The development of thermophysiological responses during four consecutive exercise/rest sessions in the cold was studied in men wearing chemical protective clothing and a face mask. Six men repeated four exercise/rest sessions during 8 h at –10°C. Each session consisted of step exercise (240 W · m−2) for 60 min and rest for another 60 min. Rectal and skin temperatures were measured continuously and thermal sensations were obtained at 30-min intervals. Entering the cold from a warm environment and the onset of exercise resulted in a decrease in skin temperatures during the first session and the decrement in the temperatures of the extremities continued for 10–20 min during the following period of exercise. Torso skin temperature was at its lowest during the first rest period. After the first session of cold exposure the range and the level of variation in mean body temperature ( b) followed a pattern which was repeated until the end of the experiment. However, the torso skin temperatures increased gradually until the fourth session, while the temperatures of the extremities, in contrast, tended to decrease up to the third session. In conclusion, the present results indicated that although b, reflecting the whole body heat balance, showed a typical pattern of change after the first session (2 h), the torso area was warming until the end of the cold exposure while the extremities continued to cool down up to the third session (6 h), obviously due to a prolonged redistribution of the circulation. Zusammenfassung Der Einfluß von p-Chlorphenylalanin-methylester-hydrochlorid (PCPA) und Reserpin auf die biogenen Amine des Glomus caroticum von Kaninchen wurde ultrastrukturell und fluoreszenzmikroskopisch untersucht. Die elektronenmikroskopische Analyse ergab keine eindeutigen Kriterien für arzneimittelinduzierte Veränderungen. Fluoreszenzmikroskopisch ließ sich nach Applikation von Reserpin eine ausgeprägte Senkung des Catecholamin- und Indolamin-Gehaltes und nach PCPA eine Abnahme des Serotonins erkennen.
Electron microscopic and fluorescence microscopic studies of biogenic amines in the rabbit glomus caroticum after treatment with reserpine and PCPA
Summary The effects of p-chlorophenylalanine-methylester-hydrochloride (PCPA) and reserpine on biogenic amines of the rabbit carotid body were investigated ultrastructurally and by fluorescence microscopy. The electron microscopic analysis did not indicate significantly that structural changes result from treatment with reserpine or PCPA. Fluorescence microscopy indicated that PCPA lowered serotonin, and reserpine lowered both catecholamines and indolamines.

Summary Sectors of anterior segments of vervet eyes were exposed to solutions of different osmolarities (Cercopithecus aethiops). After hypertonic incubation followed by isotonic fixation, as well as after fixation directly in hypertonic fixative, the ciliary epithelium showed constant changes. These changes consisted of a shrinkage pattern with dilations of intercellular clefts in the superficial region of the epithelium, whereas no dilations occurred in the basal layer. The basal junctional complex of the superficial cell layer was always intact. The detailed structure of this complex is described. The conclusion is drawn that it functions as a barrier to the molecules of the solutes in question, and that it may also have this function in vivo with regard to molecules of similar size.  相似文献

Summary Karyomeres or chromosome vesicles occur regularly at all cell divisions in cleavage embryos ofOphryotrocha labronica up to the 16-cell stage. They are formed as separate units, containing one or several nucleolus-like bodies (NLB) as well as intranuclear annulate lamellae (IAL), but coalesce later into a compound nucleus, in connection with copious blebbing and simultaneous appearance of cytoplasmic annulate lamellae (CAL). Labelling of the early embryos with3H-thymidine revealed marked localization of the synthesized DNA to the karyomere envelope region, whereas3H-uridine incorporation, indicating RNA synthesis, was sparse and notably absent in the NLB. On the other hand the latter structure like the envelopes preferentially incorporated3H-myoinositol, and displayed considerable labelling with3H-leucine. Summary The fat body of the adult housefly is composed of two types of cells, the lipid-and glycogen-rich fat body cells and the oenocytes. A comparison of the fine structure of the abdominal fat body in 4-day old and 31–35 day old male houseflies indicated an increase in lipid and a decrease in glycogen content in the fat body cells of old flies. Oenocytes of old flies exhibit deteriorative alterations with an accumulation of secondary lysosomes. The disappearance of larger individuals and the decrease in individual body condition suffered by Atlantic cod Gadus morhua in the eastern Baltic during the past two decades can be expected to affect the stock reproductive output. To investigate this, female G. morhua were collected during the spawning and pre‐spawning period in 2015?2016. The current individual potential fecundity (FP) of eastern Baltic G. morhua was estimated and analysed in relation to total length (LT) and indices of nutritional status such as body condition (K) and hepato‐somatic index (IH) using generalized linear models. In addition, the current prevalence of atresia and its potential relation to K were investigated. Moreover, a calibration curve to estimate FP from oocyte diameter, based on the autodiametric oocyte counting method, was established for the first time for eastern Baltic G. morhua and can be used for future fecundity studies on this stock. The results showed that FP was mainly positively related to fish length, but K and IH also contributed significantly to the variation in FP. The model predicted that fish with K = 1·2 have a FP 51% higher than fish of the same LT with K = 0·8. The prevalence of fecundity regulation by atresia was 5·8%, but it was found only in fish in the pre‐spawning maturity stage and with low K. Temporal changes in biological features such as the length composition and individual body condition of eastern Baltic G. morhua, should be accounted for when estimating stock reproductive potential.  相似文献

Atlantic salmon Salmo salar smolts were produced with similar energetic states as wild S. salar and the effect of low energetic state on smolt migration was tested. The total energetic state of the fish (body lipids and proteins) in the spring was correlated with Fulton's condition factor (K). Fish at a low energetic state swam slower but migrated further than fish at a higher energetic state when tested in two experimental streams. During a period of starvation throughout the winter and spring, fish conserved their body‐lipid reserves at 1·5% by using more protein as an energy source and the metabolic shift occurred between 3·5 and 1·5% body lipids. An energetic state of approximately 3·5 kJ g?1 (K ≈ 0·65) appeared to be the critical limit for survival.  相似文献

In many disease-related and functional amyloids, the amyloid-forming regions of proteins are flanked by globular domains. When located in close vicinity of the amyloid regions along the chain, the globular domains can prevent the formation of amyloids because of the steric repulsion. Experimental tests of this effect are few in number and non-systematic, and their interpretation is hampered by polymorphism of amyloid structures. In this situation, modeling approaches that use such a clear-cut criterion as the steric tension can give us highly trustworthy results. In this work, we evaluated this steric effect by using molecular modeling and dynamics. As an example, we tested hybrid proteins containing an amyloid-forming fragment of Aβ peptide (17–42) linked to one or two globular domains of GFP. Searching for the shortest possible linker, we constructed models with pseudo-helical arrangements of the densely packed GFPs around the Aβ amyloid core. The molecular modeling showed that linkers of 7 and more residues allow fibrillogenesis of the Aβ-peptide flanked by GFP on one side and 18 and more residues when Aβ-peptide is flanked by GFPs on both sides. Furthermore, we were able to establish a more general relationship between the size of the globular domains and the length of the linkers by using analytical expressions and rigid body simulations. Our results will find use in planning and interpretation of experiments, improvement of the prediction of amyloidogenic regions in proteins, and design of new functional amyloids carrying globular domains.  相似文献

In Greenland, free‐living red coralline algae contribute to and dominate marine habitats along the coastline. Lithothamnion glaciale dominates coralline algae beds in many regions of the Arctic, but never in Godthåbsfjord, Greenland, where Clathromorphum sp. is dominant. To investigate environmental impacts on coralline algae distribution, calcification and primary productivity were measured in situ during summers of 2015 and 2016, and annual patterns of productivity in L. glaciale were monitored in laboratory‐based mesocosm experiments where temperature and salinity were manipulated to mimic high glacial melt. The results of field and cold‐room measurements indicate that both L. glaciale and Clathromorphum sp. had low calcification and photosynthetic rates during the Greenland summer (2015 and 2016), with maximum of 1.225 ± 0.17 or 0.002 ± 0.023 μmol CaCO 3 · g?1 · h?1 and ?0.007 ±0.003 or ?0.004 ± 0.001 mg O2 · L?1 · h?1 in each species respectively. Mesocosm experiments indicate L. glaciale is a seasonal responder; photosynthetic and calcification rates increase with annual light cycles. Furthermore, metabolic processes in L. glaciale were negatively influenced by low salinity; positive growth rates only occurred in marine treatments where individuals accumulated an average of 1.85 ± 1.73 mg · d?1 of biomass through summer. These results indicate high freshwater input to the Godthåbsfjord region may drive the low abundance of L glaciale , and could decrease species distribution as climate change increases freshwater input to the Arctic marine system via enhanced ice sheet runoff and glacier calving.  相似文献
