Water-soluble O-acetylated galactoglucomannan (GGM) isolated from spruce thermomechanical pulp (TMP) by hot-water extraction was characterized by 1D and 2D (homo- and heteronuclear) NMR analysis. The backbone was found to consist of (1-->4)-linked mannopyranosyl and glucopyranosyl units in a ratio of 10:1.9-2.6. The mannopyranosyl units were acetylated at C-2 and C-3 with a degree of acetylation around 0.28-0.37 as determined by NMR. A slightly larger amount of 2-O-acetylated mannopyranosyl was detected when compared to the 3-O-acetylated component. Approximately every 10th mannopyranosyl unit was substituted at C-6 by a single alpha-galactopyranosyl unit. Fine structure determination based on sequence-specific chemical shift variations showed that the distribution of glycosyl residues is random. Small amounts of other minor polysaccharide species including xylans and galactans could also be identified by NMR.  相似文献

Saplings of four clones of Sitka spruce and cherry were grown for three and two growing seasons, respectively, in open top chambers at two CO2 concentrations (≈ 350 and ≈ 700 μmol mol–1) to determine whether the increase in total biomass brought about by enhanced [CO2] is a result of a transient or persistent effect in nonlimiting conditions. Classical growth analysis was applied to both species and mean current relative growth rate of total dry mass (RT) and leaf dry mass (RL), and period relative growth rate of total dry mass ( ) and leaf dry mass ( ) were calculated. Sitka spruce saplings and cherry seedlings showed a positive growth response to elevated [CO2], and at the end of the experiments both species were ≈ 40% larger in elevated [CO2] than in ambient [CO2]. As a result, the period mean and were significantly higher in elevated [CO2]. The differences in plant dry mass at the end of the experiments were a consequence of the more rapid growth in the early phase of exposure to elevated [CO2]. After this initial phase mean RT and RL were similar or even lower in elevated [CO2] than in ambient [CO2]. NAR of both species was much higher in elevated [CO2], whereas both LAR, SLA, and LMR showed the opposite trend. The higher LAR and SLA of plants in ambient [CO2] contributed to a compensation by which they maintained RT similar to that of elevated [CO2] saplings despite lower NAR and photosynthetic rate. However, when the same size the trees were similar amongst the [CO2] treatments, indicating that one of the main effect of elevated [CO2] on tree growth is to speed-up early development in all aspects.  相似文献

Abstract 1 During 1968–74, an outbreak of the European spruce sawfly Gilpinia hercyniae, defoliated many stands of Sitka spruce in commercial forests in mid‐Wales. The needle trace method was used to determine retrospectively the temporal pattern and intensity of defoliation in a stand of Sitka spruce in Hafren Forest that had been damaged severely at the time of the sawfly outbreak. 2 An initial calibration experiment, designed to test the reliability of the needle trace method when applied to Sitka spruce, indicated that artificially induced defoliation of up to 75% was detected by the technique with an accuracy of ±7%. Higher rates of defoliation were underestimated by up to 17%. 3 For the main sample of trees, the needle trace method demonstrated that retention of needle sets (average needle retention) was reduced by up to 33–38% over a 10‐year period coincident with the G. hercyniae outbreak and the years immediately afterward when the canopy was recovering. 4 Analysis of needle loss within separate needle cohorts and age‐classes revealed that 1‐year‐old needles were the most severely defoliated. The density of 1‐year‐old needles was reduced by 51–78% in 1970–73. 5 Defoliation at the time of the G. hercyniae outbreak was associated with reductions in annual height, radial, and volume increments of 24–49%, 30–59% and 32–56%, respectively. Radial and volume increments suffered their greatest reductions 1 year later than height increments. 6 The study demonstrates that the needle trace method can be applied successfully to Sitka spruce to quantify defoliation caused by an insect outbreak occurring many years previously, and that the technique can provide data on needle loss that is valuable for interpreting reductions in tree growth.  相似文献

Gibberellins Al (GA1), GA3, GA4, GA9, and after enzymatic hydrolysis of GA-conjugate-like fractions, GA9 and GA15, were identified in shoots of Sitka spruce [ Picea sitchensis (Bong.) Carr.] of different ages by combined gas chromatography-mass spectrometry (GC-MS). The purification and separation of the GAs involved the use of reverse phase and normal phase high performance liquid chromatography (HPLC). The Tan-ginbozu dwarf rice bioassay and binding to antibodies raised against GA1, GA4 and GA9 were used for detection of GA-like substances. The qualitative differences between the three ages of plant material were the presence of GA3 and GA1 in the 48-year-old material and the absence of detectable amounts of GA4 in the same material. This indicates a difference in GA metabolism which may reflect the difference in ability to form reproductive buds.  相似文献

Abstract. We investigated changes in the composition and abundance of understory species after fire in the southern boreal forest around Lake Duparquet, Québec. Ten plots of 100 m2 were sampled in each of eight sites varying in post-fire age from 26 to 230 yr, with 20 1-m2 quadrats in each of these 80 plots. Variation in the understory was described by DCA ordination and interpreted as a regeneration succession series. Thickness of the organic layers, stand age and canopy composition were all correlated with vegetational change. This change was not constant throughout succession; some old sites showed an increase in the diversity and abundance of certain pioneer species. This was partly related to openings in the canopy resulting from a major outbreak of spruce budworm, which affected sites dominated by Abies balsamea. The ordinations were performed on both the 100-m2 plots and the 1-m2 quadrats. Heterogeneity within sites was larger at the 1-m2 scale and there was a great deal of overlap in the position of the quadrats in ordination space. At the smaller scale of analysis, stand age and thickness of the organic layers were not correlated with the changes observed in the understory.  相似文献

Abstract. The concentrations and contents of organic matter and nutrients in organic deposits on the forest floor were estimated along a 231-yr chronosequence following fire at the southern limit of the boreal forest in eastern Canada. The sampling design was stratified to take into account the variability related to the presence of the principal tree species as well as to the presence of large gaps created by a recent spruce budworm (Choristoneura fumiferana) outbreak. The forest floor showed a steady accumulation of organic matter and total nutrients with time-since-fire and a 50 % decrease in the concentrations of available P and K, but not N (as determined by aerobic incubation). The increase in forest-floor weight was accompanied by an increased storage of available N, Ca and Mg. The availability of N and Ca was more strongly affected by tree species and gaps than by time-since-fire. A high N-availability was observed under Betula papyrifera and in gaps, while high a Ca-availability was found near Populus tremuloides and Thuja occidentalis. In old sites, the forest floor of gaps, created by a recent spruce budworm outbreak, had a necromass similar to that of a young forest, but the low concentrations of available P and K of an old forest.  相似文献

Picea rubens (red spruce) and P. mariana (black spruce) are closely related species which are difficult to differentiate morphologically. They are sympatric with P. glauca (white spruce) in the northern portion of their ranges. In order to identify potential interspecific polymorphisms, the chloroplast trnK intron and rpl33-psaJ-trnP region were sequenced, and the nuclear-encoded ITS region of the rDNA repeat was partially sequenced. Thirteen chloroplast and 12 nuclear candidate interspecific single nucleotide polymorphisms (SNPs) were identified. The species-specificity of several SNPs was determined by surveying DNAs amplified from trees representing range-wide provenance tests; these included 46 red spruce from 11 provenances, 84 black spruce from 30 provenances and 90 white spruce from 22 provenances. Two SNPs (1 chloroplast and 1 nuclear), which distinguish black spruce from red and white spruce, were consistent among 96–100% of the trees surveyed. Five SNPs (4 chloroplast and 1 nuclear), which distinguish white spruce from red and black spruce, were consistent among 100% of surveyed trees. These species-specific SNPs were used to identify anonymous spruce samples in a blind test, and their utility for small amounts of tissue, as little as single needles, was demonstrated. Scoring these SNPs is much less labor intensive than previous molecular methods for taxa differentiation (restriction fragment length polymorphisms or random amplified polymorphic DNAs), therefore they can be applied to large population studies. This study investigates the early effects of forest regeneration with selective, and small scale clear-cutting, on ground beetle (Coleoptera, Carabidae) community composition in a homogenous, mature spruce forest in Southern Bavaria (Höglwald), Germany. Carabid beetles were sampled with pitfall-, emergence-, and window-traps, from 1999 to 2001 during a pre-treatment year, the year of cutting, and the year after cutting. In the spruce stand we found a relatively low species richness with few dominating species. Selective cutting preserved this carabid assemblage. At the clear-cuts carabid species richness increased in the year of cutting, because of the invasion of small open field species, and the persistence of most forest species. Also, number of individuals increased due to higher numbers caught in the window-traps. The first open habitat species appeared just a few months after felling. However, in the next year the numbers of individuals, especially of forest species,were drastically reduced. Also, the number of species decreased, and was just slightly higher than on the control plot (mature stand). According to the DCA (detrended correspondence analysis) forest interior species had the same habitat preferences as net building spiders (Amaurobiidae, Linyphiidae) and other families of beetles (Staphylinidae, Curculionidae). Several groups of open habitat species responded positively on different patches found in the clear-cut: (1) diversity ofground vegetation, respectively coverage of shrubs, (2) favour for moist patterns, and water filled ruts (together with Gastropoda), or (3) low coverage of ground vegetation (together with free hunting spiders, Lycosidae). Different structures side by side (mature forest, selective cutting, open areas) may improve diversity on a forest scale. Small openings can serve as an important retreat for open habitat species. However, if clear-cuts become the dominant element, forest species maybe threatened. With selective cutting species richness of carabids is not improved; however, the remaining forest carabid species may be preserved during the early phase of the regeneration process.  相似文献

The present study was carried out to assess the possible cytogenetic changes in peripheral blood lymphocytes as a means of monitoring human populations subjected to environmental electromagnetic fields. The mean frequencies of chromosome aberrations (CA) and sister-chromatid exchanges (SCE) were determined in 72-h whole blood cultures from 15 workers (mean age 31.4 ± 5.6 years) occupationally exposed to 50 Hz electromagnetic field from a 132-230 kV electric supply substation.

Compared to a control group of eight men (mean age 31.6 ± 6.12 years), the percentage of aberrant cells was significantly increased (12.83 ± 1.28% for exposed and 7.00 ± 0.6% for nonexposed). No statistical difference was observed in the mean SCE values between the exposed (5.40 ± 0.15) and the nonexposed (5.12 ± 0.55) groups.

Furthermore, the cell proliferation index (CPI) and the mitotic index (MI) were analyzed. The two indices were significantly lower in the exposed group than in the nonexposed one: 1.44 versus 1.60 and 1.45 versus 1.79 for the two indices, respectively. The smoking habit did not influence any of the parameters investigated.  相似文献

ABSTRACT

After a 3-year exposure to elevated CO2, young trees of Sitka spruce (Picea sitchensis (Bong.) Carr.) were planted in native, nutrient-deficient forest soil and grown for two more years with three CO2 treatments in open-top chambers, and with two nutrient treatments (with and without supplied N). Elevated CO2 resulted in larger fresh mass, dry mass, leaf area and leaf thickness in two-year old needles, but had no effect on one-year old and current needles. Tree height, basal diameter and biomass production also increased, regardless of N supply. In trees without added N, elevated CO2 resulted in higher root-to-shoot and absorbing roots-to-stump ratios. Regardless of N supply, trees grown in elevated CO2 had lower photosynthetic rates on a leaf area basis. Photosynthesis reduction was accompanied by a decline in Rubisco activity and leaf N concentration. Under elevated CO2, added N elevated photosynthesis and Rubisco activity, suggesting a dependence on N availability of the photosynthetic response to elevated CO2. Stomatal conductance of trees grown with added N decreased in response to elevated CO2. This may account for the larger reduction in intercellular CO2 concentration, and hence photosynthesis, in the trees supplied with N than in those without N supply.  相似文献
