We have expressed two T7 RNA polymerase genes by electroporation into tobacco protoplasts. One of the genes was modified by inserting nucleotides encoding a viral nuclear localization signal (NLS) from the large T antigen of SV40. Both T7 RNA polymerase genes directed synthesis of a ca. 100 kDa protein in the electroporated protoplasts. T7 RNA polymerase activity was detected in extracts of protoplasts electroporated with both genes. Immunofluorescence analysis of these protoplasts indicated that only the polymerase carrying the NLS accumulated in the cell nucleus. These experiments suggest that mechanisms involved in the transport from the cytoplasm to the nucleus are similar in plant and animal cells. This system demonstrates the feasibility of T7 RNA polymerase-based approaches for the high-level expression of introduced genes in plant cells.  相似文献

Summary The inheritance of yellow berry, a grain disorder in durum and bread wheats, was studied in six intervarietal crosses in bread wheat. The trait was found to be controlled by either two or three dominant genes. Monosomic analysis using Chinese Spring monosomic series showed the presence of two major dominant genes on chromosomes 1A and 7A, and four modifiers on 4A, 4B, 6A and 6D, which influence the expression of yellow berry in bread wheat.  相似文献

We studied mechanisms of vegetation change in fens subject to succession from open water to floating mats and finally herbaceous rich-fens. Earlier research showed that these systems are characterized by transient seed banks. Our main question was whether seedlings of later successional fen stages are already present in earlier stages, remaining subordinate in the vegetation until conditions become suitable for them. If, however, conditions during succession change in a way that only a limited set of species can survive as seedlings during each of the successional stages, no seedling bank will exist. The transient character of the seeds would then imply that seeds will not germinate and will subsequently die and that seeds that have germinated in the “wrong” stage will not become established. We hypothesized that: (1) germination and seedling survival of fen species are significantly better in the successional fen stage for which these species are characteristic, (2) as a consequence no seedling banks occur in these fens. In a field experiment, seeds of five characteristic fen species in the standing vegetation of three successional fen stages i.e. raft fen, quaking fen and rich fen were sown in each of these stages in a turf pond in the Tienhoven area, The Netherlands. Germination and seedling survival were measured over two growing seasons together with environmental variables. Germination was higher in the “own” stage for all species groups as was survival for quaking fen species and rich fen species. For both these stages, percentage of germination and survival of four out of five characteristic species were significantly higher in the “own” stage. Germination and survival can be considered stage-dependent and it was concluded that seedling banks do not exist in these fens. Site-specific environmental variables act as a sieve and differentiate on species presence already during early life history stages. We found clues that the environmental sieve acts at the level of nutrient availability, tolerance for high sulphide concentration and light climate. Because of the transient seed bank and absence of a seedling bank in these fen wetlands, successful establishment of species necessitates a continuous dispersal of characteristic species until the environmental conditions permit establishment. This also implies that species of the whole successional sere should be present within dispersal distance.  相似文献

Question: The vegetation in a polder after partial tidal restoration does not resemble the targeted salt‐marsh vegetation. Is this difference in vegetation due to lack of dispersal or unsuitable abiotic conditions? What could be done for a better restoration of the site? Location: Northwestern France. Methods: Seeds were trapped at the single inlet of the polder with a 200‐μ m mesh net to estimate inputs of seeds from the bay. In parallel, seed dispersal was studied in the polder by placing Astroturf® seed traps on the surface of the sediment at three different elevations in three distinct areas. Abiotic conditions such as flooding frequency, water table level and soil salinity were monitored. Results: All but one species from the adjacent salt marshes were trapped at the inlet. Not all of these species were on the seed traps inside the polder. Seed dispersal was not homogeneous in the polder and seed trap content mostly discriminated in function of their elevation. Salinity and water logging at the bottom of the slope were very high compared to tolerance of most halophytes but decreased rapidly higher up the slope. Conclusions: The development of salt marsh target species is highly restricted by limited hydrochory inside the polder but also by unfavourable soil conditions induced by the actual hydrological regime. Halophytes are excluded at the bottom of the slope by abiotic conditions and out‐competed by sub‐halophytes higher up. In order to restore salt marsh vegetation inside the polder, a larger opening should be induced in order to increase the flooded surface, and diminish water logging and flooding frequencies.  相似文献

Objective: This empirical study was designed to explore the role of ecological features of species in the spatial patterning of a grassland community. Location: Banks of the river Rhône in France. Material and Methods: First, we explored the spatial pattern of 29 species recorded in the community using spatial autocorrelation analysis of species cover values. Second, we then explored the relationship between the patterns found and a set of life attributes that characterized the ecological features of species for resource foraging or dispersion. Finally, we explored the spatial relationship of groups of species that shared the same ecological features using cross‐correlation analysis. Results: We found a significant relationship between the spatial pattern and life attributes of the species highlighting three groups of species: (1) species characterized as competitors, reproducing by runner clonal organs and forming large, dense patches; (2) species characterized as competitive‐rud‐erals, dispersing exclusively by seed production and forming small periodic patches; and (3) species classified as CSR, characterized by rosette morphology and short rhizomes as clonal organs without any significant spatial autocorrelation. Spatial segregation was found between group 1 and group 2 up to 14 m; no significant cross‐correlation between groups 1 and 3 between 0 and 3.5 m, and association between groups 2 and 3 up to 14 m. Conclusions: These results helped to understand how species attributes (relative to stature or dispersion abilities); external factors (such as disturbance) and biotic processes (competition) interplay in structuring the plant community under study in space.  相似文献

Questions: 1. What is the scale and extent of spatial variability in factors affecting Betula invasion of heaths? 2. How much effect does each factor have on within‐patch patterns of invasion? 3. How can this understanding aid in managing Betula invasions? Location: Lowland heath of southern England. Methods: Determinants of Betula (both B. pubescens and B. pendula) invasion: biomass density, necromass density, mean vegetation height, P‐availability, soil water content and total Betula seed bank density, were measured at two sites on a 5‐ha sampling grid. Spatial pattern was assessed using geostatistics. Contributions of each determinant to within‐site heterogeneity in predicted Betula seedling densities were estimated by varying variables over their full and interquartile ranges in a statistical model derived from experimental data. Results: Salient spatial trends were revealed: strong autocorrelation over distances of < 50 m for soil factors and more extensive autocorrelation (0 to > 150 m) in vegetation variables and Betula seed bank densities. The latter resulted in single across‐site gradients, the former small, distinct patches. All patterns were overlain with variance that was present at distances of < 17.6 m. Variables displaying spatial pattern also accounted for within‐site heterogeneity in predicted Betula seedling densities but their relative contribution to this varied between sites. Conclusions: Identifiable spatial autocorrelation in factors controlling patch‐scale invasion patterns allows managers to target invasion prone patches, potentially reducing management intensities. Furthermore, management effort may be optimised by spatially de‐coupling Betula seed from safe‐sites. This plan may adaptable to the management of other weeds and open‐land ecosystems.  相似文献

The selection of appropriate seeds is essential for the success of phytoremediation/restoration projects. In this research, the growth and elements uptake by the offspring of mesquite plants (Prosopis sp.) grown in a copper mine tailing (site seeds, SS) and plants derived from vendor seeds (VS) was investigated. Plants were grown in a modified Hoagland solution containing a mixture of Cu, Mo, Zn, As(III) and Cr(VI) at 0, 1, 5 and 10 mg L−1 each. After one week, plants were harvested and the concentration of elements was determined by using ICP-OES. At 1 mg L−1, plants originated from SS grew faster and longer than control plants (0 mg L−1); whereas plants grown from VS had opposite response. At 5 mg L−1, 50% of the plants grown from VS did not survive, while plants grown from SS had no toxicity effects on growth. Finally, plants grown from VS did not survive at 10 mg L−1 treatment, whilst 50% of the plants grown from SS survived. The ICP-OES data demonstrated that at 1 mg L−1 the concentration of all elements in SS plants was significantly higher compared to control plants and VS plants. While at 5 mg L−1, the shoots of SS plants had significantly more Cu, Mo, As, and Cr. The results suggest that SS could be a better source of plants intended to be used for phytoremediation of soil impacted with Cu, Mo, Zn, As and Cr.  相似文献

ADP-glucose pyrophosphorylase (AGPase) plays a key role in regulating starch biosynthesis in cereal seeds and is likely the most important determinant of seed strength. The Escherichia coli mutant glgC gene (glgC16), which encodes a highly active and allosterically insensitive AGPase, was introduced into maize (Zea mays L.) under the control of an endosperm-specific promoter. Developing seeds from transgenic maize plants showed up to 2–4-fold higher levels of AGPase activity in the presence of 5 mM inorganic phosphate (Pi). Transgenic plants with higher cytoplasmic AGPase activity under Pi-inhibitory conditions showed increases (13–25%) in seed weight over the untransformed control. In addition, in all transgenic maize plants, the seeds were fully filled, and the seed number of transgenic plants had no significant difference compared with that of untransformed control. These results indicate that increasing cytoplasmic AGPase activity has a marked effect on sink activity and, in turn, seed weight in transgenic maize plants.  相似文献
