Ischemic heart disease is a leading cause of death worldwide. Myocardial ischemia results in reduced coronary flow, followed by diminished oxygen and nutrient supply to the heart. Reperfusion to an ischemic myocardium often augments the ischemic damage, known as ischemia-reperfusion (I/R) injury. Number of studies demonstrated that the hyperlipidemic myocardium is rather sensitive and more vulnerable to I/R-induced myocardial injury. Repeated brief ischemia and reperfusion cycles, termed as ischemic preconditioning, given before a sustained ischemia is known to reduce myocardial damage occur as a result of I/R. A plethora of evidence supports the fact that preconditioning is one of the promising interventional strategies having an ability to limit I/R-induced myocardial injury. Despite this fact, the preconditioning-mediated cardioprotection is blunted in chronic hyperlipidemic condition. This suggests that preconditioning is moderately a ‘healthy heart protective phenomenon’. The mechanisms by which chronic hyperlipidemia abrogates cardioprotective effects of preconditioning are uncertain and are not completely understood. The impaired opening of mitochondrial-KATP channels, eNOS uncoupling and excessive generation of superoxides in the hyperlipidemic myocardium could play a role in attenuating preconditioning-mediated myocardial protection against I/R injury. Moreover, hyperlipidemia-induced loss of cardioprotective effect of preconditioning is associated with redistribution of both sarcolemmal and mitochondrial Connexin 43. We addressed, in this review, the potential mechanisms involved in hyperlipidemia-induced impairment of myocardial preconditioning. Additionally, novel pharmacologic interventions to attenuate hyperlipidemia-associated exaggerated I/R-induced myocardial injury have been discussed.  相似文献

Carbon dioxide (CO2) is always maintained at ambient levels by ventilation in commercial egg incubators. However, elevated CO2 levels during the early and late periods have been reported to improve the quality of chicks and shorten the hatch window. This study investigated the effect of precise CO2 supplementation during the early and late periods of incubation on embryo growth and incubation performance by developing and using a CO2 supplementation system to increase the CO2 level in an experimental egg incubator. The CO2 level was maintained at 1% in the early period (from the beginning to the 10th day of incubation, E0E10) and in the late period (from internal pipping (IP) to the 21st day of incubation (E21), IP–E21) in an incubator for the treatment group, whereas the CO2 level was maintained at the ambient level in the other incubators for the control group. A comparative assessment of embryonic development, hatching characteristics, and hormone and nutrient levels was conducted for each trial. The experiment comprised three trials, with 300 Jing Hong No. 1 breeding eggs in each incubator. The elevated CO2 treatment significantly shortened the chick hatching time (H0) by 4 h (P < 0.05) and the hatch window by 3 h (P < 0.05) without affecting hatchability, chick weight at 1 d of age, brooding period, or quality score. At external pipping (EP), the heart weight, intestinal weight, relative intestinal weight, and relative heart weight in the treatment group were significantly higher than those in the control group (P < 0.05). In addition, the embryonic intestine, relative intestine, and relative heart weights of the newly hatched chicks in the treatment group were significantly higher than those in the control group (P < 0.05) at H0. The treatment significantly increased the concentration of corticosterone in the embryonic plasma during the period from IP to EP (P < 0.05), promoted the secretion of triiodothyronine and tetraiodothyronine (P < 0.05), and increased the glycogen content of the embryonic liver on E21 (P < 0.05). This result indicates that elevated CO2 (1%) during the early and late periods of incubation accelerated embryonic organ development and shortened the chick hatching time and hatch window without affecting hatchability or hatchling quality, which can be explained by the synergistic functions of the secretion of plasma corticosterone and thyroid hormones and the accumulation of liver glycogen between the early and late periods of incubation.  相似文献

For the present study we asked whether the endometrial fluid lipidomic may be a useful approach to predict endometrial receptivity in freeze‐all cycles. For this case‐control study, endometrial fluid samples were collected from 41 patients undergoing freeze‐all cycles. Samples were split depending on the pregnancy outcome: positive group (n = 24) and negative group (n = 17). Data were acquired by the matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry. Principal component analysis (PCA) and partial least squares discriminant analysis (PLS‐DA) were applied. A list of potential biomarker ion ratios was obtained and the values were used to build a receiver operating characteristic (ROC) curve to predict pregnancy success. The lipid categories were attributed by LIPID MAPS database. Ion ratios were established according to their correlations and used for the analysis. The PCA showed a tendency of separation between the studied groups, whereas the PLS‐DA was able to clearly distinguish them. Fifteen ratios (13 hyper‐represented in the negative and two hyper‐represented in the positive group) were selected according to their importance for model prediction. These ratios were used to build the ROC curve, which presented an area under curve of 84.0% (95%CI: 69.2–97.4%; p = 0.009). These findings suggest that lipidomic profiling of endometrial fluid may be a valuable tool for identifying the time interval comprising the window of implantation.  相似文献

The response of insects to four thinning intensities (control, light-thin, heavy-thin, or light-thin with gaps) was examined in 40-to 60-year-old Douglas-fir stands on the Willamette National Forest in Oregon, USA. In both 2000 and 2001, flying insect assemblages were collected from window traps placed over the forest floor in the center of each treatment block during two annual intervals: 1 ) June through August (early-season) and 2) August through October (late-season). The seasonal responses by these insects showed statistically significant differences in species richness (F= 22.21,P <0.001) and mean abundance (F=34.87,P <0.001). The greatest numbers of taxa generated via indicator species analysis were in the early season and within the treatment of light-thin with gap (L./ G). In particular, the woodborer beetle,Buprestis (Burprestidae), was strongly associated with L/G (Indicator Value = 52.4, P ≤0.001 ). Two beetle taxa,Ampedus (Elateridae, IV=84.7, P ≤0.001 ) and members of Cermabycidae (IV=58.4, P ≤0.001 ), were correlated significantly with early-season, whereasMelanoplus (Acrididae) was associated with late-season (IV=21.3, P >0.05) and L/G (IV=29.2, P ≤0.05). For all thinning treatments, the numbers of species and individuals were higher in the early season than the late season. Non-metric multidimensional scaling showed that Axis 1 (65%) and Axis 2 (27%) explained 92% of the variance. The former was strongly associated with thinning intensity, having higher coefficients of species richness (r=0.645) and species diversity (r=0.583). The multi-response permutation procedures showed a statistically significant difference for thinning intensity (T-statistics=-4.6322,A-statistics=0.0479,P <0.0001 ). These results suggest that heavier thinning can result in more diverse populations of flying insects, including herbivores and predators.  相似文献

In orthodontics, tooth movement is typically described using the rectangular coordinate system (XYZ); however, this system has several disadvantages when performing biomechanical analyses. An alternative method is the finite helical axis (FHA) system, which describes movement as a rotation about and a translation along a single axis located in space. The purpose of this study was to examine differences between the FHA and the XYZ systems in analyzing orthodontic tooth movement. Maxillary canine retraction was done using sliding mechanics or a retraction spring with midpalatal orthodontic implants used as measuring references. Tooth movement calculated with the FHA was compared with the corresponding movement in the rectangular coordinate system weekly over a 2-month interval in eight patients. The FHA showed that sliding mechanics controlled rotation of the canine better than the retraction spring (Ricketts retractor), and that the Ricketts retractor controlled tipping better. Changes in the FHA direction and position vectors with time showed that the biomechanical forces are not uniform during the treatment period. In both mechanics, the FHA provided a simple biomechanical model for canine retraction.  相似文献

The concept of optothermal window (OW) detection was used for the first time to assess the color of several products (juice, purée, paste) derived from thermally processed tomatoes. Unlike traditional techniques that operate either in the reflectance or transmission mode, the method proposed here actually relies on indirect measurement of absorbance in optically opaque and scattering samples. Very good correlation between the magnitude of the OW signal and the color-related parameters [colorimetric index L* and tomato paste index (TPI)] was observed.  相似文献

Motoneuron responses were elicited by global visual motion and stepwise displacements of an illuminated stripe. Stimulus protocols were identical to those used in previous behavioral studies of compensatory eyestalk reflexes. The firing rates and directional selectivity of the motoneuron responses were measured with respect to four stimulus dimensions (spatial frequency, contrast, angular displacement and velocity). The directional selectivity of the motoneuron response was correlated to the previously measured gain of the reflex for each stimulus dimension. The information theoretical analysis is based upon Kullback-Leibler (K-L) distances which measure the dissimilarity of responses to different stimuli. K-L distances for single neurons are strongly influenced by the mean rate difference of the responses to any pair of stimuli. Because of redundancy, the joint K-L distances of pairs of neurons were less than the sum of the K-L distances of the individual neurons. Furthermore, the joint K-L distances were only weakly influenced by correlations among coactivated neurons. For most of the stimulus dimensions, the K-L distances of single motoneurons were not sufficient to account for the stimulus discriminations exhibited by the eyestalk reflex which typically required the summed output of 2 to 5 motoneurons. Thus the behaviorally relevant information is encoded in the motoneuron ensemble. The minimum time required to discriminate the direction of motion (the encoding window) for a single motoneuron is about 380 to 480 ms (including a 175 ms response latency) for stepwise displacements and up to 1.0 s for global motion. During this period a motoneuron fires 2 to 3 impulses.  相似文献

Using available commercial robotics and instrumentation, we developed a fully automated and rigorous steady state enzyme kinetic assay for dipeptidyl peptidase IV (DPP IV; E.C. 3.4.14.5). The automated assay was validated with isoleucyl thiazolidide, a potent inhibitor of DPP IV with K(is)=110nM. Signal window analysis indicated that the assay had a 98% probability of detecting an inhibitor yielding 15% inhibition, with a predicted false positive rate of 0.13%. A mechanistic inhibition version of the automated assay was validated with isoleucyl 4-cyanothiazolidide, a very potent inhibitor of DPP IV. Isoleucyl 4-cyanothiazolidide was a competitive inhibitor of purified porcine DPP IV with K(is)=1 nM. Similar K(is) values were obtained for purified rat DPP IV and for DPP IV activity in human plasma from normal and diabetic donors. The pH dependence of K(is) for isoleucyl 4-cyanothiazolidide yielded a bell-shaped profile, with pK(a)=5.0 and pK(b)=7.6. To date, over 100,000 data points have been generated in profiling targeted compound libraries and in the analysis of tight-binding inhibitors of DPP IV. The data also show that robotic analysis is capable of producing full mechanistic inhibition analysis in a timely fashion to support drug discovery.  相似文献

摘要 目的:探讨肺表面活性物质(PS)治疗新生儿呼吸窘迫综合征(NRDS)前给予经鼻持续气道正压通气(nCPAP)呼吸支持的最佳时间窗。方法:选择2017年1月至2019年12月期间我院收治的NRDS患儿100例。根据随机数字表法分为A组(给予PS前预先进行小于2 h的nCPAP,n=33)、B组(给予PS前预先进行2-4 h的nCPAP,n=33)和C组(立即给予PS,n=34)。对比三组患儿的血气分析指标、肺功能指标、临床指标和并发症发生率。结果:A组、B组给予PS后4h、给予PS后24 h动脉血氧分压(PaO2)、pH值高于C组,且B组高于A组(P<0.05),而动脉二氧化碳分压(PaCO2)低于C组,且B组低于A组(P<0.05)。A组、B组给予PS后4 h、给予PS后24 h潮气量(VT)、肺动态顺应性(CD)高于C组,且B组高于A组(P<0.05),而吸气阻力(Raw)低于C组,且B组低于A组(P<0.05)。B组用药后3天内需气管插管行机械通气例数少于A组和C组,住院时间短于A组和C组(P<0.05),A组、C组的用药后3天内需气管插管行机械通气例数、住院时间对比无明显差异(P>0.05)。三组患儿并发症发生率未见统计学差异(P>0.05)。结论:给予PS前预先进行2-4h的nCPAP,可较好地改善患儿血气分析指标和肺功能,有助于改善患儿预后。  相似文献
