8-OxodG (8-oxo-2′-deoxyguanosine) is representative of nucleoside damage and shows a genotoxicity. To significantly reveal the contributions of 7-NH and C8-oxygen to the mutagenic effect of 8-oxodG by DNA polymerases, we evaluated the effects of the 8-halo-7-deaza-dG (8-halogenated 7-deaza-2′-deoxyguanosine) derivatives by DNA polymerases. 8-Halo-7-deaza-dGTPs were poorly incorporated by both KF(exo−) and human DNA polymerase β opposite dC or dA into the template DNA. Furthermore, it was found that KF(exo−) was very sensitive to the introduction of the C8-halogen, while polymerase β can accommodate the C8-halogen resulting in an efficient dCTP insertion opposite the 8-halo-7-deaza-dG in the template DNA. These results indicate that strong hydrogen bonding between 7-NH in the 8-oxo-G nucleobase and 1-N in the adenine at the active site of the DNA polymerase is required for the mutagenic effects. Whereas, I-deaza-dGTP shows an antiproliferative effect for the HeLa cells, suggesting that it could become a candidate as a new antitumor agent. 相似文献

A facile and robust microwave-assisted solution phase parallel synthesis protocol was exercised for the development of a 38-member library of N,N′-disubstituted thiourea analogues (1–38) by using an identical set of conditions. The reaction time for synthesis of N,N′-disubstituted thiourea analogues was drastically reduced from a reported duration of 8–12 h for conventional methods to only 1.5–2.0 min. All the derivatives (1–38) were characterized by physico-analytical techniques such as elemental analysis in combination with FT-IR, 1H, 13C NMR and by single crystal XRD analysis have also been performed. These compounds were screened for their in vitro urease inhibition activities. Majority of compounds exhibited potent urease inhibition activities, however, the most significant activity was found for 16, with an IC50 value of 1.23 ± 0.1 μM. Furthermore, the synthesized compounds were screened for their cytotoxic potential against lungs cancer cell lines. Cell culture studies demonstrated significant toxicity of the compounds on the cell lines, and the levels of toxicity were altered in the presence of various side groups. The molecular docking studies of the most potent inhibitors were performed to identify the probable binding modes in the active site of the urease enzymes. 通过分析TLR9基因的多态性,探究新疆不同绵羊品种抗病力的分子机制。以塔什库尔干羊、和田羊和多浪羊共107个样本为研究对象,采用PCR、测序法和生物信息学方法分析TLR9基因的SNP及氨基酸差异。发现7个SNP,其中3个位点为同义突变,4个位点为非同义突变,确定为12个等位基因,其中新发现6个等位基因。首次发现氨基酸G437R的突变。6个等位基因对应的TLR9蛋白3-D结构存在差异,可能影响机体对PAMP的识别。绵羊TLR9基因外显子2具有较高的多态性,导致LRR空间结构改变,进而影响机体对疾病的感受性。本研究为新疆南疆地区绵羊抗病育种提供基础数据。 相似文献

The study was conducted to explore the mechanisms of sex differences in the response to chronic hepatitis B(CHB) in terms of DNA methylation, SNP genotype, and gene expression. Genomic DNA was isolated from peripheral blood mononuclear cells(PBMCs) of CHB patients and healthy controls and evaluated using the Human Methylation 450 K Assay. The DNA methylation level at hg37 chromosome(CHR) X: 7810800 was further validated using pyrosequencing.SNP genotypes, VCX m RNA expression of PBMCs, and plasma VCX protein concentration were further examined using SNaPshot, RT-qPCR, and Western blot, respectively. Results showed that a total of 5529 CpG loci were differentially methylated between male and female CHB patients. DNA methylation level and CC+CT frequency at CHR X: 7810800,VCX mRNA expression of PBMCs, and plasma VCX protein concentration were higher in female than in male CHB patients. The CHR X: 7810800 locus was hypermethylated in CHB patients with CC+CT genotypes in comparison with those with the TT genotype. In cases of CC+CT genotypes, VCX mRNA expression was negatively correlated with the DNA methylation level. CHB patients with higher levels of HBV DNA, AST, and GGT or higher GPRI scores exhibited lower VCX expression. In conclusion, SNPs and DNA methylation at the CHR X: 7810800 locus cooperatively regulate VCX expression in CHB. The upregulated VCX expression in female CHB patients might represent a mechanism of protection from more severe liver dysfunction and extensive fibrosis, as observed in male CHB patients. 相似文献

The development of novel leishmanicidal agents that are capable of being replaced by the available therapeutic options has become a priority. In the present study, the synthesis and leishmanicidal activity of a series of 5-(nitroheteroaryl-2-yl)-1,3,4-thiadiazole derivatives are described. All compounds appeared to be potent anti-leishmanial agents against both promastigote and amastigote forms of Leishmania major (L. major). Amongst the synthesized compounds, 2-([1,4′-bipiperidin]-1′-yl)-5-(5-nitrofuran-2-yl)-1,3,4-thiadiazole (IIa) and 1-(5-(1-methyl-5-nitro-1H-imidazole-2-yl)-1,3,4-thiadiazol-2-yl)-4-(piperidine-1-yl) piperidine (IIc) are the most effective. Infection index was statistically declined in the presence of all compounds. The analysis of redox-related factors revealed that exposure of L. major cells to IIa and IIc led to an increase in reactive oxygen species (ROS). Furthermore, two compounds were able to increase ROS and NO levels in infected macrophages in a dose-independent manner. In addition, we showed that these compounds induced cell death in promastigotes. Altogether, our results indicated the anti-leishmanial potential of IIa and IIc is mediated by apoptosis through an imbalance in the redox system resulting in the elevation of ROS. This new class of compound seems to hold great promise for the development of new and useful anti-leishmanial agents. 相似文献

We synthesized novel phenylenediamine derivatives and evaluated them as labeling agents to label proteins in close proximity to a single electron transfer catalyst. We found that N’-acyl-N-methylphenylenediamine labels tyrosine effectively in a model experiment using tris(bipyridine)ruthenium (Ru(bpy)32+) as the single electron transfer catalyst. By changing the substituents on the nitrogen atom of the phenylenediamine derivatives, the electrochemical properties of the labeling agent can be drastically changed. On the other hand, horseradish peroxidase (HRP) also catalyzes the reaction with almost the same oxidation potential as Ru(bpy)32+ (~+1.1?V). HRP proximity labeling is applicable to signal amplification in immunohistochemistry. We evaluated the phenylenediamine derivatives as labeling agents for HRP proximity labeling and signal amplification, and found that N’-acyl-N-methylphenylenediamine is a novel and efficient agent for signal amplification using HRP in immunohistochemistry. 相似文献

The interaction between integrin macrophage differentiation antigen associated with complement three receptor function (Mac-1) and intercellular adhesion molecule-1 (ICAM-1), which is controlled tightly by the ligand-binding activity of Mac-1, is central to the regulation of neutrophil adhesion in host defense. Several "inside-out" signals and extracellular metal ions or antibodies have been found to activate Mac-1, resulting in an increased adhesiveness of Mac-1 to its ligands. However, the molecular basis for Mac-1 activation is not well understood yet. In this work, we have carried out a single-molecule study of Mac-1/ICAM-1 interaction force in living cells by atomic force microscopy (AFM). Our results showed that the binding probability and adhesion force of Mac-1 with ICAM-1 increased upon Mac-1 activation. Moreover, by comparing the dynamic force spectra of different Mac-1 mutants, we expected that Mac-1 activation is governed by the downward movement of its alpha7 helix. 相似文献

目的:探讨用单光子发射型计算机断层(single photon emission computedtomography,SPECT)心肌灌注显像,评估心肌梗死(AMI)经冠脉介入治疗(PCI)后的心肌灌注疗效。方法:采用99mTc-tetrofosmin(P53)SPECT心肌灌注显像对54例行PCI治疗的AMI患者评估心肌灌注情况,并追踪记录6个月内心脏事件发生率。结果:SPECT显示无复流组22例,有复流组32例,两组心肌梗死患者近期预后差异有统计学意义(P<0.05)。无复流组不良事件发生率较有复流组有增加趋势;另外,急诊PCI组的预后明显好于择期PCI组,差异有统计学意义(P<0.05)。结论:SPECT心肌灌注显像可对AMI患者梗死相关血管(IRA)再通治疗疗效进行可靠的无创性评价。 相似文献

The potential of ballast water to act as a major introduction vector for toxic dinoflagellates and other phytoplankton is beyond doubt; however, evidence that links the suspected introduced species with a source population is less convincing, especially without supporting historical and biochemical data, or consideration of palaeobiogeographical scenarios that may explain current species distributions. This paper presents new molecular data based on LSU-rDNA and rDNA-ITS sequences that demonstrate an unequivocal and recent link between Temperate Asian and Australasian populations of the toxic dinoflagellates Gymnodinium catenatum and toxic strains of the Alexandrium “tamarensis complex”. We integrate our data with supporting evidence from historical distribution records, sediment dating studies, toxin profiles, mating studies and previous molecular studies. We contrast the observed patterns of genetic and biochemical variation with those expected from various palaeobiogeographical scenarios explaining the evolution and natural dispersal of both species. While definitive proof is impossible, the total evidence indicates that these toxic dinoflagellates were introduced to Australasia during the past 100 years, most probably via ballast water from bulk-cargo shipping from Japan and/or south-east Asia. 相似文献
