly lower infarct volumes than their counterparts fed the PE-0 diet (30 ± 3% vs. 17 ± 3%, p < 0.01). Genistein-treated Wistar, but not WKY or SHR, rats showed significantly lower infarct volumes than their vehicle-treated controls (27 ± 2% vs. 15 ± 2%, p < 0.01). Our results demonstrate that: (1) the neuroprotective action of either chronic or acute exposure to soy isoflavones is strain-dependent, since it was shown in Wistar but not WKY or SHR rats; and (2) the soy-based diet does not prevent development of hypertension in SHR rats. 相似文献
