Abstract: Somatostatin-like immunoreactivity (SLI) from bovine retina was purified and its structure determined. Retinal tissue (1868 g) extracted with 3% acetic acid yielded 18.6 nmol SLI. This peptide was purified by chromatography on an affinity column made with anti-somatostatin antiserum, a reverse-phase C-18 HPLC column, and three sequential applications on a reverse-phase phenyl HPLC column. The peptide was purified 103,000-fold from the initial extract with an overall yield of 14.4%. Amino acid sequence determination by an automatic Edman degradation technique revealed the sequence to be as follows: Ser - Ala - Asn - Ser - Asn - Pro - Ala - Met - Ala - Pro - Arg - Glu - Arg - Lys - Ala - Gly - (Cys) - Lys - Asn - Phe - Phe - Trp - Lys - Thr - (Phe, Thr, Ser, Cys). The apparent identity of this peptide with somatostatin octacosapeptide (S28) purified from other mammalian tissue indicates the phylogenetic conservation of its structure and facilitates the use of the retina as a model system for studying the neurotransmitter function of somatostatin.  相似文献

Summary The response of somatotrophs to large doses of purified bovine growth hormone was examined in uninephrectomized rats drinking 1% sodium chloride in an attempt to prolong the action of the hormone. Growth hormone produced a triphasic response in the ultrastructure of somatotrophs. Uninephrectomy and salt alone induced no changes in somatotrophs as compared to unoperated animals consuming tap water. In the first phase (3 days) the cross sectional area of the cell increased, the size of the largest granule per cell as well as the number of granules per cell increased significantly. All these parameters then reached control level after 7 days of injections during a second phase. Suppression of somatotrophs by exogenous growth hormone was evident by 16 days (third phase) when the area of the cell, the size of the largest secretory granule as well as the number of granules per cell all decreased significantly. Somatotrophs apparently did not react to suppression by a lysosomal mechanism inasmuch as no increase in the number of lysosome-derived structures was seen even at 16 days.Supported by research grant HL 06975 from the National Heart and Lung Institute. Summary The cytology of the growth-hormone (GH) cells of the goldfish pituitary were examined following electrothermic lesions of the anterior praeoptic hypothalamus and telencephalon. Following lesions of the nucleus preopticus (NPO) light microscopy of the pituitary revealed a significant increase in the nuclear diameter and a degranulation of the GH cells. Lesions of the telencephalon anterior or dorsal to the NPO had no cytological effect on the GH cells. The ultrastructural appearance of the GH cells of NPO-lesioned fish was characterized by a marked degranulation of the cytoplasm and a proliferation of the rough endoplasmic reticulum indicative of enhanced secretory activity. The GH cells of the proximal pars distalis (PPD) are directly innervated by peptidergic (type A) and aminergic-like (type B) neurosecretory axons. Following lesions of the NPO, there was a marked reduction in the number of type A fibers in the PPD. These results suggest that the type A fibers innervating the GH cells originate in the NPO and act to inhibit the secretory activity of the GH cells.  相似文献

Growth hormone deficiency is linked to cardiovascular disease and particularly increased peripheral vascular resistance. Surprisingly, its role in endothelial nitric oxide (NO) synthetase (eNOS) regulation and NO release is basically unknown. We therefore studied the effects of different doses of somatotropin in cultures of a human endothelial cell line (EAhy926). We investigated expression and activity of eNOS, as well as other target genes known to be deregulated in cardiovascular disease including E-selectin and the lectin-like oxidized low density lipoprotein receptor. Treatment of cultured human endothelial cells with somatotropin resulted in significant (P<0.05) increases of eNOS gene and protein expression, as well as NO release, whereas production of intracellular reactive oxygen species was significantly reduced, at the highest somatotropin dose level. The enhanced eNOS gene/protein expression and enzyme activity correlate well. Our findings are suggestive for a novel role of growth hormone in endothelial biology, and particularly NO production.  相似文献

Exercise-induced increases in blood somatotropin (hGH) have always been considered in terms of quantity of the circulating molecules. Knowing that the hypophysis can release several GH species, we investigated the differential release in blood of total hGH (hGHT) and the main hGH variant (hGH20K) molecules in six trained male swimmers exposed to three different conditions known to favor GH release in blood: 45 min--70% maximum oxygen uptake (VO2max) bicycling and swimming, and 20 min of sauna bathing. Based on the binding specificity of hGH antibodies, hGH20K was isolated then assayed using the Nichols immunoradiometric assay system. All three experimental conditions produced significant (P less than 0.001) elevations in blood hGHT and hGH20K. In all three cases, mean blood hGH20K contribution to blood hGHT was relatively constant (11.9, SE 0.7%). Rises in rectal temperature were not statistically related to the changes in blood hGHT. This demonstration of a relatively constant elevation in hGH20K during bicycling, swimming, and sauna bathing can hardly explain the large differences in blood hGHT responses reported in literature under similar conditions.  相似文献
