Nymphs feeding on ears of four rabbits and four calves (Bos taurus) were examined during first and third or fourth infestations and also during the moulting period. The gut caecae were removed and examined by histochemistry and light and electron microscopy. Attachment sites of the nymphs were biopsied from all hosts and cell counts made by light microscopy. Resistance was expressed by reduction in numbers of ticks completing engorgement and reduced engorgement weights. The gut was comprised of a proliferative stem cell; a digestive cell that differentiated into a sessile type ingesting by pinocytosis and a motile type ingesting by phagocytosis; and a cell secreting a glycoprotein with acid phosphatase activity into the lumen. The gut grew during the early stages of feeding to accommodate the expansion during engorgement. On rabbits and cattle resistant to ticks the stem cells were damaged, with moribund nuclei and poorly differentiated cytoplasm. Thus there were fewer digestive and secretory cells and the gut did not expand to accommodate a full blood meal. The attachment sites were dominated by mononuclear cells and neutrophils in both host species at the first infestations but at the third or fourth infestations there was a considerable increase in proportions of eosinophils and basophils. Summary A soluble lipid-protein complex in bovine retinal pigment epithelium is shown to carry endogenous 11-cis retinaldehyde, in the extent of 15% of the total 11-cis retinaldehyde found in this tissue. The complex, analyzed with respect to its chemical composition, exhibits a lipid composition close resembling the lipid composition of the rod outer segment membrane; the SDS-PAGE evidences the presence of a number of protein bands, two of which of 34 and 27 kDa appear glycoproteins. Finally, the lipid-protein complex exhibits a discrete level of a Cathepsin D-like protease activity. From the above, the possibility is discussed that the soluble lipid-protein complex could represent some phagolysosomal inclusion occurring in the pigment epithelial cells upon rod outer segment phagocytosis. 相似文献

This paper describes the results of an ultrastructural study on the subcellular events occurring in nematode-infecting (trophic) hyphae of the nematophagous fungus Arthrobotrys oligospora. In early stages of the infection process (30 min-4 h), the infection bulb and developing trophic hyphae are characterized by a highly proliferated endoplasmic reticulum (ER). Its membranes often appeared vesiculated and occur in close association with the cell membrane of the cells. Upon further invasion of the nematode, lipid droplets developed in the trophic hyphae; these droplets were first observed 4–5 h after the infection but were abundantly present after 24–36 h. Along with the formation of lipid droplets proliferation of microbodies was observed. These organeles were characterized by the presence of catalase and thiolase and were frequently observed in close association with the lipid droplets. Later on the lipid droplets disappeared. During this period new vegetative mycelium developed from the trap that had originally captured the nematode. Our results suggest that part of the nutrients released from the nematode are first converted into lipids by the fungus which in turn are degraded via the -oxidation pathway and further metabolized to support growth of new vegetative hyphae. 相似文献

The biological impact of consumption and utilization of celery was examined for the generalist herbivore, Spodoptera exigua (Hübner). An analysis of leaves and petioles demonstrated that both plant parts contained high levels of water (>85%), but that leaves contained twice as much total organic nitrogen as petioles. Developmental and reproductive rates were significantly higher (P<0.01, t-test) for larvae fed on leaves. Penultimate and final instars consumed significantly more (P0.05, t-test) dry weight of plant material when fed petioles as opposed to leaves. Although growth rates were similar regardless of food source for penultimate instars, conversion of food to body weight was lower for larvae limited to petioles. In the final instar, both growth and conversion rates were lower for larvae fed petioles than for those given leaves. Thus, since larvae exhibit preferences for the nutritionally less suitable petioles during later larval stages, environmental factors other than nutrition must influence movement and distribution of S., exigua in celery.
Résumé L'impact biologique de la consommation et de l'utilisation du céleri a été examiné sur un herbivore généraliste Spodoptera exigua Hübner. Une analyse de feuilles et de pétioles montre que ces deux parties de la plante ont une haute teneur en eau (>85%), mais que les feuilles contiennent deux fois plus d'azote organique total que les pétioles. Les taux de développement et de reproduction on été significativement plus élevés (<0,01 test t) pour les chenilles nourries sur feuilles. Les 2 derniers stades ont consommé significativement plus (P<0,05 test t) de poids sec de matière végétale en s'alimentant sur pétioles qu'en s'alimentant sur feuilles. Bien que les taux de croissance aient été voisins quel que soit l'aliment pour les avant-derniers stades, la conversion d'aliment en poids de corps était inférieure pour les chenilles ne consommant que des pétioles. Au dernier stade, tant les taux de croissance que de conversion des chenilles élevées sur pétioles étaient inférieurs à ceux des chenilles élevées sur feuilles. Ainsi, puisque les chenilles présentent une préférence pour les pétioles moins nutritifs au dernier stade larvaire, des paramètres écologiques autres que l'alimentation doivent influencer les mouvements et la distribution de S. exigua sur céleri.相似文献

ABSTRACT. The digestion of dandelion ( Taraxacum officinale ) pollen by adult worker honeybees ( Apis mellifera Linn.; Hymenoptera: Apidae) was initiated at the germination pores. 30 min after feeding, pollen had reached the anterior midgut and the germination pores had become swollen. This permitted further removal of protoplasm during the next 2 h of digestion as the pollen passed into the middle portion of the midgut. 3 h after feeding, pollen grains had reached the posterior midgut where some had ruptured to release both 'naked' protoplasm and masses of protoplasm but many remained intact or were only partially digested; undigested pollen grains passed unchanged to the rectum. The lipid-rich pollenkitt layer was removed from the exine during digestion. Our studies indicate that dandelion pollen was not utilized completely by honeybees. 相似文献

Vertebrate dung is central to the dung beetle life cycle, constituting food for adults and a protective and nutritive refuge for their offspring. Adult dung beetles have soft mandibles and feed primarily on nutritionally rich dung particles, while larvae have sclerotized mandibles and consume coarser dung particles with a higher C/N ratio. Here, using the dung beetles Euoniticellus intermedius and E. triangulatus, we show that these morphological adaptations in mandibular structure are also correlated with differences in basic gut structure and gut bacterial communities between dung beetle life stages. Metagenome functional predictions based on 16S rDNA characterization further indicated that larval gut communities are enriched in genes involved in cellulose degradation and nitrogen fixation compared to adult guts. Larval gut communities are more similar to female gut communities than they are to those of males, and bacteria present in maternally provisioned brood balls and maternal ‘gifts’ (secretions deposited in the brood ball along with the egg) are also more similar to larval gut communities than to those of males. Maternal secretions and maternally provisioned brood balls, as well as dung, were important factors shaping the larval gut community. Differences between gut microbiota in the adults and larvae are likely to contribute to differences in nutrient assimilation from ingested dung at different life history stages. 相似文献

The preparation of hair for the determination of elements is a critical component of the analysis procedure. Open-beaker,
closedvessel microwave, and flowthrough microwave digestion are methods that have been used for sample preparation and are
discussed. A new digestion method for use with inductively coupled plasma-mass spectrometry (ICP-MS) has been developed. The
method uses 0.2 g of hair and 3 mL of concentrated nitric acid in an atmospheric pressurelow-temperature microwave digestion
(APLTMD) system. This preparation method is useful in handling a large numbers of samples per day and may be adapted to hair
sample weights ranging from 0.08 to 0.3 g. After digestion, samples are analyzed by ICP-MS to determine the concentration
of Li, Be, B, Na, Mg, Al, P, S, K, Ca, Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ge, As, Se, Rb, Sr, Zr, Mo, Pd, Ag, Cd, Sn, Sb,
I, Cs, Ba, Pt, Au, Hg, Tl, Pb, Bi, Th, and U. Benefits of the APLTMD include reduced contamination and sample handling, and

The bioavailability of trace elements in infant formulas is affected by different physiological and dietetic factors. In vitro
methods based on element dialyzability have been proposed to estimate the bioavailability. Infant formulas of the same type
but from different manufacturers can differ in the salt used for supplementation and in the contents of other components that
can affect mineral bioavailability. The aim of our study is to estimate the dialyzability of iron, zinc, and copper of formulas
marketed in Spain, in order to detect possible differences in formulas of the same type coming from different manufacturers.
At the same time, the effects of the type of formula, the composition of the protein fraction, and the mineral content on
the element dialyzability are also studied. Differences are found in the dialysis percentages of the elements studied in formulas
of the same type but from different manufacturers. The formulas giving the highest dialysis percentages for the three considered
elements are the hypoallergenic ones based on protein hydrolysates. No differences are observed in formulas having whey or
casein as the main protein fraction. Significant correlations are obtained between the element contents and the dialyzability
