Summary Non-destructive observations of root growth and distribution can be obtained from counting root intercepts with observation tubers inserted in the root zone. This paper describes the technique of inserting clear acrylic tubes horizontally into large undisturbed and repacked soil cores. Counts of roots intersecting scribed lines on the sides of the tubes were made with a fibrescope. Comparison was made between observation root tubes of different diameter (25 and 38.5 mm).The r2 values for the relationships between root intercept counts and destructively determined values of root length density (RLD) ranged from 0.78 to 0.96. The larger diameter tubes had higher r2 values. Theoretical calibration of the technique does not appear to be possible since analysis indicated that fewer roots intersected the scribed lines on the observation tube than would have been expected from a non-disturbed, randomly distributed root system. It is not known if this discrepancy is due to non-randomness or to an artifact associated with the insertion of the observation tube. Roots were not more prolific at the edge of the soil cores. Comparison of values of root length per unit soil surface area, rates of downward root growth and water uptake rates were within the ranges previously reported for wheat roots of field crops grown on clay soils. Observed root growth and distribution was found to be sensitive to four soil and water treatments imposed. It is concluded that the technique will allow quantitative analysis of root growth and distribution in undisturbed soil cores.  相似文献

Abstract The spontaneous aggregation of Candida albicans germ tubes, which occurs in medium 199, has been studied. The disaggregating activity of various substances, such as sugars, enzymes and sulphydryl compounds has been tested by addition of these substances to the culture medium. Glutathione and dithiothreitol were the most effective in inhibiting aggregation without disturbing germ tube formation.
In a buffer solution, pH 7, the previously aggregated tubes were irreversibly dispersed by pronase. In the presence of glutathione and dithiothreitol, the process was reversible, since after washing with distilled water, germ tubes aggregated immediately.  相似文献

Abstract. We have cine-filmed the random motion of microscopic particles, mostly starch grains from ruptured plastids, in sieve tubes of Heracleum mantegazzianum L. and H. sphohdylium Somm. and Lev. Our frame-by-frame analysis of the positions of the particles shows that they move much less than calculated when generally accepted estimates for the viscosity of sieve-lube sap are inserted in the Stokes-Einstein and other equations for Brownian motion. Our analysis of a film, of similar particles, made by previous workers leads us to disagree with their conclusion that particle movement in sieve tubes was greater than should be expected for ordinary Brownian motion. Particles in their film and in ours moved less than expected even when we allow for the possibility that the particles are restricted by cell walls and by each other. We suggest that the particles move less than expected because the viscosity of sieve-lube sap may be higher than has been assumed by physiologists.  相似文献

The superior sensitivity of current mass spectrometers makes them prone to contamination issues, which can have deleterious effects on sample analysis. Here, bis(2,2,6,6-tetramethyl-4-piperidyl) sebacate (marketed under the name Tinuvin 770) is identified as a major contaminant in applications using liquid chromatography coupled with mass spectrometry (LC-MS). Tinuvin 770 is often added to laboratory and medical plastics as a UV stabilizer. One particular lot of microcentrifuge tubes was found to have an excess of this compound that would leach into samples and drastically interfere with LC-MS data acquisition. Further analysis found that Tinuvin 770 readily leached into polar and nonpolar solvents from the contaminated tube lot. Efforts to remove Tinuvin 770 from contaminated samples were unsuccessful. A prescreening method using MALDI-TOF MS is presented to prevent system contamination and sample loss.  相似文献

A monoclonal antibody, 12C9, an anti-idiotypic mimic of dothistromin, a toxin produced by Dothistroma pini, was found to label the cell wall of sieve elements in a number of different plant tissues and species. The antibody labeled apple leaf tissue, tobacco leaf mid vein, leaf and meristem, and Coprosma robusta leaf mid vein. Labeling was restricted to cell walls of sieve elements and did not label the companion cells or the lumen of the cells. The antibody labeled over a wide range of dilutions. This antibody could be used to differentiate sieve elements from other types of phloem. It could also be used to co-localize sieve elements and microorganisms such as phytoplasmas stained with DAPI.  相似文献

The paper presents selected figures of chick embryogenesis as depicted in the classic studies of Caspar Friedrich Wolff (1734–1794), Christian Heinrich Pander (1794–1865) and Karl Ernst von Baer (1792–1786). My main objective here is (1) to demonstrate how the imagery of Wolff, Pander and Baer attempted to project an image of a 3-dimensional rotating body into static figures on paper by means of linear contours, and (2) to ponder on the efficacy and pervasiveness of dots, lines and arrows for depicting embryogenesis.  相似文献

A major question in pollen tube growth in planta remains: do the pollen tube walls form a barrier to interaction with the environment? Using cryo‐FESEM, we directly assessed the 3D construction and porosity of tobacco pollen tube walls. Fractured mature primary walls showed a 40–50 nm spaced lattice of continuous fibers interconnected by short rods in the primary wall. These observations agree with TEM observations of sectioned walls. In the secondary callose wall, for which no structure is visible using TEM, cryo‐FESEM also revealed a 50 nm lattice consisting of longer fibers, approximately 10–15 nm wide, with rod‐like, thinner interconnections at angles of approximately 90° with the longer fibers. Such architecture may reflect functional needs with respect to porosity and mechanical strength. The wall does not form a mechanical barrier to interaction with the environment and is gained at low cost. Cryo‐FESEM additionally revealed another special feature of the wall: the tubes were tiled with scales or rings that were highly conspicuous after pectin extraction with EDTA. These rings cause the typical banding patterns of pectin that are commonly seen in pollen tubes during oscillatory growth, as confirmed by staining with toluidine blue as well as by DIC microscopy. Growth analysis by VEC‐LM showed that the ring‐ or scale‐like structures of the primary wall consist of material deposited prior to the growth pulses. The alternating band pattern seen in the callose wall is probably imposed by constrictions resulting from the rings of the primary wall.  相似文献

We characterize a novel, pollen-specific, microtubule-associated protein, SB401, found in Solanum berthaultii. This protein binds to and bundles taxol-stabilized microtubules and enhances tubulin polymerization in a concentration-dependent manner, particularly at lower temperatures. Electron microscopy revealed that the protein decorates the entire length of microtubules. Cross-linking and electrophoresis studies showed that SB401 protein forms dimers, and suggest that dimerization could account for bundling. Double immunofluorescent staining of pollen tubes of S. berthaultii showed that SB401 protein co-localized with cortical microtubule bundles. SB401 protein also binds to and bundles actin filaments, and could connect actin filaments to microtubules. SB401 protein had a much higher affinity for microtubules than for actin filaments. In the presence of both cytoskeletal elements, the protein preferentially bound microtubules to form bundles. These results demonstrate that SB401 protein may have important roles in organizing the cytoskeleton in pollen tubes.  相似文献

An immature vasa vasorum in the adventitia of arteries has been implicated in induction of the formation of unstable atherosclerotic plaques. Normalization/maturation of the vasa vasorum may be an attractive therapeutic approach for arteriosclerotic diseases. Nerve growth factor (NGF) is a pleotropic molecule with angiogenic activity in addition to neural growth effects. However, whether NGF affects the formation of microvessels in addition to innervation during pathological angiogenesis is unclear. In the present study, we show a new role for NGF in neovessels around injured arterial walls using a novel in vivo angiogenesis assay.  相似文献
