Summary Cucumber seedlings were grown in a Portsmouth soil-sand system to study how varying soil clay and organic matter content might modify cucumber seedling response to ferulic acid, a reported allelopathic agent. Leaf area expansion of cucumber seedlings, soil respiration, and soil solution concentrations of ferulic acid were monitored. Leaf area, mean absolute rates of leaf expansion, and shoot dry weight of cucumber seedlings were significantly reduced by ferulic acid concentrations ranging from 10 to 70 μg/g dry soil. Ferulic acid was applied every other day, since it rapidly disappeared from soil solution as a result of retention by soil particles, utilization by microbes and/or uptake by roots. The amount of ferulic acid retained (i.e., adsorbed, polymerized,etc.) by soil particles appeared to be secondary to microbial utilization and/or uptake by roots. Varying clay (5.3 to 9.8 g/cup) and organic matter (2.0 to 0.04g/cup) contents of the soil appeared to have little impact on the disappearance of ferulic acid from soil solution under “ideal” growth conditions for cucumber seedlings unless larger amounts of ferulic acid were added to the soil; in this case 200 μg/g. The addition of ferulic acid to the soil materials substantially increased the activity of the soil microbes. This latter conclusion is based on recovery of ferulic acid from soil solution and soil respiration measurements. We investigated 12 natural forest stands in eastern Austria for soil ciliate diversity, viz., eight beech forests and two lowland and Pinus nigra forests each. The stands span a wide range of climatic (e.g., 543–1759 mm precipitation, 160–1035 m above sea-level) and abiotic (e.g., pH 4–7.4) factors. Samples were taken twice in autumn and late spring and analysed with the non-flooded Petri dish method. Species were identified in vivo, in silver preparations, and in the scanning electron microscope. A total of 233 species were found, of which 30 were undescribed, a surprising number showing our ignorance of soil ciliate diversity, even in Central Europe. Species number varied highly from 45 (acidic beech on silicate) to 120 (floodplain forest) and was strongly correlated with pH and overall habitat quality, as measured by climate, the C/P quotient (ratio of r-selected colpodean and k-selected polyhymenophorean ciliates), and the proportion of mycophagous ciliate species; multivariate analysis showed further important variables, viz., the general nutrient status (glucose, nitrogen, C/ N ratio) and microbial (urease) activity. The highest species number occurred in one of the two floodplain soils, supporting the intermediate disturbance hypothesis. The three main forest types could be clearly distinguished by their ciliate communities, using similarity indices and multidimensional scaling. Individual numbers varied highly from 135–1 (lowland forest) to 10,925 ml–1 (beech on silicate) soil percolate and showed, interestingly, a weak correlation with soil protozoan phospholipid fatty acids. Eight of the 30 new species found and a forgotten species, Arcuospathidium coemeterii (Kahl 1943) nov. comb., are described in detail, as examples of how species were recognized and soil protozoan diversity should be analyzed: Latispathidium truncatum bimicronucleatum, Protospathidium fusioplites, Erimophrya sylvatica, E. quadrinucleata, Paragonostomum simplex, Periholosticha paucicirrata, P. sylvatica, and Australocirrus zechmeisterae.  相似文献

In 2005 and 2006, air samples were collected at the base of a Douglas-fir watershed to monitor seasonal changes in the delta13CO2 of ecosystem respiration (delta13C(ER)). The goals of this study were to determine whether variations in delta13C(ER) correlated with environmental variables and could be used to predict expected variations in canopy-average stomatal conductance (Gs). Changes in delta13C(ER) correlated weakly with changes in vapor pressure deficit (VPD) measured 0 and 3-7 days earlier and significantly with soil matric potential (psi(m)) (P value <0.02) measured on the same day. Midday G (s) was estimated using sapflow measurements (heat-dissipation method) at four plots located at different elevations within the watershed. Values of midday Gs from 0 and 3-7 days earlier were correlated with delta13C(ER), with the 5-day lag being significant (P value <0.05). To examine direct relationships between delta13C(ER) and recent Gs, we used models relating isotope discrimination to stomatal conductance and photosynthetic capacity at the leaf level to estimate values of stomatal conductance ("Gs-I") that would be expected if respired CO2 were derived entirely from recent photosynthate. We compared these values with estimates of Gs using direct measurement of transpiration at multiple locations in the watershed. Considering that the approach based on isotopes considers only the effect of photosynthetic discrimination on delta13C(ER), the magnitude and range in the two values were surprisingly similar. We conclude that: (1) delta13C(ER) is sensitive to variations in weather, and (2) delta13C(ER) potentially could be used to directly monitor average, basin-wide variations in Gs in complex terrain if further research improves understanding of how delta13C(ER) is influenced by post-assimilation fractionation processes.  相似文献

Sixteen medicinal herbs were selected from a database on traditional herbal materials as well as literature on Korean plant resources. Then ethanol (70%, v/v) extracts of these herbs were tested for inhibition of the urease activity of Helicobacter pylori. The urease activity of H. pylori was strongly (82%) inhibited by extract of Forsythia suspensa Vahl. Active compounds in extract of Forsythia suspensa Vahl were first separated by batch mode solvent extraction, followed by purification by silica gel and octadecyl silica gel column chromatography using solvents of different polarity. According to NMR analysis of the last chromatographic fraction, we identified the presence of betulinic acid and oleanolic acid, which are known to have anti-inflammatory, anti-cancer, and anti-HIV viral activities.  相似文献

Knapweeds (Centaurea spp.) are damaging invaders of grasslands and other North American rangelands. A field study was conducted to determine conditions that promote diffuse knapweed (C. diffusa) emergence and establishment in a native Colorado grassland (North America). Knapweed was planted in native grassland under treatments with different opening sizes, levels of competition, knapweed seed burial and season of seeding. There was no effect of opening size where competing natives were alive, but knapweed emergence in 5- and 15-cm openings was higher than 0-cm openings where natives were killed. Reducing competition reduced fall diffuse knapweed emergence, but did not affect spring emergence. Seed burial increased knapweed emergence, but the effect varied by season. Although diffuse knapweed emergence reached 35%, only four plants survived from 3,600 seeds. This native grassland did not prevent knapweed emergence or establishment, but both were so low that rapid knapweed invasion is unlikely.  相似文献

Abstract: Substrate utilization of microbial cells extracted from soil with a 0.85% aqueous sodium chloride solution, was determined to estimate effects on soil microorganisms at the community level with microtiter plates (Biolog GN®) containing 95 different sources of organic carbon. A consistent pattern of utilized substrates was obtained after 24 h of microtiter plate incubation at 28°C. The absorbance values (OD590) obtained from a microtiter plate reader after background correction were transformed by using the average absorbance values of oxidized substrates as a threshold to distinguish between well utilized and poorly or non-utilized substrates and thereby reduce variances between replicates. Doubling times of the extracted soil microorganisms in the microtiter plates were tested with 12 substrates and ranged from 1.96 h to 3.23 h, depending on the carbon source. The carbon source utilization assay was used to assess the effects of soil inoculation with Corynebacterium glutamicum with and without a genetically engineered plasmid (pUN1; 6.3 kb), which encoded for the synthesis of the mammalian protease inhibiting peptide, aprotinin. Additionally, aprotinin itself was added at two concentrations to soil samples. An identical decrease in the number of carbon sources utilized, especially carbohydrates, occurred upon soil inoculation with both C. glutamicum strains after inoculation with 106 cells g−1 soil. This effect was only detectable during the first three weeks of incubation, as long as cell numbers of C. glutamicum (pUN1) were above 105 cfu g−1. Soil amendment with aprotinin resulted in utilization of additional substrates, most of them carbohydrates. With 0.1 mg aprotinin g−1 soil this stimulation lasted 2 days and with 10 mg g−1 it lasted for 7 days.  相似文献

Understanding patterns and processes involved in changes in soil and vegetation after agricultural abandonment is a key issue for management policies leading to land restoration and reclamation in Mediterranean semiarid environments. We selected a number of active and abandoned fields in two regions of olive groves in Catalonia (NE Spain), in order to analyze changes in certain soil properties and vegetation cover variables, as well as their interrelationships. The soil chemical properties considered, summarized into a single PCA axis, showed significant spatial (regional) differences and no temporal (successional) pattern, indicative of the strong influence of the parent rock material. The soil physical variables examined also showed significant regional variability, but such differences could be partially explained by successional changes. The variability at the landscape level and the observed successional trends for soil physical properties are interpreted as a result of both the influence of the bedrock and contrasting management practices prior to abandonment in each region. Although we did not find significant regional or successional variability in a global measure of plant cover, we did find significant and different patterns of variability for each of the main plant functional groups considered. Regional and post-agricultural variability in soil physical properties seem to be the prime factors determining the abundance of the main plant functional groups.  相似文献

为探索适合格木(Erythrophleum fordii)人工林在幼龄阶段的种植密度,在不同林分密度(2 m×1 m、2 m×2 m、2 m×3 m、3 m×3 m)的6 a生格木人工林下设置标准样地,采用土壤质量评价和灰色关联度等方法,探究不同密度下格木幼林的土壤理化与林下植被特征。结果表明,密度2 m×3 m下的林木胸径、树高最优,较最低水平高16.7%、27.9%;土壤总孔隙度最大,全N、硝态N、铵态N含量最高,灌木草本多样性最高。相关性分析表明土壤化学性质对灌木草本的多样性影响最大。不同林分密度下格木幼林土壤理化性质及林下植物多样性有显著差异,因此,选择合适的林分密度对人工林土壤肥力的可持续利用及林分的经营培育至关重要。  相似文献
