哈民忙哈遗址位于科尔沁沙地腹心地带,是迄今在西辽河以北地区发现的最重要的史前聚落遗址。本文全面考察2010~2014年发掘的44座房址的废弃情境,将它们分为三种类型。不同类型房址体现的不同废弃行为方式,分别对应灾难性、不预期返回和预期返回三种废弃模式。科尔沁沙地脆弱的生态环境促使人们采取流动性居住方式、依赖多元化的生计方式,从而形成了不预期返回和预期返回废弃模式。但这种人地关系的短暂平衡仍然潜伏着危机,当一系列因素耦合作用时,酿成了一场可怕的灾难,导致聚落形成最后阶段的灾难性废弃模式。  相似文献

Efficient light harvesting in a photosynthetic antenna system is disturbed by a ragged and fluctuating energy landscape of the antenna pigments in response to the conformation dynamics of the protein. This situation is especially pronounced in Photosystem I (PSI) containing red shifted chlorophylls (red Chls) with the excitation energy much lower than the primary donor. The present study was conducted to clarify light-harvesting dynamics of PSI isolated from Synechocystis sp. PCC6803 by using single-molecule spectroscopy at liquid?nitrogen temperatures. Fluorescence emission at around 720?nm from the red Chls in single PSI trimers was monitored at 80–100?K. Intermittent variations in the emission intensities, so-called blinking, were frequently observed. Its time scale lay in several tens of seconds. The blinking amplitude depended on the redox state of the phylloquinone (A1). Electrochromic shifts of Chls induced by the negative charge on A1 were calculated based on the X-ray crystallographic structure. A Chl molecule, Chl-A839 (numbering according to PDB 5OY0), bound near A1 was found to have a large electrochromic shift. This Chl has strong exciton coupling with neighboring Chl (A838) whose site energy was predicted to be determined by interaction with an arginine residue (ArgF84) [Adolphs et al., 2010]. A possible scenario of the blinking was proposed. Conformational fluctuations of ArgF84 seesaw the excitation-energy of Chl-A838, which perturbs the branching ratio of excitation-energy between the red Chl and the cationic form of P700 as a quencher. The electrochromic shift of Chl-A839 enhances the effect of the conformation dynamics of ArgF84.  相似文献

The Standard Genetic Code is organized such that similar codons encode similar amino acids. One explanation suggested that the Standard Code is the result of natural selection to reduce the fitness ``load' that derives from the mutation and mistranslation of protein-coding genes. We review the arguments against the mutational load-minimizing hypothesis and argue that they need to be reassessed. We review recent analyses of the organization of the Standard Code and conclude that under cautious interpretation they support the mutational load-minimizing hypothesis. We then present a deterministic asexual model with which we study the mode of selection for load minimization. In this model, individual fitness is determined by a protein phenotype resulting from the translation of a mutable set of protein-coding genes. We show that an equilibrium fitness may be associated with a population with the same genetic code and that genetic codes that assign similar codons to similar amino acids have a higher fitness. We also show that the number of mutant codons in each individual at equilibrium, which determines the strength of selection for load minimization, reflects a long-term evolutionary balance between mutations in messages and selection on proteins, rather than the number of mutations that occur in a single generation, as has been assumed by previous authors. We thereby establish that selection for mutational load minimization acts at the level of an individual in a single generation. We conclude with comments on the shortcomings and advantages of load minimization over other hypotheses for the origin of the Standard Code. Exposed riverine sediments (ERS) are an important invertebrate habitat. Ground beetle data from pitfall trap surveys of 165 sites in the catchments of the Rivers Carron, Nith, Spey, Tweed and Eden and from 48 sites in the catchments of the Rivers Blyth, Breamish, Coquet, Glen, Pont, South Tyne, Till, Tweed and Tyne were used to produce ERS habitat classifications in Scotland and northern England respectively. Within each habitat type the quality of sites were assessed using indices based on rarity and fidelity values and using species richness as a measure of diversity. Considerable variation in these indices was seen within and between habitat types, showing potential for their use in assessing site quality and for ranking sites within habitats. The use of standardised invertebrate survey and distribution data, together with species assemblage information, for the conservation of invertebrate habitat types as an alternative to the use of selected indicator species, is discussed.  相似文献

Post-disturbance survival is a key factor in the onset of secondary successions. Here we analyse capture-recapture data from two before/after disturbance studies to estimate the effect of fire on local bird survival. Analyses of six bird species at two Mediterranean shrubland sites were combined using a meta-analysis approach. Two warblers, Sylvia undata and S. melanocephala, were studied at one site altered by prescribed burning, and five passerines (Luscinia megarhynchos, Turdus merula, Parus major, P. caeruleus and S. melanocephala) at one site disturbed by wildfire. Based on the combined analysis, annual survival probability significantly decreased from 0.49 to 0.18 (i.e. a 64% decline) after the fire. Our results further suggest a trend for a higher decrease in annual survival associated with wildfire (–72%, from 0.51 to 0.14) than with prescribed burning (–35%, from 0.41 to 0.27), although this should be properly tested with a specific experimental design. In S. undata, a decline in survival in the long-term cannot account for the drop in density observed the first spring after fire. We suggest that a decrease in recruitment rate and an increase in the proportion of non-breeders immediately after the fire may contribute more strongly to the decline in the breeding population. Our results tend to support the idea that bird populations may respond to moderate disturbances with noticeable time lags, because of individual site tenacity.  相似文献

Single-photon counting fluorimetry was used to record the time course of the expression of interleukin-10 receptors labelled with fluorescent antibodies on the surface of adipocytes over 24h, following an immune challenge to the rat popliteal lymph node. Homologous perinodal and remote-from-node samples from the stimulated and unstimulated popliteal depots were compared in rats fed on plain chow and chow supplemented with 10% w/w suet, fish or vegetable oils. Receptor expression was maximal 6 h after stimulation, and returned to baseline after 24 h, and was similar in the stimulated and unstimulated depots. Fewer receptors were elicited in tissues from rats fed lipid-supplemented diets compared with the control diet, with fewest of all following the fish oil diet. These data suggest that interleukin-10 is involved in local interactions between perinodal adipocytes and lymph node lymphoid cells. Both triacylglycerols and phospholipids contained more polyunsaturates and fewer saturates in perinodal adipose tissue than in samples from sites not associated with lymphoid tissue. These data are consistent with paracrine interactions between perinodal adipocytes and activated lymphoid cells.  相似文献

The pollen competition hypothesis predicts that when the number of pollen grains deposited onto stigmas exceeds the number of ovules, selection can operate in the time frame between deposition and fertilization. Moreover, because of the overlap in gene expression between the two phases of the life cycle, selection on microgametophytes may alter the resulting sporophytic generation. The extent to which pollen competition occurs in nature has been unclear, because tests of the predictions of the pollen competition hypothesis have used cultivars and/or artificial growth conditions and hand-pollination techniques. In this study we used a wild species, Cucurbita foetidissima, in its natural habitat (southern New Mexico) to determine the amount and timing of the arrival of pollen onto stigmas, the relationship between pollen deposition and seed number, and the effects of the intensity of pollen competition on progeny vigor. We found that ~900 pollen grains are necessary for full seed set and that a single visit by a pollinator results in the deposition of 653.0 ± 101.8 pollen grains. About 29% of the flowers receiving a single pollinator visit had 900 or more pollen grains on its stigma. Moreover, within 2 h of anthesis, >4000 pollen grains were deposited onto a typical stigma, indicating that multiple pollinator visits must have occurred. Fruits produced by multiple visits had greater seed numbers (206 vs. 147) than fruits produced by a single visit. Finally, the progeny produced by multiple pollinator visits were more vigorous than those produced by single visits with respect to five measures of vegetative growth (MANCOVA, Wilks' lambda = 0.96, F(6,370) = 2.54, P < 0.02. These data demonstrate that conditions for pollen competition exist in nature and support the prediction that pollen competition enhances offspring vigor.  相似文献

利用多聚酶链式反应(PCR)技术,对天然天花粉蛋白(nTCS)基因在Tyr14和Arg22两个保守残基处同时进行定点突变,即Tyr14变成Phe,Arg22变成Leu,然后克隆到pET-8c高效表达载体上,构建成重组质粒pETY14F/R22L.经序列分析,定点突变的结果与预先设计的完全一致,突变后的天花粉蛋白命名为Y14F/R22LTCS.将pETY14F/R22L转化到E.coliBL21(DE3,pLysS)中,进行表达.经CM-SepharoseCL-6B柱纯化,SDS-PAGE鉴定,纯度可达90%.RIP活性测定显示,Y14F/R22LTCS的活性比nTCS降低了7.5倍,活性变化不显著,因此,TCS的Try14和Arg22对维持其活性部位构象并不是必需的.但由于Y14F/R22LTCS在E.coli中的表达量与nTCS相比明显下降,因此,Tyr14和Arg22可能与TCS翻译后的折叠有关.  相似文献

Assessment of the potential risks posed by chlorinated solvents in groundwater is the key to establish the extent of the contamination and derive achievable remedial targets should remediation deems necessary. This article first presents the application of the American Society for Testing and Materials (ASTM) Risk Based Corrective Actions (RBCA) Guidance to quantitatively evaluate human health and environmental risk for a former chemical works in Shanghai, China. The observed maximum trichloroethylene (TCE) concentration in groundwater at the site reached 1220 mg/l that exceeded its solubility of 1070 mg/l at 10°C (Soil annual average temperature is 10°C in Shanghai). The maximum concentration for cis-1, 2-DCE (DCE) was also found to be elevated at 264 mg/l. A critical exposure pathway was considered to be indoor vapor intrusion of TCE into the buildings with excess lifetime cancer risk for children being 1.7 × 10?3. This cancer risk exceeded regulatory limits of 1 × 10?4 to 1 × 10?6 for The Netherlands, the United Kingdom, and the United States. The calculated groundwater remedial targets for TCE and DCE are 7 mg/l and 904 mg/l, respectively, in order to protect child residents from inhalation of indoor vapors within the close proximity of the source area.  相似文献

Prion protein (PrP) is a glycosylphosphatidylinositol (GPI)-anchored protein, and the C-terminal GPI anchor signal sequence (GPI-SS) of PrP is cleaved before GPI anchoring. However, mutations near the GPI anchor attachment site (the ω site) in the GPI-SS have been recognized in human genetic prion diseases. Moreover, the ω site of PrP has not been identified except hamster, though it is known that amino acid restrictions are very severe at the ω and ω + 2 sites in other GPI-anchored proteins. To investigate the effect of mutations near the ω site of PrP on the conversion and the GPI anchoring, and to discover the ω site of murine PrP, we systematically created mutant murine PrP with all possible single amino acid substitutions at every amino acid residue from codon 228 to 240. We transfected them into scrapie-infected mouse neuroblastoma cells and examined the conversion efficiencies and the GPI anchoring of each mutant PrP. Mutations near the ω site altered the conversion efficiencies and the GPI anchoring efficiencies. Especially, amino acid restrictions for the conversion and the GPI anchoring were severe at codons 230 and 232 in murine PrP, though they were less severe than in other GPI-anchored proteins. Only the mutant PrPs presented on a cell surface via a GPI anchor were conversion competent. The present study shows that mutations in the GPI-SS can affect the GPI anchoring and the conversion efficiency of PrP. We clarified for the first time the ω site of murine PrP and the amino acid conditions near the ω site for the conversion as well as GPI anchoring.  相似文献
