Plant defensins are cysteine-rich cationic peptides, components of the innate immune system. The antifungal sensitivity of certain exemplars was correlated to the level of complex glycosphingolipids in the membrane of fungi strains. Psd1 is a 46 amino acid residue defensin isolated from pea seeds which exhibit antifungal activity. Its structure is characterized by the so-called cysteine-stabilized α/β motif linked by three loops as determined by two-dimensional NMR. In the present work we explored the measurement of heteronuclear Nuclear Overhauser Effects, R1 and R2 15N relaxation ratios, and chemical shift to probe the backbone dynamics of Psd1 and its interaction with membrane mimetic systems with phosphatidylcholine (PC) or dodecylphosphocholine (DPC) with glucosylceramide (CMH) isolated from Fusarium solani. The calculated R2 values predicted a slow motion around the highly conserved among Gly12 residue and also in the region of the Turn3 His36-Trp38. The results showed that Psd1 interacts with vesicles of PC or PC:CMH in slightly different forms. The interaction was monitored by chemical shift perturbation and relaxation properties. Using this approach we could map the loops as the binding site of Psd1 with the membrane. The major binding epitope showed conformation exchange properties in the μs-ms timescale supporting the conformation selection as the binding mechanism. Moreover, the peptide corresponding to part of Loop1 (pepLoop1: Gly12 to Ser19) is also able to interact with DPC micelles acquiring a stable structure and in the presence of DPC:CMH the peptide changes to an extended conformation, exhibiting NOE mainly with the carbohydrate and ceramide parts of CMH.  相似文献

Salp15 is a tick saliva protein that inhibits CD4+ T cell differentiation through its interaction with CD4. The protein inhibits early signaling events during T cell activation and IL-2 production. Because murine Experimental Autoimmune Encephalomyelitis development is mediated by central nervous system-infiltrating CD4+ T cells that are specific for myelin-associated proteins, we sought to determine whether the treatment of mice with Salp15 during EAE induction would prevent the generation of proinflammatory T cell responses and the development of the disease. Surprisingly, Salp15-treated mice developed more severe EAE than control animals. The treatment of EAE-induced mice with the tick saliva protein did not result in increased infiltration of T cells to the central nervous system, indicating that Salp15 had not affected the permeability of the blood-brain barrier. Salp15 treatment did not affect the development of antibody responses against the eliciting peptide or the presence of IFNγ in the sera. The treatment with Salp15 resulted, however, in the increased differentiation of Th17 cells in vivo, as evidenced by higher IL-17 production from PLP139-151-specific CD4+ T cells isolated from the central nervous system and the periphery. In vitro, Salp15 was able to induce the differentiation of Th17 cells in the presence of IL-6 and the absence of TGFβ These results suggest that a conductive milieu for the differentiation of Th17 cells can be achieved by restriction of the production of IL-2 during T cell differentiation, a role that may be performed by TGFβ and other immunosuppressive agents.  相似文献

The mammalian degradation of lysine is believed to proceed via two distinct routes, the saccharopine and the pipecolic acid routes, that ultimately converge at the level of α-aminoadipic semialdehyde (α-AASA). α-AASA dehydrogenase-deficient fibroblasts were grown in cell culture medium supplemented with either l-[α-15N]lysine or l-[ε-15N]lysine to explore the exact route of lysine degradation. l-[α-15N]lysine was catabolised into [15N]saccharopine, [15N]α-AASA, [15N]Δ1-piperideine-6-carboxylate, and surprisingly in [15N]pipecolic acid, whereas l-[ε-15N]lysine resulted only in the formation of [15N]saccharopine. These results imply that lysine is exclusively degraded in fibroblasts via the saccharopine branch, and pipecolic acid originates from an alternative precursor. We hypothesize that pipecolic acid derives from Δ1-piperideine-6-carboxylate by the action of Δ1-pyrroline-5-carboxylic acid reductase, an enzyme involved in proline metabolism.  相似文献

We consider models of nucleotidic substitution processes where the rate of substitution at a given site depends on the state of the neighbours of the site. We first estimate the time elapsed between an ancestral sequence at stationarity and a present sequence. Second, assuming that two sequences are issued from a common ancestral sequence at stationarity, we estimate the time since divergence. In the simplest non-trivial case of a Jukes-Cantor model with CpG influence, we provide and justify mathematically consistent estimators in these two settings. We also provide asymptotic confidence intervals, valid for nucleotidic sequences of finite length, and we compute explicit formulas for the estimators and for their confidence intervals. In the general case of an RN model with YpR influence, we extend these results under a proviso, namely that the equation defining the estimator has a unique solution.  相似文献

A mathematical model of the G protein signaling pathway in RAW 264.7 macrophages downstream of P2Y6 receptors activated by the ubiquitous signaling nucleotide uridine 5’-diphosphate is developed. The model, which is based on time-course measurements of inositol trisphosphate, cytosolic calcium, and diacylglycerol, focuses particularly on differential dynamics of multiple chemical species of diacylglycerol. When using the canonical pathway representation, the model predicted that key interactions were missing from the current network structure. Indeed, the model suggested that accurate depiction of experimental observations required an additional branch to the signaling pathway. An intracellular pool of diacylglycerol is immediately phosphorylated upon stimulation of an extracellular receptor for uridine 5’-diphosphate and subsequently used to aid replenishment of phosphatidylinositol. As a result of sensitivity analysis of the model parameters, key predictions can be made regarding which of these parameters are the most sensitive to perturbations and are therefore most responsible for output uncertainty.  相似文献

利用PCR、克隆测序、序列拼接获得山羊(Capra hircus)BMP15基因全长。利用F-CSGE技术分析两个外显子,发现山羊BMP15编码序列的第901处发生了A→G单碱基突变,该突变使得第301位氨基酸(成熟蛋白质第32位氨基酸)由丝氨酸变为甘氨酸。利用LDR技术对济宁青山羊、鲁北白山羊和沂蒙黑山羊进行突变检测,并进行其与产羔数的关联分析。结果表明,该突变对济宁青山羊产羔数没有显著影响,但对鲁北白山羊及沂蒙黑山羊产羔数均有显著影响(P0.05)。GG型和AG型的鲁北白山羊产羔数分别比AA型多0.34只(P0.01)和0.31只(P0.01)。AG型沂蒙黑山羊的产羔数比AA型多0.13只(P0.01)。初步表明,BMP15是控制鲁北白山羊和沂蒙黑山羊多胎性状的一个主效基因或是与之存在紧密遗传连锁的分子标记。  相似文献

The formation of a primary endocytic vesicle is a dynamic process involving the transient organization of adaptor and scaffold proteins at the plasma membrane. Epsins and Eps15-like proteins are ubiquitin-binding proteins that act early in this process. The yeast epsins, Ent1 and Ent2, carry functional ubiquitin-interacting motifs (UIMs), whereas the yeast Eps15-like protein, Ede1, has a C-terminal ubiquitin-associated (UBA) domain. Analysis of mutants lacking early endocytic adaptors reveals that the ubiquitin-binding domains (UBDs) of Ent2 and Ede1 are likely to function primarily to mediate protein–protein interactions between components of the early endocytic machinery. Cells that lack epsin and Ede1 UBDs are able to internalize activated, ubiquitinated receptors. Furthermore, under conditions in which epsin UIMs are important for receptor internalization, receptors internalized via both ubiquitin-dependent and ubiquitin-independent signals require the UIMs, indicating that UIM function is not restricted to ubiquitinated receptors. Epsin UIMs share function with non-UBD protein–protein interaction motifs in Ent2 and Ede1, and the Ede1 UBA domain appears to negatively regulate interactions between endocytic proteins. Together, our results suggest that the ubiquitin-binding domains within the yeast epsin Ent2 and Ede1 are involved in the formation and regulation of the endocytic network.  相似文献
