自来水中人肠道病毒的存在已引起人们极大的关切和忧虑。本文报道了武汉东湖水和以东湖为水源的自来水中病毒和指示细菌的存在水平。水源水经过预加氯消毒、絮凝沉淀、砂滤和最后加氯消毒处理而成的自来水中细菌总数检测范围是41—500/升,总大肠菌是2—13/升、粪大肠菌为0—4/升、大肠菌噬菌体是0—13.6PFU/升,平均2.48PFU/升,肠道病毒为0—78PFU/升,平均为6.7PFU/升。水源水经制水工艺处理去除指示细菌、大肠菌噬菌体和肠道病毒的效率分别为97.95—99.99%,90.63%和53.18%。这种结果说明自来水制水工艺能有效地降低指示细菌,大肠菌噬菌体,而去除肠道病毒效率较低,揭示肠道病毒对环境压专的耐受性明显地比指示细菌强。 相似文献

Abstract Identifying the source effect on heavy metals to human health risk is essential for devising and implementing restoration policies for polluted soils. For this purpose, eight heavy metals (As, Cd, Hg, Cr, Cu, Ni, Pb, and Zn) in soil profile samples (0–10, 10–20, 20–30, and 30–40?cm) collected in the area around aluminum-plastic manufacturing facilities (APMF) were determined. An absolute principal component score multiple linear regression (APCS-MLR) model supported by a health risk assessment (HRA) model was developed to determine the source apportionment of soil heavy metals and contribution rate of pollution sources to human health risk. Results showed significant accumulations of eight metals in the topsoil (0–20?cm), parent material, transportation, APMF, and agricultural practices were the four major contributing sources for heavy metals in soils, with average contribution percentages of 21.69%, 24.99%, 29.60%, and 14.25%, respectively. Carcinogenic risk factors for adults (1.23E-04) and children (1.32E-04) were found to be above the acceptable level (1E-06 to 1E-04). The health risk quantification results indicated that parent material, APMF, transportation, agricultural practices, and unidentified factors accounted for 55.76%, 14.48%, 12.09%, 10.13%, and 7.54% of the carcinogenic risk for children and adults. The adverse impacts of Cd, Zn, and Pb accumulations in soil coming from APMF activities were significant and need to receive more attention. 相似文献

Rapid auditory processing and acoustic change detection abilities play a critical role in allowing human infants to efficiently process the fine spectral and temporal changes that are characteristic of human language. These abilities lay the foundation for effective language acquisition; allowing infants to hone in on the sounds of their native language. Invasive procedures in animals and scalp-recorded potentials from human adults suggest that simultaneous, rhythmic activity (oscillations) between and within brain regions are fundamental to sensory development; determining the resolution with which incoming stimuli are parsed. At this time, little is known about oscillatory dynamics in human infant development. However, animal neurophysiology and adult EEG data provide the basis for a strong hypothesis that rapid auditory processing in infants is mediated by oscillatory synchrony in discrete frequency bands. In order to investigate this, 128-channel, high-density EEG responses of 4-month old infants to frequency change in tone pairs, presented in two rate conditions (Rapid: 70 msec ISI and Control: 300 msec ISI) were examined. To determine the frequency band and magnitude of activity, auditory evoked response averages were first co-registered with age-appropriate brain templates. Next, the principal components of the response were identified and localized using a two-dipole model of brain activity. Single-trial analysis of oscillatory power showed a robust index of frequency change processing in bursts of Theta band (3 - 8 Hz) activity in both right and left auditory cortices, with left activation more prominent in the Rapid condition. These methods have produced data that are not only some of the first reported evoked oscillations analyses in infants, but are also, importantly, the product of a well-established method of recording and analyzing clean, meticulously collected, infant EEG and ERPs. The major allergens of honeybee venom, hyaluronidase (Hyal) and phospholipase A2, can induce life-threatening IgE-mediated allergic reactions in humans. Although conventional immunotherapy is effective, up to 40% of patients develop allergic side effects including anaphylaxis and thus, there is a need for an improved immunotherapy. A murine monoclonal anti-Hyal IgG1 antibody (mAb 21E11), that competed for Hyal binding with IgEs from sera of bee venom allergic patients, was raised. The fragment of these IgG antibodies which bind to antigen (Fab) was produced and complexed (1:1) with Hyal. The crystal structure determination of Hyal/Fab 21E11 complex (2.6 A) enabled the identification of the Hyal-IgG interface which provides indirect information on the Hyal-IgE interaction (B-cell epitope). The epitope is composed of a linear array of nine residues (Arg138, His141-Arg148) located at the tip of a helix-turn-helix motive which protrudes away from the globular core and fits tightly into the deep surface pocket formed by the residues from the six complementarity determining regions (CDRs) of the Fab. The epitope is continuous and yet its conformation appears to be essential for Ab recognition, since the synthetic 15-mer peptide comprising the entire epitope (Arg138-Glu152) is neither recognized by mAb 21E11 nor by human IgEs. The structure of the complex provides the basis for the rational design of Hyal derivatives with reduced allergenic activity, which could be used in the development of safer allergen-specific immunotherapy. 相似文献

以三江源区不同建植期人工草地中典型杂草甘肃马先蒿为研究对象,以建植前为对照,比较建植期为5年、10年的垂穗披碱草人工草地中马先蒿形态特征、器官生物量和资源投资状况.结果表明:(1)甘肃马先蒿在不同建植期人工草地中个体形态特征如株高、花柱高、花数目存在显著差异,而根系长、分枝数、叶片数差异不显著;不同阶段各器官生物量发生改变,根系生物量、茎叶生物量、繁殖器官生物量变化差异显著,而总生物量无显著差异.(2)综合建植前与建植后5、10年这3个时期样地中甘肃马先蒿在形态特征与资源投资状况的变化特征发现:总生物量、茎叶生物量、繁殖器官生物量、花数都呈现先下降再上升的V"字形变化趋势,根系生物量、株高逐渐增大,而花序长度则持续下降;资源相对投资比例也随建植期的不同表现出明显的差异,地上部分和地下部分的相对投资随时间的增加而上升,繁殖投资则表现出逐年降低的趋势.(3)以总生物量为个体大小参数,在同一时期内甘肃马先蒿各器官的绝对生物量具有明显的大小依赖性,随着个体大小的增加,茎叶生物量、根系生物量和花生物量均显著增加.但不同建植期人工草地中各器官生物量的个体大小依赖性响应程度不同,其中茎叶生物量和根系生物量的大小依赖程度随建植年限的增加而增加,而花生物量的个体大小依赖性却随建植期的延长而减弱. 相似文献

GeoCAT is an open source, browser based tool that performs rapid geospatial analysis to ease the process of Red Listing taxa. Developed to utilise spatially referenced primary occurrence data, the analysis focuses on two aspects of the geographic range of a taxon: the extent of occurrence (EOO) and the area of occupancy (AOO). These metrics form part of the IUCN Red List categories and criteria and have often proved challenging to obtain in an accurate, consistent and repeatable way. Within a familiar Google Maps environment, GeoCAT users can quickly and easily combine data from multiple sources such as GBIF, Flickr and Scratchpads as well as user generated occurrence data. Analysis is done with the click of a button and is visualised instantly, providing an indication of the Red List threat rating, subject to meeting the full requirements of the criteria. Outputs including the results, data and parameters used for analysis are stored in a GeoCAT file that can be easily reloaded or shared with collaborators. GeoCAT is a first step toward automating the data handling process of Red List assessing and provides a valuable hub from which further developments and enhancements can be spawned. 相似文献

In this paper I analyse the ethical implications of the two main competing methodologies in genomic research. I do not aim to provide another contribution from the mainstream legal and public policy perspective; rather I offer a novel approach in which I analyse and describe the patent-and-publish regime (the proprietary regime) led by biologist J. Craig Venter and the 'open-source' methodologies led by biotechnology Nobel laureate John Sulston. The 'open-source methodologies' arose in biotechnology as an alternative to the patent-and-publish regime in the wake of the explosion in computer technology. Indeed, the tremendous increase in computer technology has generated a corresponding increase in the pace of genomics research. I conclude this paper by arguing that while the patent-and-publish method is a transactional method based on the exchange of extrinsic goods (patents in exchange for research funds), the free and open-source methodology (FLOSS) 1 is a transformational method based on a visionary ideal of science, which leads to prioritizing intrinsic goods in scientific research over extrinsic goods. 相似文献

S-box (SAM-I) riboswitches are a widespread class of riboswitches involved in the regulation of sulfur metabolism in Gram-positive bacteria. We report here the 3.0-Å crystal structure of the aptamer domain of the Bacillus subtilis yitJ S-box (SAM-I) riboswitch bound to S-adenosyl-l-methionine (SAM). The RNA folds into two sets of helical stacks spatially arranged by tertiary interactions including a K-turn and a pseudoknot at a four-way junction. The tertiary structure is further stabilized by metal coordination, extensive ribose zipper interactions, and SAM-mediated tertiary interactions. Despite structural differences in the peripheral regions, the SAM-binding core of the B. subtilis yitJ riboswitch is virtually superimposable with the previously determined Thermoanaerobacter tengcongensis yitJ riboswitch structure, suggesting that a highly conserved ligand-recognition mechanism is utilized by all S-box riboswitches. SHAPE (selective 2′-hydroxyl acylation analyzed by primer extension) chemical probing analysis further revealed that the alternative base-pairing element in the expression platform controls the conformational switching process. In the absence of SAM, the apo yitJ aptamer domain folds predominantly into a pre-binding conformation that resembles, but is not identical with, the SAM-bound state. We propose that SAM enters the ligand-binding site through the “J1/2-J3/4” gate and “locks” down the SAM-bound conformation through an induced-fit mechanism. Temperature-dependent SHAPE revealed that the tertiary interaction-stabilized SAM-binding core is extremely stable, likely due to the cooperative RNA folding behavior. Mutational studies revealed that certain modifications in the SAM-binding region result in loss of SAM binding and constitutive termination, which suggests that these mutations lock the RNA into a form that resembles the SAM-bound form in the absence of SAM. 相似文献
