In the rat hypothalamus [3H]imipramine binding is inhibited by tricyclic and nontricyclic antidepressant drugs in a complex manner, with biphasic curves and Hill coefficients less than 1.0. 5-Hydroxytryptamine (serotonin) inhibited with high affinity a decreasing proportion of the [3H]imipramine binding sites as the [3H]imipramine concentration was raised. In the absence of sodium ions, IC50 values for the inhibition by tricyclic and nontricyclic antidepressants were increased by approximately 1,000-fold, and the inhibition curves became classically monophasic with Hill coefficients close to 1.0. These data are interpreted as suggesting that [3H]imipramine binds to two independent sites in the rat hypothalamus. One site is sodium-dependent with a high affinity for the drugs tested; the other is sodium-independent and has a low affinity for these drugs.  相似文献

Sodium arsenite (NaAsO2), at 10% of its median lethal dose, was administered to rats with and without vitamin C pretreatment. Liver microsomal fraction was isolated and the activity of Ca2+-ATPase was assayed. Sodium arsenite was found to inhibit the activity of the liver microsomal Ca2+-ATPase to 50% to that of control rats. The specific activity of the enzyme in rats administered sodium arsenite with vitamin C pretreatment was not significantly different from that of control rats.  相似文献

Aelurostrongylus abstrusus (Railliet, 1898) is a metastrongylid nematode of major clinical relevance in felids, causing aelurostrongylosis. In spite of its clinical importance in cats, the genetics, epidemiology and biology of this parasite are not entirely understood. mt DNA can provide markers for studies of these areas, but genetic data are scant for A. abstrusus and related lungworms. Here, the mt genome was amplified by long-range polymerase chain reaction (long-PCR) from a single male adult of A. abstrusus, sequenced using 454 technology and annotated using an established bioinformatic pipeline. This circular mt genome is 13,913 bp and contains two ribosomal RNA, 12 protein-coding and 22 transfer RNA genes, consistent with most other chromadorean nematodes. This genome should provide a source of markers for future investigations of the epidemiology and ecology of A. abstrusus. Molecular tools, employing such mt markers, are likely to find utility for explorations into the epidemiology, biology and systematics of this parasite, and the diagnosis of feline aelurostrongylosis.  相似文献

We used markers based on inter-simple sequence repeats (ISSR) to examine the genetic diversity of Aspergillus flavus from peanut-cropped soils in China. Of the 100 primers, 22 primers produced clear and reproducible ISSR bands, and the di-nucleotide accounted for 73% of those primers. The size of DNA fragments ranged from 100 to 2000 bp. The primer UBC 834 produced the largest number of polymorphic bands (10), followed by UBC 809, UBC 817, UBC 895, and UBC 899, which all amplified 7 polymorphic bands. Using the five primers, the tested strains were clearly separated based on genetic similarity coefficients (GSC). The range of GSC was from 0.59 to 0.90. In unweighted pair-group method with arithmetic averages (UPGMA) analysis, the A. flavus samples grouped in five clusters. The study showed that the ISSR technology is an effective molecular approach for studying diversity of A. flavus from peanut-cropped soils in China.  相似文献

Primary cortical neurons exposed to the mitochondrial toxin NaN3 (0.1–3 mM) were submitted to oxidative stress with H2O2 (30–150 μM), to mimic conditions observed in neurodegenerative disorders. The effects of such treatment on a series of parameters useful in characterizing neuronal damage were investigated: (i) the basal release of glutamate, evaluated as 3H-d-Aspartate efflux, was sharply, concentration-dependently, increased; (ii) the phosphorylation status of intracellular markers known to be involved in the neurodegenerative processes, in particular in Alzheimer disease: tau and GSK3β were increased, as well as the protein level of β-secretase (BACE1) and p35/25 evaluated by Western blotting, while (iii) the cell metabolic activity, measured with the MTT method, was reduced, in a concentration- and time-dependent manner. The latter effect, as well as tau hyperphosphorylation, was prevented both by a mixture of antioxidant drugs (100 μM ascorbic acid, 10 μM trolox, 100 μM glutathione) and by the anti-Alzheimer drug, memantine, 20 μM. Since it is well known that hippocampal cholinergic neurons are particularly affected in Alzheimer disease, the effects of NaN3 and H2O2 were also studied in electrically stimulated rat hippocampal slices, evaluating the 3H-Choline efflux, as an index of acetylcholine release. The neurotoxic treatment depressed the neurosecretory function and the mixture of antioxidant drugs, as well as memantine, were able to restore it. The neuronal damage induced by the in vitro protocol adopted in the present work displays peculiarities of neurodegenerative disorders, e.g. Alzheimer disease, underlining the role of mitochondrial failure and oxidative stress, which appear to occur upstream the neurodegenerative process; such protocol could be utilized to test the efficacy of neuroprotective treatments.  相似文献

目的:探讨奥氮平联合丙戊酸钠治疗难治性精神分裂症的疗效.方法:将80例精神分裂症患者按随机数字法分为联合用药组(奥氮平+丙戊酸钠组)40例和单用药组(奥氮平组)40例.联合用药组服用奥氮平起始量为10 mg/d,1周末加至20 mg/d,同时使用丙戊酸钠起始量为600 mg/d,最大剂量1200 mg/d.单药组服用奥氮平,用法同联合用药组.2组均为8周1疗程.对2组采用阳性与阴性症状量表(PANSS)评定疗效,采用不良反应量表(TESS)评定治疗中不良反应.于治疗2周末、4周末、8周末测定奥氮平血药浓度.结果:第8周末,两组PANSS评分较治疗前均下降(P均<0.05),联合用药组较单用药组联合用药组PANSS(P<0.001)、阳性症状(P<0.001)、阴性症状(P<0.05)均明显改善,两组不良反应差异无统计学意义(P>0.05).结论:奥氮平联合丙戊酸钠治疗难治性精神分裂症可提高疗效且安全性高.  相似文献

Sodium azide (NaN3) is known as an inhibitor of catalase, and a nitric oxide (NO) donor in the presence of catalase and H2O2. We showed here that catalase-catalyzed oxidation of NaN3 can generate reactive nitrogen species which contribute to tyrosine nitration in the presence of H2O2. The formation of free-tyrosine nitration and protein-bound tyrosine nitration by the NaN3/catalase/H2O2 system showed a maximum level at pH 6.0. Free-tyrosine nitration induced by peroxynitrite was inhibited by ethanol and dimethyl-sulfoxide (DMSO), and augmented by superoxide dismutase (SOD). However, free-tyrosine nitration induced by the NaN3/catalase/H2O2 system was not affected by ethanol, DMSO and SOD. NO-2 and NO donating agents did not affect free-tyrosine nitration by the NaN3/catalase/H2O2 system. The reaction of NaN3 with hydroxyl radical generating system showed free-tyrosine nitration, but no formation of nitrite and nitrate. The generation of nitrite (NO-2) and nitrate (NO-3) by the NaN3/catalase/H2O2 system was maximal at pH 5.0. These results suggested that the oxidation of NaN3 by the catalase/H2O2 system generates unknown peroxynitrite-like reactive nitrogen intermediates, which contribute to tyrosine nitration.  相似文献
