Summary Chromaffin granules from bovine adrenal medullary chromaffin cells have been found to contain small vesicular structures bounded by unit membranes. Detection of these intragranular vesicles within intact cells requires the use of quick-freezing methods. The intragranular vesicles are labile to fixation by aldehydes which explains why they have not been described in intact cells until now. They are found in approximately 60% of the dense-core chromaffin granules in cells and 85% of isolated granules. They are usually clustered in groups of one to as many as five between the core and the inner surface of the granule membrane. The intragranular vesicles are independent vesicles in that they do not appear as simple invaginations of the granule membrane in either serial thin-section or freeze-etch views. Furthermore, they are released from the cell along with granule contents during nicotine-induced secretion of catecholamines. The structural heterogeneity provided by the intragranular vesicles may be related to the functional heterogeneity of granule contents observed in many recent biochemical studies.  相似文献

Representative conditional yeast secretory mutants, blocked in transport of secretory and plasma membrane proteins from the endoplasmic reticulum (sec 18), from the Golgi body (sec 7) and in transport of secretory vesicles (sec 1), accumulated exoglucanase, a constitutive yeast activity, when incubated at the restrictive temperature (37°C). Different proportions of the accumulated activity were released by mutant cells under permissive conditions. The presence or absence of cycloheximide during the secretion period made no differences in the results. More than 90% of the internal activity was bound to membrane in wild type cells. However, only the soluble pool underwent changes during the accumulation or secretion periods. The bulk of secretory invertase accumulated by sec 1 was also soluble. By contrast sec 7 and sec 18 accumulated membrane-bound as well as soluble invertase forms and both were secreted in similar proportions in each mutant. More than 90% of the accumulated invertase was secreted at the permissive temperature in sec 18 cells. That percentage was significantly lower for exoglucanase (<65%). Concomitantly, invertase accumulated by this mutant exited from the cells with a lower half time (t 1/2=150 min). Résumé Chez le Chilopode Lithobius forficatus L., les cellules parenchymateuses des glandes cérébrales présentent, durant l'intermue, un cycle sécrétoire caractérisé essentiellement par l'évolution de l'activité golgienne. Pendant les périodes de sécrétion, les ribosomes, groupés en polysomes, et les éléments ergastoplasmiques sont très abondants; pendant les périodes de repos, les ribosomes sont isolés et l'ergastoplasme est vésiculeux. Les autres organites cellulaires montrent peu de variations.Le cycle sécrétoire ne se superpose pas exactement au cycle d'intermue; il commence lors de la prémue. Peu de temps après l'exuviation, il est possible d'observer des figures qui semblent liées au rejet du produit de sécrétion. L'étude ultrastructurale des glandes cérébrales en fonction de l'intermue confirme l'existence de l'activité cyclique mise en évidence par les résultats expérimentaux.
Ultrastructural signs of cyclic secretory activity in the cerebral glands of Lithobius forficatus during intermolt
Summary During the molting cycle of Lithobius forficatus L. (Chilopoda), the parenchymal cells of the cerebral glands exhibit signs of cyclic secretory activity consisting essentially of changes in the appearance of the Golgi complex. During active periods of secretion, polysomes and ergastoplasmic elements are very numerous; free ribosomes and vesiculated ergastoplasmic cisternae characterize the periods of inactivity. Other cytoplasmic organelles undergo few variations. The secretory cycle as determined by cytological criteria does not exactly match the molting cycle. The first ultrastructural signs of activity appear during premolt. Shortly after ecdysis, images suggesting the release of secretory material can be observed. The electron microscopic data on cyclic changes during intermolt substantiate the existence of activity cycles in the cerebral glands as determined by physiological experiments.

本文把气孔及其下腔看作截面为椭圆形的柱形区域,提出一个水汽从气孔下腔内所有细胞表面扩散到气孔外端的三维扩散模型。根据 Fick 定律和质量守恒定律建立了支配该模型的水汽扩散方程。用有限差分法,借助于计算机求得水汽从气孔下腔的所有细胞表面扩散到气孔内端所遇到的阻力及其近似表达式。并从理论上对该阻力的倒数——导度随气孔面积而变化的方式做了分析和解释。通过将本模型求得的气孔下腔阻力计算公式与 Brown 等以及 Cooke 的公式比较,发现在气孔开度变化相当大的范围内用后面两公式计算的阻力偏大0.5—1倍左右。此外,计算结果还表明:在气孔下腔水散失总量中,腔内表皮细胞表面上的水散失量占86—96%,而保卫细胞表面上的水散失量又占后者的88—93%,副卫细胞表面上的水散失量仅7—12%。  相似文献

Abstract: An increase in extracellular Ca2+ concentration from 0.25 to 10 m M enhanced secretion of norepinephrine and epinephrine induced by a high extracellular K+ concentration (75 m M ). The increment in extracellular Ca2+ concentration also increased the observed peak inward Ca2+ current in response to long (10-s) depolarizing pulses from a holding potential of −55 mV to +5 mV, from about −26 to −400 pA. However, the total amount of Ca2+ influx into the cell only increased when the extracellular Ca2+ concentration was raised from 0.25 to 1 m M and then remained constant up to 10 m M extracellular Ca2+. ATP is cosecreted with catecholamines following a depolarizing stimulus. Kinetic studies indicated that ATP secretion had two components with time constants, in the presence of 2.5 m M extracellular Ca2+, of ∼4 and 41 s, being the fast component of secretion produced by the exocytosis of ∼220 chromaffin granules. The results suggest that, for a given depolarizing stimulus, the size and rate of release for the fast and slow components of secretion are dependent on extracellular Ca2+ concentration.  相似文献

The role of fructan metabolism in the assimilate relations of the grain of wheat (Triticum aestivum L.) was investigated by determination of the dry matter and fructan content of grain components at short intervals during grain filling. During the initial phase of rapid expansion, most of the assimilates entering the grain were partitioned to the outer pericarp. A large fraction of these assimilates were used for the synthesis of fructan. Dry matter deposition and fructan synthesis in the outer pericarp ceased at about 5d after anthesis. At the same time, the endosperm and the inner pericarp and testa started to accumulate dry matter at a fast rate. This was also associated with significant fructan synthesis in the latter tissues. The outer pericarp lost about 45% of its former maximum dry weight between 9 and 19 d after anthesis. This loss was due almost entirely to the near complete disappearance of water-soluble carbohydrates, most of which was fructan. The inner pericarp and testa accumulated dry matter until about mid-grain filling. The fructan contents of the inner pericarp and testa and the endosperm decreased slowly towards the end of grain filling. Most of the fructans in the inner pericarp and testa and the endosperm had a low molecular weight, whereas higher molecular weight fructans predominated in the outer pericarp. The embryo did not contain fructan. The presence of low molecular weight fructans in the endosperm cavity at mid-grain filling was confirmed. It is suggested that fructan synthesis is closely linked to growth-related water deposition in the different tissues of the wheat grain and serves to sequester the surplus of imported sucrose.  相似文献

The effect of lysosomal storage diseases on the ultrastructure of human mast cells has not previously been reported. Indeed, there has been little published evidence indicating that mast cells contain typical lysosomes. However, mast cell cytoplasmic granules contain hydrolases similar to those found in lysosomes, but which differ from lysosomal hydrolases in exhibiting optimal activity at higher pH. We therefore examined by transmission electron microscopy the dermal mast cells in 58 biopsies of patients exhibiting 1 of 29 different lysosomal storage diseases. We found mast cells containing abnormal lysosomes in 16 of these disorders. In 6 of these 16 diseases, the mast cells' cytoplasmic granules appeared normal. These observations indicate that human mast cells can contain lysosomes, and provide evidence that the enzymes affected by lysosomal storage diseases are active in mast cells.  相似文献

Summary Growing hyphal tips of higher fungi contain an organized assemblage of secretory vesicles and other cell components collectively known as the Spitzenkörper. Until now, the Spitzenkörper has been portrayed as a single spheroid complex located near the apical cell wall. This study demonstrates the occurrence of multiple Spitzenkörper in growing hyphal apices imaged by video-enhanced phase-contrast microscopy. In addition to the main Spitzenkörper, smaller satellite Spitzenkörper arise a few micrometers behind the apical pole. Four developmental stages were identified: (a) the satellites first appeared as faint phase-dark plaques next to the plasma membrane, (b) gradually increased in size and assumed an ovoid profile, (c) they migrated to the hyphal apex, and (d) finally they merged with the main Spitzenkörper. After the merger, the main Spitzenkörper temporarily increased in size. Satellites were observed in 14 fungi, most of which had relatively large (5–10 m diam.), fast-growing hyphae (2–33 m/min elongation rate). The average frequency of in-focus satellites was 7+/min forFusarium culmorum and 11+/min forTrichoderma viride. As with the main Spitzenkörper, satellites were present only in growing cells. They were transient and remained visible for 3–8 s before merging with the main Spitzenkörper. Within the hyphae, satellites travelled up to six times faster than the average cell elongation rate. Multiple satellites sometimes occurred simultaneously; up to three were seen within a hyphal apex at the same time. Localized cell enlargement occurred next to stationary satellites, suggesting that satellite Spitzenkörper are functional as sources of new cell surface before they reach the main Spitzenkörper; therefore, they account for some variations in the profiles of the growing hyphae. By electron microscopy, satellites consisted of small clusters of apical vesicles surrounding a group of microvesicles located next to the plasma membrane. The identification and behavior of the satellites represent clear evidence of directional mass transport of vesicles toward the hyphal apex. Our observations indicate that satellites are a common phenomenon in growing hyphal apices of septate fungi and that they contribute to growth of the hyphal apex.Abbreviations VSC vesicle supply center  相似文献
