P53 is an apoptosis marker which is involved in determining nucleus pulposus (NP) cell fate. Little is known about P53 interaction with N-Myc downstream-regulated gene 2 (NDRG2) in intervertebral disc degeneration (IVDD). Here, we studied the role of the P53-NDRG2 axis in IVDD. We found that NDRG2 was expressed in NP tissue obtained from patients with IVDD. The level of NDRG2 was positively related to the severity of IVDD, as determined by Pfirrmann grading. Subsequently, we overexpressed NDRG2 in human NP cells by adenoviral transfection and studied the effects of increased levels of NDRG2 on the viability and apoptosis of these cells. NDRG2 overexpression induced NP cell apoptosis and reduced viability in NP cells obtained from patient with IVDD. We also found that the level of P53 was elevated in NP cells from patients with IVDD and treatment with exogenous P53 upregulated NDRG2 in NP cells. Last, IVDD model was established in P53 knockout mice and the pathological changes in the intervertebral discs and NDRG2 expression were examined. P53 knockout can reduce the damage of NP tissues after IVDD surgery to some extent. Restoration of NDRG2 antagonized the effect of P53 knockout on IVDD. Collectively, this study suggests that elevated P53 in NP cells stimulates apoptosis of the cells by upregulating NDRG2 expression, thereby exacerbating IVDD. 相似文献

探讨红车轴草(TrifoliumpratenseL)提取物对胃癌细胞株BGC-823的抑制增殖效应及诱导凋亡作用.我们采用不同浓度(50、100、250、500、1000mg/L)红车轴草提取物处理BGC-823细胞,采用四甲基偶氮唑盐(MTT)法检测药物对细胞的抑制作用,倒置显微镜观察细胞的形态学改变;AO/EB染色,荧光显微镜观察细胞凋亡形态;采用DNALadder观察DNA的降解;应用流式细胞仪观察细胞凋亡过程中的细胞周期的变化和细胞凋亡.结果显示在红车轴草提取物的作用下,BGC-823细胞呈凋亡改变,DNA琼脂糖凝胶电泳呈典型的凋亡特征.细胞凋亡的同时,细胞周期阻滞于G2/M期.实验结果表明红车轴草提取物能抑制胃癌BGC-823细胞增殖,能诱导胃癌细胞凋亡. 相似文献

This study investigated the age-dependent changes in the number of BrdU- and TUNEL-positive cells in murine gingival tissue
and submandibular gland, and compared the findings with those in other tissues and organs. The cell proliferative activity
was decreased after 20 weeks of age in epithelial cells of the gingiva, tongue, buccal mucosa and skin. A decreased cell proliferative
activity was also associated with aging in the liver and kidney parenchymal cells. Meanwhile, cell death showed peculiar changes
in gingival subepithelial tissue, and mucous and serous acini of the submandibular gland. An increase of TUNEL-positive cells
was demonstrated in gingival subepithelial tissue after 20-week-old of age. A significant increase of TUNEL-positive cells
was also found in the mucous acinar cells in the 20-week-old mice and in the serous acini after 20 weeks. The fluctuation
in the number of TUNEL-positive cells in the subepithelial tissue of the skin, and BrdU- and TUNEL-positive staining ratios
in the liver was smaller than that in other tissue and organs throughout life. This study may provide useful information for
better understanding the influence of aging on the functional alteration that occurs in the gingival tissue and submandibular

Six condition indices based on RNA, total soluble protein and two metabolic enzymes [lactate dehydrogenase (LDH) and citrate synthase (CS)] were analysed in muscle tissue of individual larvae, post-flexion reared sea bass Dicentrarchus labrax using DNA and total soluble protein as standards for size. In addition, the effect of 2 days of food deprivation on the cell proliferation rates was assessed. The RNA:DNA best reflected short-term changes in feeding conditions. If standardized by DNA content, LDH activity was a better indicator of condition than any other index but RNA:DNA. Further, the analysis of cell proliferation rates in muscle from 26 day-old larvae proved useful in distinguishing continuously fed larvae from individuals subjected to 2 days of fast. 相似文献

Cells employ pro-survival and pro-adaptive pathways to cope with different forms of environmental stress. When stress is excessive, and the damage caused by it is unsustainable, cells engage pro-death pathways, which are in place to protect the host from the deleterious effects of harmed cells. Two important pathways that determine the balance between survival and death of stressed cells are the integrated stress response (ISR) and the mammalian target of rapamycin (mTOR), both of which converge at the level of mRNA translation. The two pathways have established avenues of communication to control their activity and determine the fate of stressed cells in a context-dependent manner. The functional interplay between the ISR and mTOR may have significant ramifications in the development and treatment of human diseases such as diabetes, neurodegeneration and cancer. 相似文献

以来自哈尔滨传染性法氏囊病病毒(IBDV) 强毒株(Harbin 毒株,H) 的基因组RNA为模板,用反转录聚合酶链反应(RT- PCR) 的方法得到了其A 节段的全长cDNA 片段,分5'端(1 659bp) 和3'端(1 444bp) 上下两段分别克隆到pGEMB○R - T 载体上,测定了其核苷酸顺序,在长为3 101 bp 中含有两个阅读框ORFA1 和ORFA2 ,分别编码1 012 个氨基酸的前体蛋白(VP2 - 4 -3) 和145 个氨基酸的VP5,ORFA1 和ORFA2 有部分的重叠。将核苷酸序列及推测出的氨基酸序列与已报道的IBDV 血清Ⅰ型和Ⅱ型毒株的相应序列进行了比较,结果表明:H 毒株与其它血清Ⅰ型毒株之间,在核苷酸水平上存在25bp - 267bp 的差异;在氨基酸水平上存在17 ~40 个氨基酸的差异。在VP2 - 4 - 3 内比较显示,H 毒株与P2 、Cu- 1 之间氨基酸的差异最小为1 .7% ,H 毒株与UK661 之间氨基酸的差异最大为3 .9 % 。变异主要发生在VP2 的可变区(206 - 350 位氨基酸) ,在H 毒株所特有的12 个氨基酸当中,该区就占5 个,代表1 .76 % 的变异。VP4、VP3 和VP5区各有 相似文献

Rapid method of in vitro multiplication of date palm was developed. Shoot tips were cultured on Murashige and Skoog (MS) medium supplemented with 2 mg dm–3 dimethylaminopurine (2iP) + 1 mg dm–3 naphthalene acetic acid (NAA). Shoot buds were proliferated from white nodular cultures on hormone free medium. Shoot bud proliferation strongly enhanced when cultured on MS-medium contained 3 mg dm–3 2iP + 0.5 mg dm–3 NAA. Culturing on full-strength MS medium showed higher multiplication rate compared with half-strength MS medium. Among four concentrations of sucrose used, 30 g dm–3 speeded up the bud proliferation more than 10, 20 and 40 g dm–3. However, the largest shoot buds were observed with 40 g dm–3 sucrose. Solidification of culture media by 1.75 g dm–3
