Originally regarded as just membrane constituents and energy storing molecules, lipids are now recognised as potent signalling molecules that regulate a multitude of cellular responses via receptor-mediated pathways, including cell growth and death, and inflammation/infection. Derived from polyunsaturated fatty acids (PUFAs), such as arachidonic acid (AA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), each lipid displays unique properties, thus making their role in inflammation distinct from that of other lipids derived from the same PUFA. The diversity of their actions arises because such metabolites are synthesised via discrete enzymatic pathways and because they elicit their response via different receptors. This review will collate the bioactive lipid research to date and summarise the findings in terms of the major pathways involved in their biosynthesis and their role in inflammation and its resolution. It will include lipids derived from AA (prostanoids, leukotrienes, 5-oxo-6,8,11,14-eicosatetraenoic acid, lipoxins and epoxyeicosatrienoic acids), EPA (E-series resolvins), and DHA (D-series resolvins, protectins and maresins).  相似文献

A bifunctional high molecular weight (Mr, 64,500 Da) beta-1-3, 1-4 glucan 4-glucanohydrolase was purified to homogeneity from Thermomonospora sp., exhibiting activity towards lichenan and xylan. A kinetic method was used to analyze the active site that hydrolyzes lichenan and xylan. The experimental data was in agreement with the theoretical values calculated for a single active site. Probing the conformation and microenvironment at active site of the enzyme by fluorescent chemo-affinity label, OPTA resulted in the formation of an isoindole derivative with complete inactivation of the enzyme to hydrolyse both lichenan and xylan confirmed the results of kinetic method. OPTA forms an isoindole derivative by cross-linking the proximal thiol and amino groups. The modification of cysteine and lysine residues by DTNB and TNBS respectively abolished the ability of the enzyme to form an isoindole derivative with OPTA, indicating the participation of cysteine and lysine in the formation of isoindole complex.  相似文献

Most methods of analyzing intracellular metabolites require extraction of metabolites from the cells. A concern in these methods is underestimation of metabolite levels due to incomplete extraction. In comparing extraction methods, then, it would seem that the best method for extracting a particular metabolite is the one that gives the largest yield. 研究采用直接测序法,分析日本沼虾(Macrobrachium nipponense)rDNA基因内转录间隔区ITS1的DNA序列,以筛选日本沼虾SNPs位点。共分析了32个太湖水域野生日本沼虾样本,结果表明,日本沼虾ITS1序列平均长度为1749.8bp,是迄今已报道的最长的ITS1序列,A、G、T和C的平均含量分别为29.9%、28.3%、27.7%、14.0%,G+C的含量平均为42.3%。通过序列比对,共筛选出22个SNPs位点,SNPs位点出现频率为0.0126,其中9个为C/T转换(占40.91%),4个为A/G转换(占18.18%),2个为A/T颠换(占9.09%),5个为T/G颠换(占22.73%),1个为A/C颠换(占4.55%),1个A/T或C颠换(占4.55%)。日本沼虾ITS1序列的22个SNP位点中,21个位点为2个等位基因,1个位点出现了3个等位基因,为复等位基因位点。日本沼虾ITS1序列中还发现3个具有多态性的微卫星位点、1个高度变异区以及大量的缺失、插入。研究首次对日本沼虾ITS1序列进行了分析,并发现了大量的SNP位点,为日本沼虾遗传育种研究提供了新的分子标记。    相似文献

Bone adaption upon mechanical stimulation is accompanied by changes in gene expression. In this context we investigated the influence of mechanical loading on heparin binding growth associated molecule (HB-GAM) expression, an extracellular matrix molecule which in cell culture has been shown to stimulate the differentiation of osteoblasts. We obtained information on the participating signal transduction pathways using a mitogenic loading regimen. Specific inhibitors of various signal transduction pathways were added to loaded cells and to unloaded controls. By semi-quantitative PCR studies we demonstrated a rapid decrease of HB-GAM expression in primary osteoblasts and SaOs-2 cells by 20-30% upon mechanical loading within 30min. We showed that the RGD-integrin interaction is involved in the regulation of HB-GAM expression. Furthermore, integrity of the cytoskeleton, stretch-activated, and voltage-sensitive Ca(2+) channels as well as gap junctional communication are necessary for the downregulation of HB-GAM expression by mechanical loading.  相似文献

The circadian rhythm of locomotor activity of the field mouse Mus booduga was studied and single animal phase response curves (PRCs) (n = 8) were constructed for 15-min daylight pulses of 1000 lux intensity. The light pulses, presented at different phases of the circadian cycle, evoked advancing and delaying phase shifts (ΔPHs) depending on the circadian time (CT) of light pulse application. ΔPHs by light pulses applied at the same phase are strongly correlated with the animals' circadian period (τ). The results indicate a significant correlation between (i) τ and the area under the advance zone of the PRC (A) (r = +0.72, p > 0.05), (ii) τ and the area under the delay zone of the PRC (D) (r = -0.98, p > 0.00001), (iii) τ and the difference between the area under delay and advance zone of PRC (D-A) (r = -0.97, p > 0.00001), and (iv) between τ and ΔpHs (at various phases of the circadian cycle) and further suggest that the waveform and time course of PRC depend on the animals' endogenous period (τ). Cadherins form a large family of calcium-dependent cell-cell adhesion receptors involved in development, morphogenesis, synaptogenesis, differentiation, and carcinogenesis through signal mechanotransduction using an adaptor complex that connects them to the cytoskeleton. However, the molecular mechanisms underlying mechanotransduction through cadherins remain unknown, although their extracellular region (ectodomain) is thought to be critical in this process. By single molecule force spectroscopy, molecular dynamics simulations, and protein engineering, here we have directly examined the nanomechanics of the C-cadherin ectodomain and found it to be strongly dependent on the calcium concentration. In the presence of calcium, the ectodomain extends through a defined ("canalized") pathway that involves two mechanical resistance elements: a mechanical clamp from the cadherin domains and a novel mechanostable component from the interdomain calcium-binding regions ("calcium rivet") that is abolished by magnesium replacement and in a mutant intended to impede calcium coordination. By contrast, in the absence of calcium, the mechanical response of the ectodomain becomes largely "decanalized" and destabilized. The cadherin ectodomain may therefore behave as a calcium-switched "mechanical antenna" with very different mechanical responses depending on calcium concentration (which would affect its mechanical integrity and force transmission capability). The versatile mechanical design of the cadherin ectodomain and its dependence on extracellular calcium facilitate a variety of mechanical responses that, we hypothesize, could influence the various adhesive properties mediated by cadherins in tissue morphogenesis, synaptic plasticity, and disease. Our work represents the first step toward the mechanical characterization of the cadherin system, opening the door to understanding the mechanical bases of its mechanotransduction.  相似文献
