Skin pigmentation is a human phenotype that varies greatly among human populations and it has long been speculated that this variation is adaptive. We therefore expect the genes that contribute to these large differences in phenotype to show large allele frequency differences among populations and to possibly harbor signatures of positive selection. To identify the loci that likely contribute to among-population human skin pigmentation differences, we measured allele frequency differentiation among Europeans, Chinese and Africans for 24 human pigmentation genes from 2 publicly available, large scale SNP data sets. Several skin pigmentation genes show unusually large allele frequency differences among these populations. To determine whether these allele frequency differences might be due to selection, we employed a within-population test based on long-range haplotype structure and identified several outliers that have not been previously identified as putatively adaptive. Most notably, we identify the DCT gene as a candidate for recent positive selection in the Chinese. Moreover, our analyses suggest that it is likely that different genes are responsible for the lighter skin pigmentation found in different non-African populations. Ultraviolet (UV) radiation is a proven carcinogen, responsible for more than half of all human malignancies. It also compromises skin appearance and function. Since the UV action spectra for DNA damage, skin cancer and Vitamin D3 (vit D) photosynthesis are identical and vit D is readily available from oral supplements, why has sun protection become controversial, now that some data suggest conventionally “sufficient” levels of vit D may be less than optimal for at least some population groups? First, the media and apparently some researchers are hungry for a new message. Nevertheless, after 50 years, UV exposure is still a major avoidable health hazard. Second, the controversy is fueled by a powerful special interest group: the indoor tanning industry. They target not the frail elderly or inner-city ethnic minorities, groups for whom evidence of vit D insufficiency is strongest, but rather fair-skinned teenagers and young adults, those at highest risk of UV photodamage. Third, evolution does not keep pace with civilization. When nature gave man the appealing capacity for vit D photosynthesis, the expected lifespan was far less than 40 years. Long-term photodamage was not a concern, and vit D was not available at the corner store. The medical community should avoid sensationalism and instead rigorously explore possible cause-and-effect relationships between vit D status and specific diseases while advocating the safest possible means of assuring vit D sufficiency.  相似文献

应用石蜡切片和H.E染色技术,对石磺科(Onchidiidae)3个属的代表物种:瘤背石磺(Onchidium struma)、平疣桑椹石磺(Platevindex mortoni)和里氏拟石磺(Paraoncidium reevesii)的皮肤进行了组织学观察及参数测量比较。结果表明,3种石磺的皮肤虽然厚度不一,但基本结构相似,均由角质膜、表皮和真皮构成。角质膜是一层覆盖于表皮角质层上的蛋白质薄膜;表皮由多层上皮细胞构成,包括角质层、颗粒层和生发层;真皮包括疏松层和致密层,疏松层中嵌有颗粒腺和黏液腺两种腺体。3种石磺的皮肤厚度、各组织相对厚度及腺体数量等均存在差异。将结构差异与石磺的栖息环境进行比较分析后得到:陆栖为主的瘤背石磺皮肤表皮角质化程度高,颗粒腺发达;以水栖为主的里氏拟石磺表皮角质化程度相对低,黏液腺发达;而水陆两栖的平疣桑椹石磺,皮肤角质化程度介于前述二者之间,颗粒腺与黏液腺均不发达。研究结果体现了三者不同的生态适应特征,也为深入探讨海洋无脊椎动物从海洋向陆地进化的研究提供形态学依据。  相似文献

UVA irradiation-induced skin damage and redox imbalance have been shown to be ameliorated by ergothioneine (EGT), a naturally occurring sulfur-containing amino acid. However, the responsible molecular mechanism with nanomolar concentrations of EGT remains unclear. We investigated the dermato protective efficacies of EGT (125–500 nM) against UVA irradiation (15 J/cm2), and elucidated the underlying molecular mechanism in human keratinocyte-derived HaCaT cells. We found that EGT treatment prior to UVA exposure significantly increased the cell viability and prevented lactate dehydrogenase release into the medium. UVA-induced ROS and comet-like DNA formation were remarkably suppressed by EGT with a parallel inhibition of apoptosis, as evidenced by reduced DNA fragmentation (TUNEL), caspase-9/-3 activation, and Bcl-2/Bax dysregulation. Furthermore, EGT alleviated UVA-induced mitochondrial dysfunction. Dose-dependent increases of antioxidant genes, HO-1, NQO-1, and γ-GCLC and glutathione by EGT were associated with upregulated Nrf2 and downregulated Keap-1 expressions. This was confirmed by increased nuclear accumulation of Nrf2 and inhibition of Nrf2 degradation. Notably, augmented luciferase activity of ARE may explain Nrf2/ARE-mediated signaling pathways behind EGT dermato-protective properties. We further demonstrated that Nrf2 translocation was mediated by PI3K/AKT, PKC, or ROS signaling cascades. This phenomenon was confirmed with suppressed nuclear Nrf2 activation, and consequently diminished antioxidant genes in cells treated with respective pharmacological inhibitors (LY294002, GF109203X, and N-acetylcysteine). Besides, increased basal ROS by EGT appears to be crucial for triggering the Nrf2/ARE signaling pathways. Silencing of Nrf2 or OCTN1 (EGT carrier protein) signaling with siRNA showed no such protective effects of EGT against UVA-induced cell death, ROS, and apoptosis, which is evidence of the vitality of Nrf2 translocation and protective efficacy of EGT in keratinocytes. Our findings conclude that EGT at nanomolar concentrations effectively ameliorated UVA-induced skin damage, and may be considered as a desirable food supplement for skin protection and/or preparation of skin care products.  相似文献

PurposeThis study aims at characterizing the dosimetric behavior of an INTRABEAM® miniature accelerator equipped with flat and surface applicators, converting the spherical dose distribution into a flat one.MethodsDosimetric characterization was carried out in two steps. Firstly characterization was made in standard conditions for dermatologic applications, which is with the applicator directly on contact with the skin. Secondly, characterization was made in more clinical conditions, such as obliquities and heterogeneities.ResultsBehaviors of flat and surface applicators are different. Dose distribution for surface applicators is uniform at surface, whereas for flat applicator the maximum homogeneity is shown at a particular depth in water. Some results are different from previously published studies due to differences in the X-ray source design. The study showed that in the absence of a perfect contact between the applicator and the skin of the patient, there is a dose distribution spread on the edge of the irradiation field where the contact is not made. Dose loss due to lack of backscatter radiations is significant. By contrast, influence of a denser material behind the measurement point has no significant influence on the dose at this point. Thickness of tissue treated with flat and surface applicators is only a few millimeters, depending on the applicator's size, making these applicators ideal for superficial lesions, compared to high energy electrons and iridium brachytherapy.ConclusionsThe INTRABEAM® miniature accelerator equipped with surface applicators is a reliable way of treating superficial cutaneous malignancies.  相似文献

An increasing progress on the role of Hedgehog (Hh) signaling for carcinogenesis has been achieved since the link of Hh pathway to human cancer was firstly established. In particular, the critical role of Hh signaling in the development of Basal cell carcinoma (BCC) has been convincingly demonstrated by genetic mutation analyses, mouse models of BCCs, and successful clinical trials of BCCs using Hh signaling inhibitors. In addition, the Hh pathway activity is also reported to be involved in the pathogenesis of Squamous Cell Carcinoma (SCC), melanoma and Merkel Cell Carcinoma. These findings have significant new paradigm on Hh signaling transduction, its mechanisms in skin cancer and even therapeutic approaches for BCC. In this review, we will summarize the major advances in the understanding of Hh signaling transduction, the roles of Hh signaling in skin cancer development, and the current implications of “mechanism-based” therapeutic strategies.  相似文献

目的:研究Rab23在皮肤鳞状细胞癌(SCC)中的表达及意义。方法:用免疫组化S-P法分别检测30例皮肤SCC、15例正常皮肤组织标本中Rab23的表达。结果:Rab23在皮肤SCC和正常皮肤中阳性率分别为90%和13.3%,二者差异有统计学意义(P<0.05)。结论:Rab23在皮肤SCC中高表达可能在皮肤SCC的发生发展过程中发挥作用。  相似文献

Mice lacking the pro-adhesive matricellular protein connective tissue growth factor (CTGF/CCN2) display an embryonic lethal phenotype due to defects in bone and cartilage. However, the specific role of CCN2 in skin development is unknown. Here, we generated mice deleted for CCN2 in the entire body (using a cre/lox system in which CCN2 is deleted in the entire body due to the presence of a constitutively expressed cre recombinase). We found that CCN2 was not required for the development of skin as defined by skin thickness measurements, trichrome staining and immunostaining with anti-CD31 (to detect endothelial cells) and anti-α−SMA (to detect smooth muscle cells and pericytes) antibodies. Thus, although recently we have shown that CCN2 is required for fibrogenesis in postnatal mice, CCN2 is not required for skin development during embryogenesis.  相似文献

The protein p27BBP (alias eIF6) occurs in yeast and mammalian epithelial cells. It is essential for ribosome genesis and has also been implicated in the functionality of integrins and intermediate filaments. By immunoblot, we show that homogenized integument from Sepia officinalis (Cephalopoda, Mollusca) contains a protein with immunological properties that closely resemble those of p27BBP. We also demonstrate, by immunogold electron microscopy with an indirect immunoreaction technique on ultrathin sections of human skin and Sepia integument, that p27BBP is constantly present in both species in epithelial cells, fibroblasts, and muscle fibers. It is found in the vicinity of intermediate filaments, in nucleoli, along the internal wall of the nuclear membrane, and in association with desmosomes and hemidesmosomes and occasionally occurs extracellularly. Thus, the structure and function of p27BBP seem to have been highly conserved throughout evolution; the protein appears to be essential in eukaryotic cells in which it interacts with several ultrastructural components of diverse function.Financial support was provided by funds from FIRST.  相似文献
