Procedures for asymbiotic seed germination and seedling acclimatization were developed for Bletia purpurea, a threatened North America native terrestrial orchid. Six asymbiotic orchid seed germination media (Knudson C, PhytoTechnology Orchid Seed Sowing Medium, Malmgren Modified Terrestrial Orchid Medium, Vacin &; Went Modified Orchid Medium, ½-strengh Murashige &; Skoog, and BM-1 Terrestrial Orchid Medium) were examined for their effectiveness in promoting seed germination and protocorm development of B. purpurea in either a 0/24 h or 16/8 h L/D photoperiod. Germination occurred regardless of medium or photoperiod treatment. However, advanced seedling development (Stage 6) only occurred on Vacin &; Went Modified Orchid Medium in the 16/8 h L/D photoperiod. Further effects of photoperiod on in vitro seedling development were also examined. Shoot length, leaf width, root number and length, and fresh weight and dry weight in the 16/8 h L/D photoperiod were all significantly different when compared to the 8/16 h and 12/12 L/D photoperiods. In vitro seedlings were readily acclimatized to greenhouse conditions. Seedlings showed high survival all potting media. Seedlings acclimatized in Fafard Mix 4 potting medium developed significantly longer roots. Corm formation occurred regardless of potting media used.  相似文献

We tested the effects of cold stratification, temperature, light and NaCl on seed germination and germination recovery and of NaCl on radicle growth and radicle elongation recovery of Kalidium caspicum, a small leafy succulent shrub dominant in saline deserts in northwest China. In all conditions of temperature and light/darkness, germination percentages and rates of cold-stratified seeds were significantly higher than those of nonstratified seeds. Germination of a high percentage of both nonstratified and stratified seeds was inhibited by 0.2 M NaCl, and 0.6 M NaCl completely inhibited germination. Nongerminated seeds germinated after they were transferred from NaCl solutions to distilled water. Radicle elongation significantly decreased with increase in salinity, and it was completely inhibited by ≥1.0 M NaCl; radicle elongation recovered in young seedlings pretreated by 10 days of incubation in ≤0.4 M NaCl. Results show that seed germination and early seedling growth of K. caspicum are salt tolerant, and these characteristics help explain why this species can survive and dominate salt habitats, such as those in the Junggar desert in Xinjiang, northwest China.  相似文献

Herbivores may enhance plant recruitment, but such positive interactions may be overlooked in favour of obvious negative effects of herbivory on propagules. My objective was to determine whether larval insects that feed and develop within fruit of the mangrove Avicennia marina act as mutualist herbivores by increasing the dispersal of propagules without affecting their viability and emerging successfully as adults following dispersal of the propagule by water. Surveys revealed that frugivory is common throughout the mangrove forest, and fruit had up to six exit holes where larvae had emerged as adults. Larval insects did not affect the flotation of propagules with pericarps, a thin structure that provides buoyancy for dispersal by water. In contrast, after simulating germination by removing the pericarp, the majority of propagules with three exit holes floated on average for 20 h longer than those without exit holes, which sank immediately. Based on this evidence that frugivory could increase the dispersal potential of propagules, I predicted that propagules consumed by larval insects should disperse farther than undamaged propagules, and this was tested by quantifying the potential viability of propagules stranded on beaches at increasing distances (up to 20 km) from mangrove forests. Flies and moths emerged as adults after being transported tens of kilometres within mangrove propagules, revealing a novel mode of dispersal. Proportionally fewer potentially viable propagules were supplied to beaches at increasing distances from mangrove forests, however, indicating that larval insects negatively affect recruitment and are thus not acting as mutualist herbivores. Nevertheless, when transported back to the mangrove forest, seedlings established from propagules damaged by larval insects and stranded on beaches. Therefore, although frugivory does not preclude mangrove recruitment, its negative effects in the pre-dispersal environment may be intensified with increasing dispersal distance, thus limiting the long-distance supply of propagules and recruitment of mangroves.  相似文献

A xanthone derivative, named gaboxanthone (1), has been isolated from the seed shells of Symphonia globulifera, together with known compounds, symphonin (2), globuliferin (3), guttiferone A (4), sistosterol, oleanolic acid and methyl citrate. The structure of the compound was assigned as 5,10-dihydroxy-8,9-dimethoxy-2,2-dimethyl-12-(3-methylbut-2-enyl) pyrano [3,2-b]xanthen-6(2H)-one, by means of spectroscopic analysis. The anti-plasmodial and antioxidant activities of the phenolic compounds were evaluated, respectively, in culture against W2 strain of Plasmodium falciparum and using the free radical scavenging activity of the DPPH radical, respectively. Compounds 1-4 were found to be active against the Plasmodium parasites (IC(50) of 3.53, 1.29, 3.86 and 3.17 microM, respectively). Guttiferone A (4) showed a potent free radical scavenging activity compared to the well-known antioxidant caffeic acid.  相似文献

Arabidopsis thaliana is frequently used as a model for the study of oilseed biology and metabolism. However, the very small seeds of Arabidopsis can complicate analysis of their oil content and influence the application of results to larger-seeded plants. Here, we describe how seed anatomy, light, and plant-to-plant variation influence the content and measurement of oil in Arabidopsis seeds. The anatomy of Arabidopsis and Brassica napus seeds were compared and the distribution of mass, oil and the fatty acid composition of different seed parts were determined. In Brassica, 90% of the seed oil resides in the cotyledons that contribute 74% of seed mass. By contrast, the values for Arabidopsis are 60% and 45%, respectively, with a higher fraction of the oil deposited in the radicle, hypocotyl, endosperm and seed coat. Growth of Arabidopsis plants with 600 micromol m(-2) s(-1) light resulted in a two-fold higher seed yield, a 40% increase in mass per seed and a 60% increase in oil per seed compared to growth at 100 micromol m(-2) s(-1). Factors that influence the analysis of oil content were evaluated. Intact-seed transmethylation followed by gas chromatography (GC) analysis provided reproducible analysis of Arabidopsis seed oil. However, plant-to-plant variation in oil content is large and we analyzed how this influences the ability to detect statistically valid changes in oil between different genotypes. These observations establish a reference data set on the fatty acid composition and distribution of mass and oil between tissues of Arabidopsis seeds that should help to predict the applicability of results obtained with Arabidopsis to other oilseeds.  相似文献

Plants exude a variety of substances through their roots, germinating seeds and aerial parts. Some of these released compounds seem to have an inhibitory effect against pathogens. The aim of this work was to investigate and identify antifungal proteins present in exudates from imbibed cowpea seeds (Vigna unguiculata (L.) Walp). The obtained exudation was analyzed in regard to specific protein activities by enzymatic or immunological assays for plant defense proteins, from 4 h to 48 h of seed imbibition. Our results show that cowpea seeds exudates present several defense related proteins characterized as β-1,3-glucanases, cystatins, vicilins and lipid transfer proteins (LTPs), as well as a storage vacuole membrane α-TIP protein, since the very first hours of imbibition. These exudates also have an “in vitro” inhibitory effect on the growth of the fungus Fusarium oxysporum f. sp. phaseoli. Our results suggest that seed exudates should promote seed protection from soil pathogens.  相似文献

Pyruvate, orthophosphate dikinase (PPDK; E.C.2.7.9.1) is most well known as a photosynthetic enzyme in C4 plants. The enzyme is also ubiquitous in C3 plant tissues, although a precise non-photosynthetic C3 function(s) is yet to be validated, owing largely to its low abundance in most C3 organs. The single C3 organ type where PPDK is in high abundance, and, therefore, where its function is most amenable to elucidation, are the developing seeds of graminaceous cereals. In this report, we suggest a non-photosynthetic function for C3 PPDK by characterizing its abundance and posttranslational regulation in developing Oryza sativa (rice) seeds. Using primarily an immunoblot-based approach, we show that PPDK is a massively expressed protein during the early syncitial-endosperm/-cellularization stage of seed development. As seed development progresses from this early stage, the enzyme undergoes a rapid, posttranslational down-regulation in activity and amount via regulatory threonyl-phosphorylation (PPDK inactivation) and protein degradation. Immunoblot analysis of separated seed tissue fractions (pericarp, embryo + aleurone, seed embryo) revealed that regulatory phosphorylation of PPDK occurs in the non-green seed embryo and green outer pericarp layer, but not in the endosperm + aleurone layer. The modestly abundant pool of inactive PPDK (phosphorylated + dephosphorylated) that was found to persist in mature rice seeds was shown to remain largely unchanged (inactive) upon seed germination, suggesting that PPDK in rice seeds function in developmental rather than in post-developmental processes. These and related observations lead us to postulate a putative function for the enzyme that aligns its PEP to pyruvate-forming reaction with biosynthetic processes that are specific to early cereal seed development.  相似文献

Seeds with ‘intermediate’ storage physiology store poorly under cold and dry conditions. We tested whether the poor shelf life can be attributed to triacylglycerol phase changes using Cuphea carthagenensis (Jacq.) seeds. Viability remained high when seeds were stored at 25°C, but was lost quickly when seeds were stored at 5°C. Deterioration was fastest in seeds with high (≥0.10 g g−1) and low (0.01 g g−1) water contents (g H2O g dry mass−1), and slowest in seeds containing 0.04 g g−1. A 45°C treatment before imbibition restored germination of dry seeds by melting crystallized triacylglycerols. Here, we show that the rate of deterioration in C. carthagenensis seeds stored at 5°C correlated with the rate that triacylglycerols crystallized within the seeds. Lipid crystallization, measured using differential scanning calorimetry, occurred at 6°C for this species and was fastest for seeds stored at 5°C that had high and very low water contents, and slowest for seeds containing 0.04 g g−1. Germination decreased to 50% (P50) when between 16 and 38% of the triacylglycerols crystallized; complete crystallization took from 10 to over 200 days depending on water content. Our results demonstrate interactions between water and triacylglycerols in seeds: (1) water content affects the propensity of triacylglycerols to crystallize and (2) hydration of seed containing crystallized triacylglycerols is lethal. We suggest that these interactions form the basis of the syndrome of damage experienced when seeds with intermediate storage physiologies are placed in long-term storage.  相似文献

小百草岭具有野生种子植物141科、576属、1406种。区系统计分析表明:(1)科级组成中,热带性质的科有55科,占总科数的56.12%,具较强的热带性质;属级组成中,温带性质的属有293属,占总属数的56.35%,表现出较强的温带性质,从科到属的区系成分变化表明:古热带起源的区系在喜马拉雅造山运动及滇中高原形成过程中,区系成分逐渐分化发展,在亚热带山地相应的海拔地段保留了相应的喜热成分,又分化出适应亚热带山地的喜温成分,从而使其区系的整体外貌呈现出热、温带成分的共存发展,是亚热带山地植物区系的一个基本特征;(2)区系成分新老兼备,既有第三纪古老孑遗的类群,也有新近分化衍生的类群,众多的中国特有分布型(种)中,表现出与周边地区较为广泛的地理联系,而与横断山地区共有的成分占据了明显优势,揭示了两地间紧密的区系联系。  相似文献
